The high sensitivity that can be attained using an enzymatic system and mediated by catechol has been verified by on-line interfacing of a rotating biosensor and continuous-flow/stopped-flow/continuous-flow processing. Horseradish peroxidase, HRP, [EC 1.11.1.7], immobilized on a rotating disk, in the presence of hydrogen peroxide catalyzed the oxidation of catechol, whose back electrochemical reduction was detected on glassy carbon electrode surface at -200mV. Thus, when ciprofloxacin (CF) or norfloxacin (NF) was added to the solution, these piperazine-containing compounds participate in Michael addition reactions with catechol to form the corresponding aminoquinone derivative, decreasing the peak current obtained in proportion with the increase of its concentration. CF was used as the model piperazine-containing compound for the study. The influence of indicator composition on the nature of the analytical response has been assessed through examining the electrochemical properties of three derivatives. Interference by electroactive species (ascorbate, urate, and tyrosine) and other physiological constituents (cysteine, glutathione) has also been assessed.  相似文献

Understanding peroxidase function in plants is difficult because of the lack of substrate specificity, the high number of genes and their diversity in structure. In the present study, the relative expression of 22 genes coding putative peroxidases (E.C 1.11.1.x) in Arabidopsis was studied. The relative expression of AtPrx37 showed a correlation with the cessation of growth in rosette leaves as well as with the growth capacity along the flower stem. Using AtPrx37::GUS construction, its expression was associated with the vascular bundles. Furthermore, the overexpression of AtPrx37 under the control of CaMV 35S promoter rendered a dwarf phenotype with smaller plants and delayed development. The plants overexpressing AtPrx37 also showed an increase in the amount of esterified phenolic material associated with their walls. A role in the growth cessation and phenolic cross-linking during lignin deposition is postulated.  相似文献

The genus Polianthes belongs to Asparagaceae, subfamily Agavoideae (APG III, 2009) and is endemic to Mexico, spanning the states of Chihuahua and Tamaulipas in the north, and extending to the state of Oaxaca in the south. Its species are perennial, herbaceous plants with corms and bulbs. Their leaves are green or glaucous, linear or lanceolate. Inflorescences are in racemes or spikes with narrow and widely infundibuliform flowers, varying in color from white to red. The fruit is a capsule with black seeds. The number of species of Polianthes varies depending on the criteria to classify them. For example, in the first revision for the genus, Rose (1903) recognized 12 species. According to Espejo and López (1992), the genus has 15 species, whereas GarcíaMendoza and Galván (1995) reported 12 species, and Solano and Feria (2007) recognized 14 species and three varieties. Background

Fat content and fatty acid composition in swine are becoming increasingly studied because of their effect on sensory and nutritional quality of meat. A QTL (quantitative trait locus) for fatty acid composition in backfat was previously detected on porcine chromosome 8 (SSC8) in an Iberian x Landrace F2 intercross. More recently, a genome-wide association study detected the same genomic region for muscle fatty acid composition in an Iberian x Landrace backcross population. ELOVL6, a strong positional candidate gene for this QTL, contains a polymorphism in its promoter region (ELOVL6:c.-533C < T), which is associated with percentage of palmitic and palmitoleic acids in muscle and adipose tissues. Here, a combination of single-marker association and the haplotype-based approach was used to analyze backfat fatty acid composition in 470 animals of an Iberian x Landrace F2 intercross genotyped with 144 SNPs (single nucleotide polymorphisms) distributed along SSC8.

Results

Two trait-associated SNP regions were identified at 93 Mb and 119 Mb on SSC8. The strongest statistical signals of both regions were observed for palmitoleic acid (C16:1(n-7)) content and C18:0/C16:0 and C18:1(n-7)/C16:1(n-7) elongation ratios. MAML3 and SETD7 are positional candidate genes in the 93 Mb region and two novel microsatellites in MAML3 and nine SNPs in SETD7 were identified. No significant association for the MAML3 microsatellite genotypes was detected. The SETD7:c.700G > T SNP, although statistically significant, was not the strongest signal in this region. In addition, the expression of MAML3 and SETD7 in liver and adipose tissue varied among animals, but no association was detected with the polymorphisms in these genes. In the 119 Mb region, the ELOVL6:c.-533C > T polymorphism showed a strong association with percentage of palmitic and palmitoleic fatty acids and elongation ratios in backfat.

