Pouched mice (Saccostomus campestris) were born in captivity during January and March and subsequently maintained under long photoperiod (14 h light: 10 h dark) at 25°C. During their first winter (July) and the following summer (January) the pouched mice were exposed to natural photoperiod in an unheated laboratory for 3 weeks prior to measurement. The pouched mice continued to grow during the study, and were significantly heavier after summer exposure than after winter exposure 6 months earlier. Although this increase in body mass would result in a decline in their surface area to volume ratio there was no significant decline in minimal thermal conductance (C m) and winter-exposed pouched mice had a relatively lowerC m than expected. Meanwhile the smaller, winter-exposed animals displayed a significantly higher capacity for non-shivering thermogenesis, together with higher levels of basal metabolism than summer individuals. These differences were not solely attributable to the contrasting body mass of each group and it is therefore clear thatS. campestris can increase thermoregulatory heat production, and modify heat loss following exposure to short photoperiod and cold during their first winter. Despite the significant increase in metabolism, the overall energy requirements of small, winter-exposed animals were significantly lower than those for heavier pouched mice following exposure to summer conditions. These results suggest that growing pouched mice can effectively adapt to lower temperature conditions during their first winter, yet accrue considerable overall savings in total energy requirements as a result of their smaller body mass.  相似文献

In an effort to further characterize the defective proliferative response of T lymphocytes to mitogens in multiple sclerosis (MS) patients, we examined the response to and production of interleukin 2 (IL 2) by both peripheral blood lymphocytes (PBL) and cerebrospinal fluid mononuclear cells. We also examined the proportion of cells bearing receptors for IL 2 and transferrin. Chronic progressive MS patients have an abnormally low response to exogenous IL 2 as compared to controls. Whereas acute relapse patients' PBL demonstrated a normal IL 2 response during an exacerbation, they showed reduced responsiveness during remission. These abnormalities could not be explained by different dose or kinetic response optima to PHA or IL 2, nor could they be explained by depressed numbers of IL 2 or transferrin receptor-bearing lymphocytes. Production of IL 2 by PBL was also abnormal in MS patients. Chronic progressive patients produced elevated levels of IL 2, whereas acute relapse patients undergoing an exacerbation produced diminished levels of IL 2. During remission, these levels returned to that of controls'. The effect of 1200 rad x-irradiation or nylon wool removal of adherent cells was a significantly greater augmentation of IL 2 production in MS patients than in other neurologic disease or normal controls. Cerebrospinal fluid lymphocytes from MS patients had normal proportions of IL 2 receptor-bearing cells, but were deficient in their IL 2 response and production as compared to autochthonous or control PBL. The inability of some MS patients' lymphocytes to clonally expand in response to IL 2 might contribute to the pathogenicity of the disease.  相似文献

Fine needle aspiration (FNA) in HIV+ patients: results from a series of 655 aspirates
There are many selected small series or case reports of FNAs in patients with HIV infection, but large series are rare and the epidemic's characteristics have evolved over time. The current study, from a large public hospital in the USA, included women as well as men, hetero- and homosexuals, in-patients and out-patients, and deep radiologically guided aspirates as well as superficial masses. Of 655 FNAs, reactive or benign changes were present in 37%, confirmed or suspected malignancy in 13%, speciwc infection with stainable organisms in 14%, and inflammation in 16%. Twenty percent of cases were inadequate for diagnosis. Most of the identiwable infections were associated with Mycobacterium tuberculosis , with fewer atypical mycobacteria, fungi and other bacteria. Clinically signiwcant diagnoses were correlated with deep aspirate location and lesion size > 2 cm, conwrming other studies which also identiwed tenderness and recent enlargement as important indicators. The liberal use of FNA in our HIV+ population has greatly reduced the necessity for surgical nodal resection, reassured clinicians in continuing observation of reactive lymphadenopathy, and allowed immediate therapy for speciwc infection, cyst or malignancy.  相似文献

The G protein family of transmembrane signaling molecules includes Gs and Gi, the stimulatory and inhibitory regulators of adenylate cyclase. These and other characterized G proteins are comprised of beta, gamma, and alpha chains, the latter being the most variable among the proteins and thus serving to distinguish them. Previous results (Begin-Heick, N. (1985) J. Biol. Chem. 260, 6187-6193) suggested that the autosomal recessive mouse mutation obese (ob), which results in an abnormal response of adipose tissue to lipolytic hormones, is due to a defect in the gene coding for the alpha chain of Gi. In order to test this hypothesis we used a cloned cDNA probe representing murine Gi alpha mRNA in conjunction with a panel of Chinese hamster-mouse somatic cell hybrids segregating mouse chromosomes to map the Gi alpha gene in the mouse. In addition, we used a cDNA probe representing the murine Gs alpha gene to a specific mouse chromosome. Our results indicate that the Gi alpha locus maps to mouse chromosome 9, while Gs alpha is localized to region 2E1-2H3 of mouse chromosome 2. Localization of the Gi alpha gene to chromosome 9 excludes this gene as a site of the ob mutation, since the ob locus maps to chromosome 6. Furthermore, our findings indicate that certain members of the murine G protein alpha gene family have dispersed to different chromosomes since diverging from a common ancestral gene.  相似文献

The divergence in height velocity of early and late maturing girls in early adolescence is exploited to predict menarcheal age from a year's growth in height. Simple prediction equations are generated based on data from the Berkely Guidance Study, and are subsequently tested against an independent sample from the Stuart Growth Study. The success of this prediction method compares favorably with others based on x-ray determinations of bone age; it is much more successful than a proposed method that utilizes estimations of body fat. The height increment method is recommended both by its simplicity and by the relative accessability of the information involved.  相似文献

The ability of a stretch of alternating dA-dT to adopt the left-handed Z form has been assessed by examining the behavior of the sequence d(CG)6(TA)4(CG)6 contained in the plasmid pBR322. The structural transition occurring within this sequence as a function of negative superhelicity was analyzed by several methods, including (1) the supercoiling-dependent unwinding of the insert as determined by two-dimensional gel electrophoresis, (2) the binding of anti-Z-DNA antibodies to the insert, (3) the sensitivity of the sequence to a single strand specific endonuclease, and (4) the sensitivity of the insert to digestion by a restriction endonuclease that cuts within the d(CG)6 segments when in the right-handed form. These studies have shown that in negatively supercoiled DNA the two d(CG)6 portions of the insert adopt the Z form, while the central d(TA)4 segment forms an underwound structure with a helical repeat that is best approximated as being intermediate between the B form and the Z form. A statistical mechanical treatment of the unwinding of the insert as a function of negative superhelicity provides an estimate of the minimum free energy required to convert an A-T bp from the B form to the Z form, as well as the free energy associated with the conversion of an A-T bp from the B form to the unwound form. These results strongly indicate that Z DNA is an unfavored structural alternative for stretches of d(AT)n in negatively supercoiled DNA.  相似文献
