The method of Green's functions is applied to the convection-diffusion equation describing gas transport in the human lung. Explicit analytic solutions are found for the zeroth approximation to the value of steady state alveolar oxygen concentration, given the frequency of breathing, tidal volume, oxygen consumption, alveolar volume, volume of the bronchial tree, and two modelling parameters describing the geometry. The results of the calculation are compared favorably with some experimental data.  相似文献

The enzyme rhodanese (thiosulfate sulfurtransferase, EC 2.8.1.1) is inactivated on incubation with reducing sugars such as glucose, mannose, or fructose, but is stable with non-reducing sugars or related polyhydroxy compounds. The enzyme is inactivated with (ES) or without (E) the transferable sulfur atom, although E is considerably more sensitive, and inactivation is accentuated by cyanide. Inactivation of E is accompanied by increased proteolytic susceptibility, a decreased sulfhydryl titer, a red-shift and quenching of the protein fluorescence, and the appearance of hydrophobic surfaces. Superoxide dismutase and/or catalase protect rhodanese. Inactive enzyme can be partially reactivated during assay and almost completely reactivated by incubation with thiosulfate, lauryl maltoside, and 2-mercaptoethanol. These results are similar to those observed when rhodanese is inactivated by hydrogen peroxide. These observations, as well as the cyanide-dependent, oxidative inactivation by phenylglyoxal, are explained by invoking the formation of reactive oxygen species such as superoxide or hydrogen peroxide from autooxidation of alpha-hydroxy carbonyl compounds, which can be facilitated by cyanide.  相似文献

Our recent anterior drawer studies in human cadaveric knees [Guan and Butler, Adv. Bioengng 17, 5 (1990); Guan et al., Trans. orthop. Res. Soc. 16, 589 (1991)] have suggested that anterior bundles of the anterior cruciate ligament (ACL) develop higher load-related material properties than posterior bundles. This was confirmed when we reevaluated the axial failure data for these bundle-bone specimens from an earlier study [Butler et al., J. Biomechanics 19, 425-432 (1986)]. The purpose of this study was to determine, in a larger data set, if anteromedial and anterolateral bundles of the anterior cruciate ligament exhibit significantly larger load-related material properties than the posterior ligament bundles. Seven ACL-bone units from seven donors (the three tissues from the original study plus four new ones) were subdivided into three subunits, preserving the bone insertions. The subunits were failed in tension at a constant strain rate (100% s-1) and four material properties were compared within and between donors. The anterior bundles developed significantly larger moduli, maximum stresses, and strain energy densities to maximum stress than the posterior subunits. Moduli for the anterior vs posterior subunits averaged 284 MPa vs 155 MPa, maximum stresses averaged 38 MPa vs 15 MPa, and strain energy densities averaged 2.7 N m cc-1 vs 1.1 N m cc-1, respectively. No significant differences were found, however, among strains to maximum stress or between any of the other properties for the two anterior subunits. These results are important to the design of ligament replacements and suggest new experiments designed to distinguish in vivo force levels in these ACL bands, a possible reason for the material differences.  相似文献

Concentrations of ATP and ADP were measured in 156 sheep embryos by means of an ultramicrofluorescence assay. Stages of preimplantation development measured included unfertilized oocytes through blastocyst-stage embryos. ATP concentrations remained constant through the 8-cell stage; then ATP decreased significantly (p < 0.025) at the morula stage and remained low through the blastocyst stage. ADP concentrations did not change throughout the embryonic stages measured. Decreased levels of ATP with constant levels of ADP caused the ATP:ADP ratio to decrease significantly (p < 0.025) between the 8-cell and morula stages. We suggest that the increase in glucose uptake by sheep embryos observed at the morula stage of development may be due, in part, to a decrease in the ATP:ADP ratio.  相似文献

The three-dimensional structure of the tobacco mosaic virus (TMV) coat protein disk suggests a possible pathway for the early evolution of the virus self-assembly mechanism.The coat protein contains a 2-fold repeated structural pattern in the folding of both its four alpha helices (A,B,C,D), which run alternately forward and back along the radius of the disk, and the four-stranded antiparallel pleated sheet which links these helices to the hydrophobic girdle at the outer rim of the disk. Helices A and B can be approximately superposed on C and D by a screw rotation about a molecular pseudo-dyad axis which lies nearly parallel to the plane of the protein disk. This operation relates 29 pairs of α-carbon positions with a root-mean-square deviation of 1.77 Å. A second pseudo-dyad in the pleated-sheet region relates 14 more atom pairs with a deviation of 2.32 Å and forms a distorted continuation of the relationship between the helices. The helix dyad also relates repeated pairs of functionally important amino acids which take part in intersubunit contacts.We have analysed these structural repeats and tested their significance by comparing them with repeats in other “helix quartet” proteins, cytochrome b5 and the hemerythrins, as well as with an irregular helix cluster in thermolysin. TMV is noticeably more repetitive than the others, including hemerythrin which is thought to have evolved by gene duplication.We propose that the primitive TMV coat protein was a dimeric structure of two smaller units paired about a 2-fold axis. Each unit was a pair of helices, linked at the inner radius of the virus rod by a short bend, where the RNA binding site formed, and connected at the outer radius by two short strands of beta sheet. A tandem gene duplication joined the two units and formed the present helix quartet. The flexible loop which now runs into the centre of the virus and connects helix C to helix D developed later. The assembly origin RNA may have evolved from part of the coat protein RNA which codes for this loop.  相似文献

A study of the treatment of dysphasia after stroke compared the progress of two groups of disabled patients. One received conventional treatment from qualified speech therapists and the other from non-professional volunteers. Methods of assessing communication difficulties were also compared and the impact of aphasic illness on families examined. No important differences in the results of treatment were seen between the two groups. The volunteers, however, often had to assume some of the responsibilities of social workers, and transport to hospital created practical and economic problems. It is concluded that the two forms of treatment provide essentially the same benefit, although doubt must still remain because relatively few patients were studied.  相似文献

With new legislation and favorable case law developments, many providers of health care services are turning to binding arbitration agreements as a partial solution to the medical malpractice crisis. Existing data indicate tremendous advantages can be gained from the use of such agreements, if they are carefully drawn to comply with new law, tailored to the particular situation in which they will be used and coupled with appropriate procedures to secure the intended benefits.Arbitration is not a substitute for insurance, but if a provider has taken the calculated risk of forgoing insurance he should not be without a carefully drawn binding arbitration agreement.  相似文献

The phosphoprotein of rat incisors has been purified by successive gel and ion-exchange chromatography. The product gave a single band on polyacrylamide gel electrophoresis and contained approximately 34% phosphoserine and 32% aspartic acid. Alkaline elimination experiments showed all the phosphate to be present as phosphoserine. Ultraviolet spectra in the presence or absence of ATP showed that the phosphoprotein did not contain an nucleotide moiety as suggested by Veis, A., Spector, A. R. and Zamoscianyk, H. ((1972) Biochim. Biophys. Acta 257, 404-413) for bovine dentin phosphoprotein.  相似文献