Conclusions

Our results suggest that the polymorphisms studied in MAML3 and SETD7 are not the causal mutations for the QTL in the 93 Mb region. However, the results for ELOVL6 support the hypothesis that the ELOVL6:c.-533C > T polymorphism has a pleiotropic effect on backfat and intramuscular fatty acid composition and that it has a role in the determination of the QTL in the 119 Mb region.  相似文献

At 4 days after the implantation of two subcutaneous 75 mg morphine pellets in the back skin, rats were morphine-dependent. In the three layers studied in the occipital cortex we found that the values of the 2-adrenergic agonist [3H]bromoxidine binding increased with respect to animals implanted with placebo pellets. Typical behavioral and physiological symptoms of the abstinence syndrome appeared 30 minutes after administration of naloxone, [3H]bromoxidine binding values being similar to those obtained in animals implanted with placebo pellets. The pattern of response of the [3H]bromoxidine binding was similar in the hippocampus and the superficial gray layer of the superior colliculus of the mesencephalon, but the differences were not statistically significant in these areas. This paper concludes that exist brain regional differences in the 2-adrenoreceptors response under morphine-treatment and possibly under naloxone-induced morphine abstinence syndrome.  相似文献

BACKGROUND AND AIMS: Atriplex (Halimione) portulacoides is a halophytic, C(3) shrub. It is virtually confined to coastal salt marshes, where it often dominates the vegetation. The aim of this study was to investigate its growth responses to salinity and the extent to which these could be explained by photosynthetic physiology. METHODS: The responses of young plants to salinity in the range 0-700 mol m(-3) NaCl were investigated in a glasshouse experiment. The performance of plants was examined using classical growth analysis, measurements of gas exchange (infrared gas analysis), determination of chlorophyll fluorescence characteristics (modulated fluorimeter) and photosynthetic pigment concentrations; total ash, sodium, potassium and nitrogen concentrations, and relative water content were also determined. KEY RESULTS: Plants accumulated Na(+) approximately in proportion to external salinity. Salt stimulated growth up to an external concentration of 200 mol m(-3) NaCl and some growth was maintained at higher salinities. The main determinant of growth response to salinity was unit leaf rate. This was itself reflected in rates of CO(2) assimilation, which were not affected by 200 mol m(-3) but were reduced at higher salinities. Reductions in net photosynthetic rate could be accounted for largely by lower stomatal conductance and intercellular CO(2) concentration. Apart from possible effects of osmotic shock at the beginning of the experiment, salinity did not have any adverse effect on photosystem II (PSII). Neither the quantum efficiency of PSII (Phi(PSII)) nor the chlorophyll fluorescence ratio (F(v)/F(m)) were reduced by salinity, and lower mid-day values recovered by dawn. Mid-day F(v)/F(m) was in fact depressed more at low external sodium concentration, by the end of the experiment. CONCLUSIONS: The growth responses of the hygro-halophyte A. portulacoides to salinity appear largely to depend on changes in its rate of photosynthetic gas exchange. Photosynthesis appears to be limited mainly through stomatal conductance and hence intercellular CO(2) concentration, rather than by effects on PSII; moderate salinity might stimulate carboxylation capacity. This is in contrast to more extreme halophytes, for which an ability to maintain leaf area can partially offset declining rates of carbon assimilation at high salinity.  相似文献

BackgroundThe pork tapeworm (Taenia solium) is a parasitic helminth that imposes a major health and economic burden on poor rural populations around the world. As recognized by the World Health Organization, a key barrier for achieving control of T. solium is the lack of an accurate and validated simulation model with which to study transmission and evaluate available control and elimination strategies. CystiAgent is a spatially-explicit agent based model for T. solium that is unique among T. solium models in its ability to represent key spatial and environmental features of transmission and simulate spatially targeted interventions, such as ring strategy.Methods/Principal findingsWe validated CystiAgent against results from the Ring Strategy Trial (RST)–a large cluster-randomized trial conducted in northern Peru that evaluated six unique interventions for T. solium control in 23 villages. For the validation, each intervention strategy was replicated in CystiAgent, and the simulated prevalences of human taeniasis, porcine cysticercosis, and porcine seroincidence were compared against prevalence estimates from the trial. Results showed that CystiAgent produced declines in transmission in response to each of the six intervention strategies, but overestimated the effect of interventions in the majority of villages; simulated prevalences for human taenasis and porcine cysticercosis at the end of the trial were a median of 0.53 and 5.0 percentages points less than prevalence observed at the end of the trial, respectively.Conclusions/SignificanceThe validation of CystiAgent represented an important step towards developing an accurate and reliable T. solium transmission model that can be deployed to fill critical gaps in our understanding of T. solium transmission and control. To improve model accuracy, future versions would benefit from improved data on pig immunity and resistance, field effectiveness of anti-helminthic treatment, and factors driving spatial clustering of T. solium infections including dispersion and contact with T. solium eggs in the environment.  相似文献

