Auxin-induced growth, epidermal cell length, cellular osmotic potential, and cell wall composition of coleoptile segments excised from one normal and two dwarf rice strains were studied 2, 3, 4, and 5 days after soaking. The auxin-induced growth was higher at the early stages of coleoptile growth and decreased with age, being always higher in normal than in the two dwarf strains. A good correlation between auxin-induced growth and auxin-induced decrease in the minimum stress-relaxation time has been found, suggesting that the different growth capacity in response to auxin among the three different strains is due to differences in the structure of their cell walls. In fact, cell wall analysis revealed that (1) the relative α-cellulose content of the cell walls was higher in the two dwarf strains than in the normal one, and (2) the auxin-induced decrease in noncellulosic glucose was high, compared with dwarf strains, in the normal strain, which showed the higher auxin-induced growth, showing a highly significant correlation between the decrease in noncellulosic glucose and the growth in response to auxin. Thus, the different growth between normal and dwarf strains might be attributed to their different capacity to degrade β-glucan of their cell walls.  相似文献

The course of haemostasis defects was investigated in dimethylnitrosamine (DMNA) acute liver necrosis. Before 18 hr there was no evidence of disseminated intravascular coagulation (DIC) nor of abnormal fibrinolysis. At 12 hr the level of the vitamin-K-dependent factors (factors II, VII, IX and X) was reduced to 25-63% of control. Factors V and VIII:C levels decreased to about 10 and 20% by 12 hr. Factor V was the only molecule which decreased significantly by 6 hr. The rapid decrease of these proteins might be related to an early parenchymal functional impairment attested by early structural lesions observed in the endoplasmic reticulum and nucleus. The isolated decrease of factor V in the absence of any significant change in serum transaminase (SGOT and SGPT) levels is proposed, at least in the rat, as an early criterion of hepatic failure.  相似文献

The Agave angustifolia complex, distributed from Mexico to Costa Rica, comprises four species and five varieties, including three species used for mescal production. The complex is represented in the Mexican state of Oaxaca by two wild taxa, A. angustifolia var. angustifolia and A. angustifolia var. rubescens, the cultivated form A. angustifolia “Espadín” and the partially cultivated species A. rodacantha. The aims of this study were to investigate the morphological and genetic variation of the A. angustifolia complex in the state of Oaxaca and to identify traits useful for taxonomic delimitation. Four wild and three cultivated populations of A. angustifolia from Oaxaca, one population of A. tequilana from Guanajuato and one population of A. angustifolia from Sonora were sampled for morphological, genetic and cytometric analyses. We showed that cultivated populations of A. angustifolia “Espadin,” A. rhodacantha and A. tequilana could be clearly differentiated from wild populations. Furthermore, the domesticated populations of A. angustifolia, known locally as “Espadin,” had a higher ploidy level and lower genetic variation than their related wild populations. The population of A. angustifolia from Sonora could be recognized as a different entity. Populations of A. rhodacantha need to be studied throughout their entire distribution area to further evaluate their taxonomic delimitation.  相似文献

 Three cycles of S1 recurrent selection for yield were carried out in two synthetic maize populations, EPS6 from humid Spain and EPS7 from arid Spain. One-hundred S1 lines were evaluated from each cycle of selection and the ten highest-yielding S1 lines were recombined to produce the next cycle. Changes in variability and genetic distances in two synthetic maize populations, following three cycles of recurrent selection, recombining ten S1 lines in each cycle, were determined. Isozyme analysis was performed on 125 seedlings per cycle of selection (four cycles in each of two populations). Regressions of each allozyme frequency on cycles of selection were performed, genetic distances between populations were determined, and simple correlations between genetic distances and heterosis were calculated. The average heterozygosity per locus was also calculated for each population. Regression analysis did not reveal any common trend between EPS6 and EPS7 for changes in allele frequencies presumably due to selection. The number of polymorphic loci, the mean alleles per locus, and the mean heterozygosity did not show any reduction in variability. Finally, selection did not affect genetic distances among cycles of selection. The agronomic evaluation of the selection program, after three cycles of selection, revealed that the genetic variance was not significantly reduced for most traits, and that the heterosis among cycles of selection of both populations had not changed significantly. The conclusions based on isozyme data supported the deductions made from agronomic data. Three cycles of selection neither caused relevant changes on variability nor on genetic distance among cycles of selection of both maize synthetic populations. Summary Portions (1.0–1.5 cm long) of terminal shoots from selected mature treesOlea europaea L. cv. Arbequina, micrografted in one phase ontoin vitro juvenile shoots, resulted in the restoration of shoot-bud proliferation and rooting competence. Although higherin vitro survival rates were obtained after a second repeated micrografting, the reinvigoration ratio of the regenerated shoots, indicated by proliferation and rooting ability, was not improved after two phases of micrografting. Thus, one-phase micrograft allows for a successful micropropagation system for olive trees. The cuttings obtained from successive pruning of plants produced through micrografting and growth in soil showed complete restoration of rooting competence, with rooting percentages similar to those of juvenile microshoots.  相似文献

