Light quality is a significant environmental factor that influences photosynthetic pigments in cyanobacteria. In the present study, we illuminated the marine cyanobacteria Synechococcus sp. with white (350 ~ 700 nm), red (630 nm), green (530 nm), and blue (450 nm) light emitting diodes (LEDs) and measured pigment levels (chlorophyll, carotenoid, and phycobiliprotein) and expression of photosynthesis-related genes (pebA, psbB, and psaE). The amount of photosynthetic pigments (total pigments, chlorophyll, and phycobiliproteins) was higher in the green and blue LED groups than in the white and red LED groups after 8 days of culture. The cells were prepared in a 1.5 mL solution for the analysis of the total pigments, chlorophyll, and carotenoid, and in a 2 mL for analysis of phycobiliproteins. The mRNA expression levels of pebA and psbB significantly increased after 8 days of cultivation under green and blue light, while the mRNA expression levels of psaE decreased. These results indicate that green and blue light increase the accumulation of photosynthetic pigments. Adaptive evolution was employed to generate sodium (Na+)-tolerant mutants of Escherichia coli MG1655. Four mutants with elevated sodium tolerance, designated ALS1184, ALS1185, ALS1186, and ALS1187, were independently isolated after 73 days of serial transfer in medium containing progressively greater Na+ concentrations. The isolates also showed increased tolerance of K+, although this cation was not used for selective pressure. None of the adapted mutants showed increased tolerance to the nonionic osmolyte sucrose. Several physiological parameters of E. coli MG1655 and ALS1187, the isolate with the greatest Na+ tolerance, were calculated and compared using glucose-limited chemostats. Genome sequencing showed that the ALS1187 isolate contained mutations in five genes, emrR, hfq, kil, rpsG, and sspA, all of which could potentially affect the ability of E. coli to tolerate Na+. Two of these genes, hfq and sspA, are known to be involved in global regulatory processes that help cells endure a variety of cellular stresses. Pyruvate formate lyase knockouts were constructed in strains MG1655 and ALS1187 to determine whether increased Na+ tolerance afforded increased anaerobic generation of lactate. In fed-batch fermentations, E. coli ALS1187 pflB generated 76.2 g/liter lactate compared to MG1655 pflB, which generated only 56.3 g/liter lactate. 相似文献

Aqueous zinc ion batteries (AZIBs) are steadily gaining attention based on their attractive merits regarding cost and safety. However, there are many obstacles to overcome, especially in terms of finding suitable cathode materials and elucidating their reaction mechanisms. Here, a mixed‐valence vanadium oxide, V6O13, that functions as a stable cathode material in mildly acidic aqueous electrolytes is reported. Paired with a zinc metal anode, this material exhibits performance metrics of 360 mAh g?1 at 0.2 A g?1, 92% capacity retention after 2000 cycles, and 145 mAh g?1 at a current density of 24.0 A g?1. A combination of experiments and density functional theory calculations suggests that hydrated intercalation, where water molecules are cointercalated with Zn ions upon discharge, accounts for the aforementioned electrochemical performance. This intercalation mechanism facilitates Zn ion diffusion throughout the host lattice and electrode–electrolyte interface via electrostatic shielding and concurrent structural stabilization. Through a correlation of experimental data and theoretical calculations, the promise of utilizing hydrated intercalation as a means to achieve high‐performance AZIBs is demonstrated. 相似文献

We previously reported that dehydroevodiamine.HCl (DHED) has anticholinesterase and antiamnesic activities. To verify the effects of DHED on cognitive deficits further, we tested it on the scopolamine-induced amnesia model of the rat using the passive avoidance and eight-arm radial maze tests. A single (20 mg/kg p.o.) and repeated (10 mg/kg p.o.) administrations of DHED could significantly reverse the latency time shortened by scopolamine (1 mg/kg i.p.) to control level. The impaired spatial working memory induced by scopolamine (1 mg/kg i.p.) was also improved significantly by a single injection (6.25 mg/kg i.p.) and repeated administrations of DHED (10 mg/kg p.o.) in the eight-arm radial maze test. In addition, we examined the effects of DHED on the memory impairment and the histological changes of the brain after unilateral electrolytic lesion of the entorhinal cortex (EC) and middle cerebral artery occlusion in rats. The cognitive deficits caused by EC lesion and middle cerebral artery occlusion were improved significantly by repeated administrations of DHED (6.25 mg/kg i.p.) after EC lesion or ischemic insult once a day for 7 days in the passive avoidance test. Histological analysis showed that the neuronal loss in the DHED-treated group was notably reduced in the hippocampal area (CA1) of ischemic rats and in the dentate gyrus and hippocampal area (CA1 and CA3) of EC-lesioned rats compared with the nontreated group. The infarction area was decreased significantly by a single administration of DHED (6.25 mg/kg i.p.) 30 min before ischemic insult for 6 h.
