Rat hemidiaphragms were loaded with [U-14C] glucose at 2°C and subsequently incubated at 37°C with non-labeled glucose or [14C] glucose in the presence or absence of insulin. The incorporation of isotope into glycogen and lactate was determined. The results showed that insulin markedly stimulated the synthesis of glycogen from extracellular glucose while it had no effect on incorporation of isotope into glycogen from intracellular glucose. Lactate formation was not influenced by insulin. It was concluded that glucose transport in muscle is linked to glycogen sythesis and that insulin preferentially directs glucose entering the cell toward the formation of glycogen.  相似文献

We assessed the genetic structure and diversity of Reithrodontomys spectabilis, a critically endangered, endemic rodent from Cozumel Island, México. A total of 90 individuals were trapped from September 2001 to January 2005. Microsatellite data analysis revealed high genetic diversity values: a total of 113 alleles (average 12.5 per locus), H o  = 0.78, H e  = 0.80. These high values can be related to Cozumel’s size (478 km2) and extensive native vegetation cover, factors that could be promoting a suitable population size, high heterozygosity and the persistence of rare alleles in the species, as well as some long-term movement of individuals between sampling localities. A strong genetic structure was also observed, with at least four genetic groups, associated with a pattern of isolation by distance. We found a strong allelic and genetic differentiation shown between localities, with negligible recent gene flow and low inbreeding coefficients. The species life history and ecological characteristics—being nocturnal, semi-terrestrial, a good tree climber, having lunar phobia and significant edge effect—are likely affecting its genetic structure and differentiation. The high genetic diversity and population structure award R. spectabilis a significant conservation value. Our results can serve as a basis for future research and conservation of the species, particularly considering the problems the island is facing from habitat perturbation, urbanization and introduction of exotic species. In view of the structure and genetic variability observed, it is essential to establish and reinforce protected areas and management programs for the conservation of the endemic and endangered Cozumel Harvest mice.  相似文献

Characterization of spatial patterns of functional microbial communities could facilitate the understanding of the relationships between the ecology of microbial communities, the biogeochemical processes they perform and the corresponding ecosystem functions. Because of the important role the ammonia-oxidizing bacteria (AOB) and archaea (AOA) have in nitrogen cycling and nitrate leaching, we explored the spatial distribution of their activity, abundance and community composition across a 44-ha large farm divided into an organic and an integrated farming system. The spatial patterns were mapped by geostatistical modeling and correlations to soil properties and ecosystem functioning in terms of nitrate leaching were determined. All measured community components for both AOB and AOA exhibited spatial patterns at the hectare scale. The patchy patterns of community structures did not reflect the farming systems, but the AOB community was weakly related to differences in soil pH and moisture, whereas the AOA community to differences in soil pH and clay content. Soil properties related differently to the size of the communities, with soil organic carbon and total nitrogen correlating positively to AOB abundance, while clay content and pH showed a negative correlation to AOA abundance. Contrasting spatial patterns were observed for the abundance distributions of the two groups indicating that the AOB and AOA may occupy different niches in agro-ecosystems. In addition, the two communities correlated differently to community and ecosystem functions. Our results suggest that the AOA, not the AOB, were contributing to nitrate leaching at the site by providing substrate for the nitrite oxidizers.  相似文献

Members of the ATP binding cassette (ABC) transporter family perform a critical function in maintaining lipid homeostasis in cells as well as the transport of drugs. In this review, we provide an update on the ABCG-transporter subfamily member proteins, which include the homodimers ABCG1, ABCG2 and ABCG4 as well as the heterodimeric complex formed between ABCG5 and ABCG8. This review focusses on progress made in this field of research with respect to their function in health and disease and the recognised transporter substrates. We also provide an update on post-translational regulation, including by transporter substrates, and well as the involvement of microRNA as regulators of transporter expression and activity. In addition, we describe progress made in identifying structural elements that have been recognised as important for transport activity. We furthermore discuss the role of lipids such as cholesterol on the transport function of ABCG2, traditionally thought of as a drug transporter, and provide a model of potential cholesterol binding sites for ABCG2.  相似文献

Inter‐individual variation in phenotypic traits has long been considered as “noise” rather than meaningful phenotypic variation, with biological studies almost exclusively generating and reporting average responses for populations and species’ average responses. Here, we compare the use of an individual approach in the investigation of extracellular acid–base regulation by the purple sea urchin Paracentrotus lividus challenged with elevated pCO2 and temperature conditions, with a more traditional approach which generates and formally compares mean values. We detected a high level of inter‐individual variation in acid–base regulation parameters both within and between treatments. Comparing individual and mean values for the first (apparent) dissociation constant of the coelomic fluid for individual sea urchins resulted in substantially different (calculated) acid–base parameters, and models with stronger statistical support. While the approach using means showed that coelomic pCO2 was influenced by seawater pCO2 and temperature combined, the individual approach indicated that it was in fact seawater temperature in isolation that had a significant effect on coelomic pCO2. On the other hand, coelomic [HCO3?] appeared to be primarily affected by seawater pCO2, and less by seawater temperature, irrespective of the approach adopted. As a consequence, we suggest that individual variation in physiological traits needs to be considered, and where appropriate taken into account, in global change biology studies. It could be argued that an approach reliant on mean values is a “procedural error.” It produces an artefact, that is, a population's mean phenotype. While this may allow us to conduct relatively simple statistical analyses, it will not in all cases reflect, or take into account, the degree of (physiological) diversity present in natural populations.  相似文献

Four liverworts, Marchantia berteroana, M. foliacea, Plagiochila stephensoniana and Porella elegantula collected in New Zealand were chemically investigated. M. berteroana contains cuparene and (?)-2-hydroxycuparene as the major components. γ-Cadinene is the major component of M. foliacea. 4-Hydroxy-3′-methoxybibenzyl is a chemical marker for P. stephensoniana which belongs to the non-pungent type of Plagiochila species. P. elegantula synthesizes perrottetianal and belongs to the non-pungent type of Porella species.  相似文献

Previous authors have suggested that cyclical succession takes place inCalluna-dominated heathland, and is determined by the different growth phases of the dominant. Characteristic species are said to invade at different stages of the life cycle ofCalluna, particularly in the initial and pioneer phases and in the degenerate phase. In the last phase, gaps are formed due to separation and dying of the oldest frame branches.This paper aims to analyze in which microhabitat and in which phase of theCalluna life cycle seedling emergence and establishment is possible.More seedlings appear in pioneerCalluna than in matureCalluna stands in experimentally manipulated micro-sites. In pioneerCalluna establishment is affected only by application of litter, in matureCalluna by increased illumination. The differences between treatments disappeared after three years, but not the difference in establishment between stands.Seedling emergence from species sown experimentally was different in the successive phases ofCalluna. Highest emergence was in the pioneer stand.In a seedling survey also most seedlings appeared and established in the pioneer phase. The number of recently germinated seeds was high in all phases. However, seedling survival was very low, except in the pioneer stands. This study does not of itself show evidence for cyclical succession inCalluna-dominated heath vegetation. Colonization of gaps by seedlings ofCalluna in mature or degenerate phases is possible, but must be an infrequent occurrence.Nomenclauture follows Clapham, Tutin & Warburg (1962).We wish to thank Dr G.
