We have demonstrated previously that the optimal method for inducing an antibody response against defined cancer antigens is covalent conjugation of the antigen to keyhole limpet hemocyanin (KLH) and use of the potent saponin adjuvant QS-21. Single molecules of glycolipids (tetrasaccharides, pentasaccharides, or hexasaccharides) and MUC1 peptides (containing between one and five MUC1 tandem repeats) conjugated to KLH have proven sufficient for antibody recognition and vaccine construction. However, cancer specificity of monoclonal antibodies against the monosaccharide Tn and disaccharide sTn comes largely from recognition of clusters (c) of these molecules on the cell surface. Tn consists of a monosaccharide (GalNAc) O-linked to serine or threonine on epithelial cancer mucins which are uniquely rich in serines and threonines. We test here several Tn constructs: Tn monosaccharide, Tn(c) prepared on a triple threonine backbone, and Tn prepared on a partially or fully glycosylated MUC1 backbone. We determine that Tn(c) is more effective than Tn, and conjugation to KLH is more effective than conjugation to BSA or polystyrene beads for inducing ELISA reactivity against Tn, and FACS reactivity against Tn-positive tumor cells. Surprisingly, MUC1 glycosylated with Tn at three or five sites per 20 amino acid MUC1 tandem repeat and conjugated to KLH, induced the strongest antibody response against Tn and tumor cells expressing Tn, and had the additional advantage of inducing antibodies against MUC1.  相似文献

Little has been published regarding the treatment of patients with long-established capsular contracture after previous submuscular or subglandular breast augmentation. This study reviews 7 years of experience in treating established capsular contracture after augmentation mammaplasty by relocating implants to the "dual-plane" or partly subpectoral position. A retrospective chart review was performed on all patients who were treated for capsular contracture using this technique between 1993 and 1999. Data collected included the date of the original augmentation, the original implant location, date of revision and type of implant used, length of follow-up, outcome, and any ensuing complications. Different surgical techniques were used, depending on whether the prior implant was located in a subglandular or submuscular plane. All patients had revisions such that their implants were relocated to a dual plane, with the superior two thirds or so of the implant located beneath the pectoralis major muscle and the inferior one third located subglandularly. Of 85 patients reviewed, 54 had their original implants in a submuscular position and 31 had their initial augmentation in a subglandular position. Of the 54 patients whose implants were initially submuscular, 23 patients (43 percent) had silicone gel implants, 15 patients (28 percent) had double-lumen implants, and the remaining 16 patients (30 percent) had saline implants. Of the 31 patients whose implants were initially subglandular, 20 patients (65 percent) had silicone gel implants, three patients (10 percent) had double-lumen implants, and the remaining eight patients (26 percent) had saline implants. Fifty-one patients (60 percent) had replacement with saline implants (37 smooth saline, 14 textured saline), whereas 34 (40 percent) had silicone gel implants (seven smooth gel, 27 textured gel). The average time from previous augmentation to revision was 9 years 9 months. The average follow-up time after conversion to the dual-plane position was 11.5 months. Only three of 85 patients required reoperation for complications, all of which involved some degree of implant malposition. Of patients converted to the dual plane, 98 percent were free of capsular contracture and were Baker class I at follow-up, whereas 2 percent were judged as Baker class II. There were no Baker level III or IV contractures at follow-up. The dual-plane method of breast augmentation has proved to be an effective technique for correcting established capsular contracture after previous augmentation mammaplasty. This technique appears to be effective when performed with either silicone or saline-filled implants.  相似文献

Assembly of a eukaryotic nucleus involves three distinct events: membrane recruitment, fusion to form a double nuclear membrane, and nuclear pore complex (NPC) assembly. We report that importin beta negatively regulates two of these events, membrane fusion and NPC assembly. When excess importin beta is added to a full Xenopus nuclear reconstitution reaction, vesicles are recruited to chromatin but their fusion is blocked. The importin beta down-regulation of membrane fusion is Ran-GTP reversible. Indeed, excess RanGTP (RanQ69L) alone stimulates excessive membrane fusion, leading to intranuclear membrane tubules and cytoplasmic annulate lamellae-like structures. We propose that a precise balance of importin beta to Ran is required to create a correct double nuclear membrane and simultaneously to repress undesirable fusion events. Interestingly, truncated importin beta 45-462 allows membrane fusion but produces nuclei lacking any NPCs. This reveals distinct importin beta-regulation of NPC assembly. Excess full-length importin beta and beta 45-462 act similarly when added to prefused nuclear intermediates, i.e., both block NPC assembly. The importin beta NPC block, which maps downstream of GTPgammaS and BAPTA-sensitive steps in NPC assembly, is reversible by cytosol. Remarkably, it is not reversible by 25 microM RanGTP, a concentration that easily reverses fusion inhibition. This report, using a full reconstitution system and natural chromatin substrates, significantly expands the repertoire of importin beta. Its roles now encompass negative regulation of two of the major events of nuclear assembly: membrane fusion and NPC assembly.  相似文献

