In recent years the incidence of Fusarium spp. isolated from Stem End Rot (SER) and Internal Core Rot (ICR) of grapefruit and oranges has increased markedly. Fusarium spp. were isolated from apparently healthy green buttons and from the tissue underneath the button of healthy fruits. Of all the SER- and ICR-rotted fruit, the incidence of Fusarium spp. alone and of Fusarium spp. with Alternaria citri was between 75 and 100%. Fusarium was also found to be the single causal agent of ICR of oranges and grapefruit. Species of Fusarium isolated from Stem End Rot and Internal Core Rot of citrus fruit were F. oxysporum, F. moniliforme and recently also F. culmorum.  相似文献

There is widespread agreement in neuroscience and psychology that the visual system identifies objects and faces based on a pattern of activation over many neurons, each neuron being involved in representing many different categories. The hypothesis that the visual system includes finely tuned neurons for specific objects or faces for the sake of identification, so‐called “grandmother cells”, is widely rejected. Here it is argued that the rejection of grandmother cells is premature. Grandmother cells constitute a hypothesis of how familiar visual categories are identified, but the primary evidence against this hypothesis comes from studies that have failed to observe neurons that selectively respond to unfamiliar stimuli. These findings are reviewed and it is shown that they are irrelevant. Neuroscientists need to better understand existing models of face and object identification that include grandmother cells and then compare the selectivity of these units with single neurons responding to stimuli that can be identified.  相似文献

Vertebrate skin appendages are incredibly diverse. This diversity, which includes structures such as scales, feathers, and hair, likely evolved from a shared anatomical placode, suggesting broad conservation of the early development of these organs. Some of the earliest known skin appendages are dentine and enamel-rich tooth-like structures, collectively known as odontodes. These appendages evolved over 450 million years ago. Elasmobranchs (sharks, skates, and rays) have retained these ancient skin appendages in the form of both dermal denticles (scales) and oral teeth. Despite our knowledge of denticle function in adult sharks, our understanding of their development and morphogenesis is less advanced. Even though denticles in sharks appear structurally similar to oral teeth, there has been limited data directly comparing the molecular development of these distinct elements. Here, we chart the development of denticles in the embryonic small-spotted catshark (Scyliorhinus canicula) and characterize the expression of conserved genes known to mediate dental development. We find that shark denticle development shares a vast gene expression signature with developing teeth. However, denticles have restricted regenerative potential, as they lack a sox2+ stem cell niche associated with the maintenance of a dental lamina, an essential requirement for continuous tooth replacement. We compare developing denticles to other skin appendages, including both sensory skin appendages and avian feathers. This reveals that denticles are not only tooth-like in structure, but that they also share an ancient developmental gene set that is likely common to all epidermal appendages.  相似文献

The morphology of the lumbar spine is crucial for upright posture and bipedal walking in hominids. The excellent preservation of the lumbar spine of Kebara 2 provides us a rare opportunity to observe a complete spine and explore its functionally relevant morphology. The lumbar spine of Kebara 2 is analyzed and compared with the lumbar spines of modern humans and late Pleistocene hominids. Although no size differences between the vertebral bodies and pedicles of Kebara 2 and modern humans are found, significant differences in the size and orientation of the transverse processes (L1‐L4), and the laminae (L5, S1) are demonstrated. The similarity in the size of the vertebral bodies and pedicles of Kebara 2 and modern humans suggests similarity in axial load transmission along the lumbar spine. The laterally projected (L2‐L4) and the cranially oriented (L1, L3) transverse processes of Kebara 2 show an advantage for lateral flexion of the lumbar spine compared with modern humans. The primary structure of glycolate oxidase from spinach has been determined. Six different types of peptide digest were investigated, utilizing CNBr, proteolytic enzymes, and chemical modifications to change a specificity of cleavage. In total, 90 peptides were purified and analyzed. The studies were aimed at correlation with crystallographic analysis of the same protein carried through in parallel and with cDNA studies which utilized initially determined amino acid sequences for synthesis of oligonucleotide probes. Continuous comparisons with the results from the crystallographic studies helped at an early stage to secure peptide overlaps, at the same time as the peptide data secured residue assignments in the electron density maps. In the end, all data agree and regions from all parts of the molecule have been checked by independent methods of analysis. The primary structure establishes the type of N-terminal post-translational processing, and yields information on segments not fully defined in electron density maps. Combined, the chemical, crystallographic, and cDNA data give extensive reliability. The peptide analysis shows that the N-terminus is blocked by acylation of the initiator methionine, which is in a primary structure typical for non-removal of the methionine in the processing events of the nascent protein chain. The molecule is comparatively rich in menthionine and some other generally less common residues, but has only one cysteine residue and no extensive hydrophobic segment. An amino acid sequence homology with flavocytochrome b2 from yeast, as expected from known similarities in tertiary structure, is observed (33% residue identities).  相似文献

Induction of nuclease and RNase activities, together with decreases in nucleic acid content are considered to be characteristics of senescence in higher plants. However, little is known about the specific identities or functions of the enzymes involved or the mechanisms controlling their activation. Here we report the identification of a 41-kDa-tomato nuclease, LeNUC1, which is specifically induced during tomato leaf senescence but not in ripening fruits. LeNUC1 is a glycoprotein, which can degrade both RNA and DNA and has optimal activity at pH 7.5–8. EDTA inhibits the activity of LeNUC1, while the addition of Co2+ or Mn2+ can restore its activity in the presence of the chelating agent. Interestingly, the activity of LeNUC1 is also induced in young leaves upon treatment with ethylene, which is known to be a senescence-promoting hormone in tomato. Constitutive activity of a 39-kDa nuclease, LeNUC2, similar in its biochemical requirements to LeNUC1, was also detected. LeNUC2 is not induced by ethylene and does not seem to be glycosylated. Based on their characteristics, LeNUC1 and LeNUC2 can be classified as Nuclease I enzymes. LeNUC1 may be involved in nucleic acid metabolism during tomato leaf senescence.  相似文献

The epithelium of the mammalian vagina arises from two distinct germ layers, endoderm from the urogenital sinus and mesoderm from the lower fused Müllerian ducts. While previously it has been reported that neonatal vaginal epithelium can be induced to differentiate as uterus, which normally develops from the middle portion of the Müllerian ducts, it has not been determined whether this ability is shared by both mesoderm- and endoderm-derived vaginal epithelia. To test if germ layer origin influences the ability of vaginal epithelium to undergo uterine differentiation, we have isolated sinus-derived and Müllerian-derived vaginal epithelia from newborn mice, combined them with uterine mesenchyme, and grown them for 4 weeks in female mice. Mesoderm-derived Müllerian vaginal epithelium in combination with uterine mesenchyme formed the simple columnar epithelium typical of uterus. Similar results were obtained with neonatal cervical epithelium, another mesodermal Müllerian duct derivative. On the other hand, sinus vaginal epithelium combined with uterine mesenchyme formed small cysts lined by a stratified squamous vaginal-like epithelium. This epithelium never showed evidence of cycling between the cornified and mucified states as is typically seen in vaginal epithelium combined with vaginal stroma. These results indicate that the ability of epithelium to form uterus is limited to mesoderm-derived epithelia and suggest that endoderm-derived sinus vaginal epithelium cannot undergo the typical differentiative modifications in response to the hormonal fluctuations of the estrous cycle when associated with uterine stroma.  相似文献