Herpes simplex virus type 1 (HSV-1) has the ability to enter many different hosts and cell types by several strategies. This highly prevalent alphaherpesvirus can enter target cells using different receptors and different pathways: fusion at a neutral pH, low-pH-dependent and low-pH-independent endocytosis. Several cell receptors for viral entry have been described, but several observations suggest that more receptors for HSV-1 might exist. In this work, we propose a novel role for the proteolipid protein (PLP) in HSV-1 entry into the human oligodendrocytic cell line HOG. Cells transfected with PLP-EGFP showed an increase in susceptibility to HSV-1. Furthermore, the infection of HOG and HOG-PLP transfected cells with the R120vGF virus–unable to replicate in ICP4-defficient cells- showed an increase in viral signal in HOG-PLP, suggesting a PLP involvement in viral entry. In addition, a mouse monoclonal antibody against PLP drastically inhibited HSV-1 entry into HOG cells. PLP and virions colocalized in confocal immunofluorescence images, and in electron microscopy images, which suggest that PLP acts at the site of entry into HOG cells. Taken together these results suggest that PLP may be involved in HSV-1 entry in human oligodendrocytic cells.  相似文献

Synopsis The nanoflagellateTetraselmis suecica was tested both as the sole food source and as a diet complement toArtemia nauplii for grunion,Leuresthes tenuis, larvae. A total of 4800 grunion larvae, obtained through artificial insemination and incubation, were cultivated under laboratory conditions. Growth and survival rates were registered for 14 days in two experimental series. In the first series the nanoflagellateT. suecica was offered as the sole food source at five different concentrations. Survival and growth increased in agreement with the increase inT. suecica concentration. In the second series,Artemia nauplii were offered at six concentration levels. This series was divided into two groups: the nanoflagellateT. suecica was added to one group at a concentration of 5000 cells ml–1; the other group was maintained without nanoflagellates. In this series, survival and growth were directly related to nauplii concentration, but significant effects of the nanoflagellates were evident only in relation to the survival; the greatest difference (58% without nanoflagellates vs. 69% with nanoflagellates) was observed at anArtemia concentration of 1000 nauplii 1–1. The mechanism responsible for increased survival ofL. tenuis larvae in presence of phytoplankton is unclear.  相似文献

Addition of NH4Cl at low concentrations to Azotobacter chroococcum cells caused an immediate cessation of nitrate uptake activity, which was restored when the added NH 4 + was exhausted from the medium or by adding an NH 4 + assimilation inhibitor, l-methionine-dl-sulfoximine (MSX) or l-methionine sulfone (MSF). In the presence of such inhibitors the newly-reduced nitrate was released into the medium as NH 4 + . When the artificial electron donor system ascorbate/N-methylphenazinium methylsulfate (PMS), which is a respiratory substrate that was known to support nitrate uptake by A. chroococcum while inhibiting glutamine synthetase activity, was the energy source, externally added NH 4 + had no effect on nitrate uptake. It is concluded that, in A. chroococcum cells, NH 4 + must be assimilated to exert its short-term inhibitory effect on nitrate uptake. A fluorescence technique has been developed for observing starch granules in plant tissues. Sections are stained with a mixture of dyes which we have named F.A.S.G.A. from the initials of the Spanish names of its components (fucsina, alcian blue, safranina, glicerina, agua), and viewed by epifluorescence microscopy. The starch granules fluoresce greenish yellow, allowing the degradative state to be observed. Cell structures which do not fluoresce are also differentiated. The stain permits identification of other structures when examined by visible light microscopy and is relatively resistant to fading over time.  相似文献
