Summary Microscopic observations of isotonic suspensions of human red blood cells demonstrate that cell shape is unaltered when the transmembrane electrical potential, orE m , is set in the range –85 to +10 mV with valinomycin at varied external K+, or K o .E m was measured with the fluorescent potentiometric indicator, diS-C3(5), as calibrated by a pH method. Repeating Glaser's experiments in which echinocytosis was attributed to hyperpolarization, we found that at low ionic strength the pH-dependent effects of amphotericin B appear to be unrelated toE m . The effects of increased intracellular Ca2+, or Ca o , on echinocytosis and onE m are separable. With Ca ionophore A23187 half-maximal echinocytosis occurs at greater Ca o than that which induces the half-maximal hyperpolarization associated with Ca-induced K+ conductance (Gardos effect). Thus, cells hyperpolarized by increased Ca o remain discoidal when Ca is below the threshold for echinocytosis. With A23187 and higher Ca o , extensive echinocytosis occurs in cells which are either hyperpolarized or at their resting potential. The Ca-activation curve for echinocytosis is left-shifted by low K o , a new observation consistent with increased DIDS-sensitive uptake of45Ca by hyperpolarized cells. These results support the following conclusions: (1) the shape and membrane potential of human red blood cells are independent under the conditions studied; (2) in cells treated with A23187, the Gardos effect facilitates echinocytosis by increasing Ca.  相似文献

The N-terminal amino acid sequences of two chloroform/methanol soluble globulins from barley and one form wheat are reported. They are homologous with N-terminal sequences previously reported for α-amylase and trypsin inhibitors from cereals and 2 S storage proteins from castor bean and rape. Three albumins were also purified from Aegilops squarrosa and Triticum monococcum. These had N-terminal amino acid sequences most closely related to the α-amylase and trypsin inhibitors. The relationships of this superfamily of seed proteins are discussed.  相似文献

High resolution 2DGE (two-dimensional gel electrophoresis) was used to characterize neuronal and glial proteins of the rat optic nerve, to examine the phases of intraaxonal transport with which the neuronal proteins are associated, and to identify the ribosomal populations on which these proteins are synthesized. Neuronal proteins synthesized in the retinal ganglion cells were identified by injecting the eye with L-[35S]methionine, followed by 2DGE analysis of fast and slow axonally transported proteins in particulate and soluble fractions. Proteins synthesized by the glial cells were labeled by incubating isolated optic nerves in the presence of L-[35S]methionine and then analyzed by 2DGE. A number of differences were seen between filamentous proteins of neurons and glia. Most strikingly, proteins in the alpha- and beta-tubulin region of the 2D gels of glial proteins were distinctly different than was observed for axonal proteins. As expected, neurons but not glia expressed neurofilament proteins, which appeared among the slow axonally transported proteins in the particulate fraction; significant amounts of the glial filamentous protein, GFA, were also labeled under these conditions, which may have been due to transfer of amino acids from the axon to the glial compartment. The fast axonally transported proteins contained relatively large amounts of high-molecular-weight acidic proteins, two of which were shown to comigrate (on 2DGE) with proteins synthesized by rat CNS rough microsomes; this finding suggests that rough endoplasmic reticulum may be a major site of synthesis for fast transported proteins. In contrast, the free polysome population was shown to synthesize the principal components of slow axonal transport, including tubulin subunits, actin, and neurofilament proteins.  相似文献

To elucidate the mechanisms whereby genes and environment influence wing size, we investigated the effects of various rearing temperatures and larval crowding conditions on the wings of the mutant miniature and wild-type fruit flies. In adults we monitored wing size, cell number, wing thickness, cell density; in larval imaginal discs we looked for cell death. Cell density was inversely proportional to wing size. Of particular interest was the finding that smaller wings tend to be thicker. Electron microscope studies showed that the miniature wing layers are grossly abnormal. We hypothesize that these abnormalities are due to abnormal cell flattening of the wing epithelial cells, and we conclude that gene and environmental effects on cell flattening may be an important component in determining cell density and hence organ size.  相似文献

Summary Chromosome preparations from seven subjects with aberrations of sex chromosomes were utilized for in situ hybridization studies with the tritium-labeled Y-derived probe p50f. Two subjects had a pseudodicentric chromosome consisting of two copies of Yp and a portion of Y long arm; two were XX males [46,XX,t(Xp;Yp)], one was missing part of the Y short arm, and another had t(5p;Yq); in addition cells from an XYY male as well as a normal 46,XY male, and a 46,XX female, were hybridized with the same probe. The hybridization technique of Harper and Saunders (1981) was used. There was excess labeling of the Yp/paracentromeric regions in the cases with the normal Y, the XYY, the pseudodicentric Y, and the 5/Y translocation. No significant label was seen on metaphases from the normal 46,XX female or the female with the partially missing Y short arm. Excess label was present on the X short arm in the cases of the XX males; there were 8% and 9.5% of cells with label. The combined cytogenetic and hybridization data indicate that one X short arm in these XX males has undergone a translocation with Yp, and that genes for sex determination probably reside on the distal half of the Y short arm.  相似文献

