The environmental physiology of three speciesof Collembola: Cryptopygus cisantarcticus, Isotoma klovstadi (Isotomidae) and Friesea grisea (Neanuridae) was investigated from November 2002 to February 2003 at Cape Hallett, North Victoria Land, Antarctica. All three species were freeze avoiding, and while supercooling points were variable on seasonal and daily scales in I. klovstadi and C. cisantarcticus, they remained largely static in F. grisea. LT50 (temperature where 50% of animals are killed by cold) was -13.6, -19.1 and -19.8 degrees C for C. cisantarcticus, I. klovstadi and F. grisea, respectively. Upper lethal temperature was 34, 34 and 38 degrees C for C. cisantarcticus, I. klovstadi and F. grisea. Critical thermal minimum onset (the temperature where individuals entered chill coma) was ca. -7, -12 and -8 degrees C for C. cisantarcticus, I. klovstadi and F. grisea, and 25% of I. klovstadi individuals froze without entering chill coma. Critical thermal maximum (the onset of spasms at high temperature) was 30, 33 and 34 degrees C for C. cisantarcticus, I. klovstadi and F. grisea. Haemolymph osmolality was approximately 720 mOsm for C. cisantarcticus and 680 mOsm for I. klovstadi, and both species showed a moderate degree of thermal hysteresis, which persisted through the season. Desiccation resistance was measured as survival above silica gel, and the species survived in the rank order of C. cisantarcticus< I. klovstadi = F. grisea. Desiccation resulted in an increase in haemolymph osmolality in I. klovstadi, and water was quickly regained by desiccation-stressed individuals that had access to liquid water, but not by individuals placed in high humidity, indicating that this species is unable to absorb atmospheric water vapour. SDS-PAGE did not suggest any strong patterns in protein synthesis either seasonally or in response to temperature or desiccation stress. Microclimate temperatures were measured at sites representative of collection sites for the three species. Microclimate temperatures were highly variable on a diurnal and weekly scale (the latter relating to weather patterns), but showed little overall variation across the summer season. Potentially lethal high and low temperatures were recorded at several sites, and it is suggested that these temperature extremes account for the observed restriction of the less-tolerant C. cisantarcticus at Cape Hallett. Together, these data significantly increase the current knowledge of the environmental physiology of Antarctic Collembola.  相似文献

Water loss in insects: an environmental change perspective   总被引:1,自引:0,他引:1  
In the context of global environmental change much of the focus has been on changing temperatures. However, patterns of rainfall and water availability have also been changing and are expected to continue doing so. In consequence, understanding the responses of insects to water availability is important, especially because it has a pronounced influence on insect activity, distribution patterns, and species richness. Here we therefore provide a critical review of key questions that either are being or need to be addressed in this field. First, an overview of insect behavioural responses to changing humidity conditions and the mechanisms underlying sensing of humidity variation is provided. The primary sensors in insects belong to the temperature receptor protein superfamily of cation channels. Temperature-activated transient receptor potential ion channels, or thermoTRPs, respond to a diverse range of stimuli and may be a primary integrator of sensory information, such as environmental temperature and moisture. Next we touch briefly on the components of water loss, drawing attention to a new, universal model of the water costs of gas exchange and its implications for responses to a warming, and in places drying, world. We also provide an overview of new understanding of the role of the sub-elytral chamber for water conservation, and developments in understanding of the role of cuticular hydrocarbons in preventing water loss. Because of an increasing focus on the molecular basis of responses to dehydration stress we touch briefly on this area, drawing attention to the role of sugars, heat shock proteins, aquaporins, and LEA proteins. Next we consider phenotypic plasticity or acclimation responses in insect water balance after initial exposures to altered humidity, temperature or nutrition. Although beneficial acclimation has been demonstrated in several instances, this is not always the case. Laboratory studies show that responses to selection for enhanced ability to survive water stress do evolve and that genetic variation for traits underlying such responses does exist in many species. However, in others, especially tropical, typically narrowly distributed species, this appears not to be the case. Using the above information we then demonstrate that habitat alteration, climate change, biological invasions, pollution and overexploitation are likely to be having considerable effects on insect populations mediated through physiological responses (or the lack thereof) to water stress, and that these effects may often be non-intuitive.  相似文献

The structure of a C11 peptidase PmC11 from the gut bacterium, Parabacteroides merdae, has recently been determined, enabling the identification and characterization of a C11 orthologue, PNT1, in the parasitic protozoon Trypanosoma brucei. A phylogenetic analysis identified PmC11 orthologues in bacteria, archaea, Chromerids, Coccidia, and Kinetoplastida, the latter being the most divergent. A primary sequence alignment of PNT1 with clostripain and PmC11 revealed the position of the characteristic His-Cys catalytic dyad (His99 and Cys136), and an Asp (Asp134) in the potential S1 binding site. Immunofluorescence and cryoelectron microscopy revealed that PNT1 localizes to the kinetoplast, an organelle containing the mitochondrial genome of the parasite (kDNA), with an accumulation of the protein at or near the antipodal sites. Depletion of PNT1 by RNAi in the T. brucei bloodstream form was lethal both in in vitro culture and in vivo in mice and the induced population accumulated cells lacking a kinetoplast. In contrast, overexpression of PNT1 led to cells having mislocated kinetoplasts. RNAi depletion of PNT1 in a kDNA independent cell line resulted in kinetoplast loss but was viable, indicating that PNT1 is required exclusively for kinetoplast maintenance. Expression of a recoded wild-type PNT1 allele, but not of an active site mutant restored parasite viability after induction in vitro and in vivo confirming that the peptidase activity of PNT1 is essential for parasite survival. These data provide evidence that PNT1 is a cysteine peptidase that is required exclusively for maintenance of the trypanosome kinetoplast.  相似文献