Ghrelin is a recently discovered stomach hormone that stimulates pituitary growth hormone (GH) secretion potently. The purpose of these experiments was to test the hypothesis that a stomach-ghrelin-pituitary-GH axis exists in which either an elevation or reduction in systemic GH levels will exert a negative or positive feedback action, respectively, on stomach ghrelin homeostasis. In rats, GH administration decreased stomach ghrelin mRNA levels and plasma ghrelin levels significantly. In GH-releasing hormone (GHRH) transgenic mice, GHRH overexpression decreased stomach ghrelin peptide levels when compared with control mice. In aged rats (25 months) stomach ghrelin mRNA and peptide levels and plasma ghrelin levels were decreased when compared with young rats (5 months). Because GH secretion is reduced in aged rats, the elevated stomach ghrelin production and secretion may reflect a decreased GH feedback on stomach ghrelin, homeostasis, and secretion. Together, these findings suggest that endogenous pituitary GH exerts a feedback action on stomach ghrelin homeostasis and support the hypothesis that a stomach-ghrelin-pituitary GH axis exists.  相似文献

Structural features of the pulvinus of the solar-tracking leaf of Lavatera cretica L. that are involved in its capacity for omnidirectional and fully reversible bending in response to vectorial excitation of the lamina were studied by light- and scanning electron microscopy. Pulvini that had bent in the plane at right angles to the midvein were bisected along that plane and the symmetrical tissues of the expanded and contracted flanks were compared. The pulvinus contains a central vascular core and exhibits a transversely furrowed exterior. These specialized features enable the fully mature tissue of this region of the petiole to bend reversibly. The epidermis, chlorenchyma, peripheral collenchyma, and cortical parenchyma in the pulvinus form concentric, radially symmetric sheaths around the vascular core and exhibit structural features in their cell walls that would allow considerable changes in cell volume and consequently enable the omnidirectional bending of this pulvinus. Thickened wall portions of the pulvinar epidermis and peripheral collenchyma exhibit a highly specialized architecture, consisting of alternating thick and thin strips, that enhances their flexibility, while maintaining mechanical support. Cell walls of the chlorenchyma and the cortical parenchyma are thin and capable of reversible infolding. Those of the cortical parenchyma also exhibit numerous prominent transverse pit fields, indicative of anisotropic orientation of their microfibrillar lattice transverse to the pulvinar axis. This orientation is compatible with elastic deformation of the cortical parenchyma cells along the pulvinar axis. Filament-like cytoplasmic strands were observed along the walls of pulvinar motor cells, predominantly transverse to the pulvinar axis, but their function (if any) in volume changes of these cells is unknown.  相似文献

Mammalian cells employ a network of DNA repair pathways. DNA repair is required during development to ensure accuracy of DNA replication in the rapidly dividing embryonic cells and to maintain genomic integrity in the mature organism. An enzyme involved in repair of replication errors generated on either normal or oxidatively damaged DNA templates, is the mammalian ortholog of the Escherichia coli MutY DNA glycosylase (MYH). We show that levels of MYH isoform, detected at the E14 embryonic stage, decrease during embryonic and neonatal rat development, while new isoforms appear and gradually increase in the neonate and adult brain. The temporally declining expression of embryonic MYH resembles the pattern of proliferating cell nuclear antigen (PCNA) decline during this period. Immunohistochemical analyses of the embryonic brain show that cells staining for MYH initially coincide with cells staining for PCNA. At later stages PCNA declines, while MYH is detected primarily outside the nucleus. MutY-like glycosylase activity for adenines misincorporated opposite oxidized guanines is detected in both, embryonic and adult brain extracts. Together, these findings suggest that in proliferating embryonic cells, MYH might be primarily involved in post replicative repair of nuclear DNA, whereas in post mitotic neurons, in the repair of mitochondrial DNA.  相似文献

Ghrelin is a recently discovered stomach hormone whose secretion increases with fasting; the fasting-induced elevation is inhibited by refeeding. The aim of this study was to determine whether all nutrient types (i.e., carbohydrates, proteins, fats) and soybean trypsin inhibitor (SBTI), a secretagogue for intestinal cholecystokinin (CCK), given individually into the stomach or intravenously can inhibit ghrelin secretion in the fasted rat. Intragastric (i.g.) administration of intact protein, a protein digest, SBTI, dextrose, or fat decreased plasma ghrelin levels significantly (p<0.05). All nutrients inhibited ghrelin secretion equally. Fat and dextrose given intravenously (i.v.) also reduced ghrelin secretion. These data demonstrate that nutrients can inhibit ghrelin secretion by both the luminal and systemic routes. Additionally, the findings show that all nutrient types given orally are capable of inhibiting ghrelin secretion, and suggest that intestinal CCK may participate in the inhibition of ghrelin secretion following oral intake of nutrients.  相似文献