1. Responses of zooplankton to nutrient enrichment and fish predation were studied in 1998 and 1999 by carrying out parallel mesocosm experiments in six lakes across Europe. 2. Zooplankton community structure, biomass and responses to nutrient and fish manipulation showed geographical and year‐to‐year differences. Fish had a greater influence than nutrients in regulating zooplankton biomass and especially the relative abundances of different functional groups of zooplankton. When fish reduced the biomass of large crustaceans, there was a complementary increase in the biomasses of smaller crustacean species and rotifers. 3. High abundance of submerged macrophytes provided refuge for zooplankton against fish predation but this refuge effect differed notably in magnitude among sites. 4. Large crustacean grazers (Daphnia, Diaphanosoma, Sida and Simocephalus) were crucial in controlling algal biomass, while smaller crustacean grazers and rotifers were of minor importance. Large grazers were able to control phytoplankton biomass even under hypereutrophic conditions (up to 1600 μg TP L?1) when grazer biomass was high (>80–90 μg dry mass L?1) or accounted for >30% of the grazer community. 5. The littoral zooplankton community was less resistant to change following nutrient enrichment in southern Spain, at high temperatures (close to 30 °C), than at lower temperatures (17–23 °C) characterising the other sites. This lower resistance was because of a greater importance of nutrients than zooplankton in controlling algal biomass. 6. Apart from the reduced role of large crustacean grazers at the lowest latitude, no consistent geographical patterns were observed in the responses of zooplankton communities to nutrient and fish manipulation.  相似文献

Aim Climate change causes shifts in species distributions, or ‘migrations’. Despite the centrality of species distributions to biodiversity conservation, the demonstrated large migration of tropical plant species in response to climate change in the past, and the expected sensitivity of species distributions to modern climate change, no study has tested for modern species migrations in tropical plants. Here we conduct a first test of the hypothesis that increasing temperatures are causing tropical trees to migrate to cooler areas. Location Tropical Andes biodiversity hotspot, south‐eastern Peru, South America. Methods We use data from repeated (2003/04–2007/08) censuses of 14 1‐ha forest inventory plots spanning an elevational gradient from 950 to 3400 m in Manu National Park in south‐eastern Peru, to characterize changes in the elevational distributions of 38 Andean tree genera. We also analyse changes in the genus‐level composition of the inventory plots through time. Results We show that most tropical Andean tree genera shifted their mean distributions upslope over the study period and that the mean rate of migration is approximately 2.5–3.5 vertical metres upslope per year. Consistent with upward migrations we also find increasing abundances of tree genera previously distributed at lower elevations in the majority of study plots. Main conclusions These findings are in accord with the a priori hypothesis of upward shifts in species ranges due to elevated temperatures, and are potentially the first documented evidence of present‐day climate‐driven migrations in a tropical plant community. The observed mean rate of change is less than predicted from the temperature increases for the region, possibly due to the influence of changes in moisture or non‐climatic factors such as substrate, species interactions, lags in tree community response and/or dispersal limitations. Whatever the cause(s), continued slower‐than‐expected migration of tropical Andean trees would indicate a limited ability to respond to increased temperatures, which may lead to increased extinction risks with further climate change.  相似文献

A Markov process with several absorbent states is applied for analyzing a breast cancer dataset. The study examines the evolution of patients until death, and shows that two well‐differentiated ways can be considered in the evolution of patients towards the death state: those who relapse and those who not. The risk groups we have considered are determined by the application of treatments radiotherapy and chemotherapy, which are introduced as covariates. Four states are distinguished: no relapse, relapse, death after metastasis, and death without metastasis, the last two absorbent. We apply a methodology that uses algorithmic procedures, avoiding differential equations. The transition probability functions and the likelihood function in the model are calculated. For the dataset, the survival functions and the mean times in states for the different group of risks are determined. We show that the metastasis is the main cause of death in this cohort, but the number of deaths by relapse is not negligible.  相似文献
