Objective: To detect BRAF V600E mutation in thyroid fine-needle aspiration (FNA) slides and needle rinses (NR). Study Design: Tumor-enriched DNA was extracted from FNA smears, formalin-fixed paraffin-embedded (FFPE) sections, or NR specimens from 37 patients with confirmed papillary thyroid carcinoma or benign findings. An allele-specific primer selectively amplified the 1799 T>A BRAF mutation while simultaneously blocking amplification of wild-type (WT) BRAF with an unlabeled probe during PCR. Mutation detection was accomplished by melting analysis of the probe. Results: Allele-specific/blocking probe PCR confirmed the BRAF mutation status for 20 of 24 paired FNA/FFPE samples previously tested by fluorescent probe real-time PCR. For the other 4 cases, the sensitive PCR method detected the BRAF mutation in all paired FNA/FFPE samples. Previously, the mutation had been detected in only the FFPE samples. The BRAF mutation was also detected in some NR specimens. Conclusion: Treatment of patients with thyroid nodules is guided by FNA biopsy, which can be scantly cellular, necessitating a sensitive test that can detect low levels of BRAF V600E mutation in a WT background. We report increased detection of BRAF V600E in FNA specimens using allele-specific/blocking probe PCR, which has an analytical sensitivity of 0.01%.  相似文献

The formation of inorganic materials with complex form is a widespread biological phenomenon (biomineralization). Among the most spectacular examples of biomineralization is the production by diatoms (a group of eukaryotic microalgae) of intricately nanopatterned to micropatterned cell walls made of silica (SiO2). Understanding the molecular mechanisms of diatom silica biomineralization is not only a fundamental biological problem, but also of great interest in materials engineering, as the biological self-assembly of three-dimensional (3D) inorganic nanomaterials has no man-made analog. Recently, insight into the molecular mechanism of diatom silica formation has been obtained by structural and functional analysis of biomolecules that are involved in this process. Furthermore, the rapid development of diatom molecular genetics has provided new tools for investigating the silica forming machinery of diatoms and for manipulating silica biogenesis. This has opened the door for the production, through genetic engineering, of unique 3D nanomaterials with designed structures and functionalities.  相似文献

In a previous investigation, I have shown that the kinetics of the Ca uniporter change fundamentally when mitochondria have transitorily lost their membrane potential. The sigmoidal kinetics, usually observed in liver mitochondria, became almost hyperbolic. This means an increase in the affinity for calcium, and hence a considerable acceleration of Ca uptake in the range of low, e.g., physiological calcium concentration. In this investigation I show that extramitochondrial calcium released from the deenergized mitochondria causes the allosteric activation of the Ca uniporter. The dependence of the allosterical activation on the extramitochondrial Ca2+ concentration and on time is described. It is also reported that it is possible to activate allosterically the Ca uniporter of energized mitochondria by a short-term elevation of the extramitochondrial Ca2+ concentration. The process of activation is reversible. It is quickly reversed by the addition of chelators for Ca2+, and it is slowly reversed when the activating Ca2+ has to be removed by the mitochondrial Ca uniporter, though the bulk of extramitochondrial calcium is taken up by it very quickly. Several kinetics of the Ca uniporter are described. The implications of continually changing kinetics of the Ca uniporter are considered for carbon tetrachloride intoxication and the action of alpha 1-adrenergic agonists in liver cells.  相似文献

Summary A new form of ectodermal dysplasia was observed in two siblings, offspring of healthy non-consanguineous parents. The main findings in both children are: hypodontia, abnormally shaped teeth, scalp hypotrichosis, pili annulati, follicular hyperkeratosis on the trunk and limbs, intensified delineation and reticular hyperpigmentation of the nape, and hyperopia; one of the siblings also has astigmatism. As both patients have normal nails and are euhidrotic, this is an ectodermal dysplasia of the pilodental subgroup. The cause is probably genetic and autosomal-recessive inheritance is most likely.  相似文献

Reports in recent years indicate that the increasing emergence of resistance to drugs be using to TB treatment. The resistance to them severely affects to options for effective treatment. The emergence of multidrug-resistant tuberculosis has increased interest in understanding the mechanism of drug resistance in M. tuberculosis and the development of new therapeutics, diagnostics and vaccines. In this study, a label-free quantitative proteomics approach has been used to analyze proteome of multidrug-resistant and susceptible clinical isolates of M. tuberculosis and identify differences in protein abundance between the two groups. With this approach, we were able to identify a total of 1,583 proteins. The majority of identified proteins have predicted roles in lipid metabolism, intermediary metabolism, cell wall and cell processes. Comparative analysis revealed that 68 proteins identified by at least two peptides showed significant differences of at least twofolds in relative abundance between two groups. In all protein differences, the increase of some considering proteins such as NADH dehydrogenase, probable aldehyde dehydrogenase, cyclopropane mycolic acid synthase 3, probable arabinosyltransferase A, putative lipoprotein, uncharacterized oxidoreductase and six membrane proteins in resistant isolates might be involved in the drug resistance and to be potential diagnostic protein targets. The decrease in abundance of proteins related to secretion system and immunogenicity (ESAT-6-like proteins, ESX-1 secretion system associated proteins, O-antigen export system and MPT63) in the multidrug-resistant strains can be a defensive mechanism undertaken by the resistant cell.

When establishing a new breeding group of Naked mole-rats three significant hurdles are to overcome: 1. Determination of sex. 2. Get over the group specific smell (between individuals of different groups). 3. Get over the reproductive blockade. This report describes experiences achieving these goals as well as establishes requirements for a successful breeding. The sex determination of nonreproductive Naked mole rats is tainted with great uncertainty, because the anogenital region of both sexes appears quite similar. For a lack of births and a failure of rearing offspring the following reasons seem to be responsible: 1. Incorrect sex determination at the time of group founding. 2. The reproductive blockade could not be overcome because of disadvantageous hierarchical relations within the group. 3. Pregnancy is interrupted due to disturbances from outside the enclosure or social stress within the group. 4. Due to these disturbances the alpha-female is not often enough willing to suckle her youngsters.  相似文献