The environmental predictability (EP) hypothesis proposes that rapid cold hardening (RCH) might be common in temperate species incapable of surviving freezing events and which also dwell in unpredictable environments. The kelp fly Paractora dreuxi serves as a useful model organism to test this prediction at an intra-specific level because larvae and adults show different responses to low temperature despite occupying a similar unpredictable thermal environment. Here, using acclimation temperatures, which simulated seasonal temperature variation, we find little evidence for RCH in the freeze-intolerant adults but a limited RCH response in freeze-tolerant larvae. In the relatively short-lived adults, survival of -11 degrees C generally did not improve after 2h pre-treatments at -4, -2, 0, 10, 20 or 25 degrees C either in summer- (10 degrees C) or winter (0 degrees C)-acclimated individuals. By contrast, survival of summer-acclimated larvae to -7.6 degrees C was significantly improved by approximately 37% and 30% with -2 and 0 degrees C pre-treatments, respectively. The finding that summer-acclimated larvae showed RCH whereas this was not the case in the winter-acclimated larvae partially supports the predictions of the EP hypothesis. However, the EP hypothesis also predicts that the adults should have demonstrated an RCH response, yet they did not do so. Rather, it seems likely that they avoid stressful environments by behavioural thermoregulation. Differences in responses among the adults and larvae are therefore to some extent predictable from differences in their feeding requirements and behaviour. These results show that further studies of RCH should take into account the way in which differences among life stages influence the interaction between phenotypic plasticity and environmental variability and predictability.  相似文献

Oxidation of pyruvate, -ketoglutarate, palmitoylcarnitine, succinate, and ferrocytochrome c by interscapular-brown-adipose-tissue (BAT) mitochondria of untrained and trained rats were measured at rest and alter running to exhaustion. At rest, BAT mitochondria from trained rats showed significantly lower activities (<50%) for the oxidation of all the substrates. In untrained rats the activities of the enzymes for the oxidation of all the substrates except pyruvate and succinate were lower at exhaustion compared to the resting state when expressed on a per-gram-fresh-Weight basis. In trained rats all of the enzyme activities increased as a result of exhaustive exercise. These differences between the two groups of rats in the post-exercise changes in oxidative capacities suggest that following an initial adaptation, resulting in a large decrease in mitochondrial oxidative activity, training protects the residual oxidative pathways against exercise-induced inactivation. These data show that unlike exposure to cold, or overfeeding, a physiological stimulus such as exercise reduces the oxidative capacity of BAT, and therefore may reduce the thermogenic activity of the tissue in endurance-trained rats as has been addressed in the scientific literature.  相似文献

Three regression models are proposed for predicting reproduction in a model dairy herd under South African conditions. Conception rate (CR%) was related to mean monthly temperature-humidity index (THI) by; CR% = 31.15THI - 0.25THI(2) - 890.2, and first service conception rate (FSCR%) to THI by; FSCR% = 173.45 - 1.79THI. Conception rate was related to numerical month of the year (M) by; CR% = 11.86M - 0.82M(2) + 26.36. The relation between mean monthly THI values and the conception rate of dairy cattle is significant. Further investigations to test the proposed regression models under various dairy herd conditions and to improve reproduction in South African dairy herds are needed.  相似文献

The effects of cold acclimation on the activity levels of cytochrome c oxidase, glutathione peroxidase and glutathione reductase in various tissues of the rat (Rattus norvegicus) were investigated. One group was individually housed at 4 +/- 1 degrees C and the other at 24 +/- 1 degrees C for 6 months. Chronic cold acclimation resulted in significantly (P < 0.05) increased cytochrome c oxidase activity levels in liver, kidney, heart, interscapular brown adipose tissue and gastrocnemius muscle. The activity of glutathione peroxidase was significantly (P < 0.05) elevated in liver, interscapular brown adipose tissue, lung and muscle, whereas glutathione reductase was only significantly (P < 0.05) elevated in interscapular brown adipose tissue as a result of chronic cold exposure. The results obtained are possibly indicative of a positive compensatory response against the increased production of oxygen derived radicals as a result of chronic cold exposure.  相似文献

While biochemical mechanisms are typically used by animals to reduce oxidative damage, insects are suspected to employ a higher organizational level, discontinuous gas exchange mechanism to do so. Using a combination of real-time, flow-through respirometry and live-cell fluorescence microscopy, we show that spiracular control associated with the discontinuous gas exchange cycle (DGC) in Samia cynthia pupae is related to reactive oxygen species (ROS). Hyperoxia fails to increase mean ROS production, although minima are elevated above normoxic levels. Furthermore, a negative relationship between mean and mean ROS production indicates that higher ROS production is generally associated with lower . Our results, therefore, suggest a possible signalling role for ROS in DGC, rather than supporting the idea that DGC acts to reduce oxidative damage by regulating ROS production.  相似文献