Summary The human 70 kdalton heat-shock protein (HSP 70) is a member of a multigene family which is expressed in response to various physiological stresses including elevated temperatures. Using a cloned genomic HSP 70 DNA sequence we demonstrate by somatic cell hybrid and restriction fragment length polymorphism (RFLP) analyses that there are a minimum of three distinct HSP 70 loci in the human genome, one of which is located on chromosome 6.  相似文献

Summary The genes encoding GS and EPSP synthase have already been cloned. Clones containing portions of the genes encoding aspartate amino transferase and asparagine synthetase tentatively have been identified by this and other laboratories. It is certain that many other genes encoding other important enzymes involved in amino acid biosynthesis will also be identified in the near future. It is evident that new techniques for the separation of proteins will greatly aid in identifying low abundance genes more rapidly and efficiently. Micro techniques need to be further developed and the necessary equipment made available to laboratories. Hopefully the cost of the required equipment will decrease and commercial enterprises will offer more contract services for work requiring the more expensive equipment. Graduate training in advanced techniques of separating proteins at high resolutions should be encour-aged expanded so that our future scientists are well versed in protein biochemistry as well as in techniques of molecular biology.  相似文献

Summary The Goldcrest is an interesting species for studies of physiological and behavioural adaptions to cold, since it is the smallest species present in Europe. Although many small birds have to lower their metabolic rate and enter hypothermia in order to make their energy reserves last throughout the night, our measurements of oxygen consumption of five Goldcrests suggest that these birds may be able to balance their nocturnal energy budget during the winter at normothermic body temperatures, provided that the available food supply is adequate. These results are in agreement with those of another small bird, the Common Bushtit, which, like the Goldcrest, reduce nocturnal heat loss by contact roosting. Since the energy saved by hypothermia decreases with decreasing ambient temperature and also decreasing body weight, this strategy might not be such an advantage for the smallest birds living at very low ambient temperatures.
Zusammenfassung Am Wintergoldhähnchen als dem kleinsten europäischen Vogel interessieren besonders seine physiologischen und Verhaltens-Anpassungen an niedere Temperaturen. Viele Kleinvögel senken ihre Stoffwechselaktivität und verfallen in Hypothermie, um mit ihren Energie-Reserven während der langen Winternächte auszukommen. Unsere Untersuchungen über den O2-Verbrauch von 5 Wintergoldhähnchen legen jedoch nahe, daß sich das nächtliche Energie-Budget auch im Tiefwinter auf Normalniveau bewegt und daß sie bei ausreichendem Nahrungsangebot ihre normale Körpertemperatur auch nachts beibehalten. Ähnliche Befunde liegen von den vergleichbaren kleinen amerikanischen Buschmeisen vor, die wie Goldhähnchen den nächtlichen Wärmeverlust durch Schlafen auf engem Gefiederkontakt reduzieren. Es ist anzunehmen, daß die Hypothermie als Überwinterungsstrategie für unsere kleinsten Vögel garnicht so vorteilhaft wäre: Die durch Hypothermie eingesparte Energie geht mit abnehmender Umgebungstemperatur und mit geringerer Körpergröße wieder verloren.

Phospholipid packing has been suggested as a relevant variable in the control of membrane fusion events. To test this possibility in a model system, a comparison was made of the fusability of erythrocytes with a normal asymmetric transbilayer distribution of plasma membrane phospholipids (tightly packed exterior lipids) and erythrocytes with a symmetric transbilayer distribution of phospholipids (more loosely packed exterior lipids), using polyethylene glycol as fusogen. Not only were lipid-symmetric cells more readily fused, but fusions of mixtures of lipid-symmetric and lipid-asymmetric cells indicated that both fusing partners must have a symmetric distribution for fusion to be enhanced. Lipid-symmetric cells may fuse more readily because loose packing of the exterior lipids enhances hydrophobic interactions between cells. Alternatively, enhanced membrane fluidity may facilitate intramembranous particle clustering, previously implicated as a potentiator of fusion. Finally, exposure of phosphatidylserine on the surface of lipid-symmetric erythrocytes may be responsible for their enhanced fusion.  相似文献
