? Premise of the study: Modeling the contemporary and future climate niche for rare plants is a major hurdle in conservation, yet such projections are necessary to prevent extinctions that may result from climate change. ? Methods: We used recently developed spline climatic models and modified Random Forests statistical procedures to predict suitable habitats of three rare, endangered spruces of Mexico and a spruce of the southwestern USA. We used three general circulation models and two sets of carbon emission scenarios (optimistic and pessimistic) for future climates. ? Key results: Our procedures predicted present occurrence perfectly. For the decades 2030, 2060, and 2090, the ranges of all taxa progressively decreased, to the point of transient disappearance for one species in the decade 2060 but reappearance in 2090. Contrary to intuition, habitat did not develop to the north for any of the Mexican taxa; rather, climate niches for two taxa re-materialized several hundred kilometers southward in the Trans-Mexican Volcanic Belt. The climate niche for a third Mexican taxon shrank drastically, and its two mitotypes responded differently, one of the first demonstrations of the importance of intraspecific genetic variation in climate niches. The climate niche of the U.S. species shrank northward and upward in elevation. ? Conclusion: The results are important for conservation of these species and are of general significance for conservation by assisted colonization. We conclude that our procedures for producing models and projecting the climate niches of Mexican spruces provide a way for handling other rare plants, which constitute the great bulk of the world's endangered and most vulnerable flora.  相似文献

Comparative folliculogenesis and spermatogenesis of four teleost fish from a Reservoir in south‐eastern Brazil. —Acta Zoologica (Stockholm). 91 : 466–473. This study provides a comparative analysis of gametogenesis of neotropical teleosts Metynnis maculatus, Megalancistrus parananus, Cichla kelberi and Satanoperca pappaterra, through histological, histochemical and histometric techniques. In the ooplasm of C. kelberi and S. pappaterra conspicuous lipid vesicles were observed, which are characteristic of pelagic eggs produced by marine fishes. Perinucleolar oocytes were identified in the testis of S. pappaterra suggesting that this species is protogynous without functional hermaphroditism, an unusual pattern for neotropical fresh‐water fishes. The spermatozoa of the studied species have rounded heads, a characteristic of fish that externally fertilise their eggs. The follicular (granulosa) cells of the vitellogenic oocytes from the studied species were cuboidal or columnar, however, in C. kelberi there were columnar follicular cells at the vegetal pole and cuboidal cells at the animal pole. Variations of the histochemical content were detected in the cortical alveoli and follicular cells of vitellogenic oocytes showing differences in the mechanisms to block polyspermy and egg adhesiveness. Larger oocytes were recorded in species which demonstrated parental care behaviour and smaller oocytes were noted in those species with fractioned spawning.  相似文献

The regeneration of somatic seedlings from selected 100-year-old cork oak trees is reported. The induction of somatic embryogenesis from leaves of epicormic shoots was significantly affected by genotype, harvesting time and their interaction. Leaves from all five selected trees produced somatic embryos when the segments of branches used as sources of epicormic shoots were collected in May. Genotype, but not the level of photosynthetically active radiation, affected the proliferation of the embryogenic lines and the number of detachable embryos that could be obtained from them. Genotype also affected several steps leading to conversion of somatic embryos, from germination to complete acclimatisation of somatic seedlings. Almost 40% of the somatic embryos from all lines germinated, showing coordinated root and shoot growth. Although the mean percentage of recovery for the whole process was low, plants could be regenerated from four of the five trees tested.  相似文献

A transformation system for selected mature cork oak (Quercus suber L.) trees using Agrobacterium tumefaciens has been established. Embryos obtained from recurrent proliferating embryogenic masses were inoculated with A. tumefaciens strains EHA105, LBA4404 or AGL1 harbouring the plasmid pBINUbiGUSint [carrying the neomycin phosphotransferase II (nptII) and -glucuronidase (uidA) genes]. The highest transformation efficiency (4%) was obtained when freshly isolated explants were inoculated with A. tumefaciens strain AGL1. Evidence of stable transgene integration was obtained by PCR for the nptII and uidA genes, Southern blotting and expression of the uidA gene. Plant tissue culture techniques are carried out under environmentally controlled conditions in phytotrons. However, electric components of phytotrons generate electromagnetic fields that may act as a environmental factor influencing plant growth and morphogenesis. Isolated somatic embryos of Quercus suber, picked from embryogenic lines, were chronically exposed to a 50 Hz and 15 μT electromagnetic field generated in a Helmholtz-coil system for 8 weeks, in order to examine if the extremely low frequency (ELF) magnetic field (MF) affected the morphogenic behaviour of embryogenic cultures during recurrent embryogenesis. Germination of somatic embryos from genotype G7.1 was carried out under the same electromagnetic field, and also under conditions in which the local geomagnetic field was suppressed. The ELF MF did not influence the growth of embryogenic clumps of the assayed genotypes, but reduced the number of detachable embryos produced by genotype G3.27. The ELF MF did not modify the percentages of germination or plant formation of somatic embryos. However, somatic embryos had better germination when cultured under the suppressed geomagnetic field condition. An experiment was conducted to determine soil and plant resistance to water flow in faba bean under field conditions during the growing season. During each sampling period transpiration flux and leaf water potential measured hourly were used with daily measurements of root and soil water potential to calculate total resistance using Ohm's law analogy. Plant growth, root density and soil water content distributions with depth were measured. Leaf area and root length per plant reached their maximum value during flowering and pod setting (0.31 m2 and 2200 m, respectively), then decreasing until the end of the growing period. Root distribution decreased with depth ranging, on average, between 34.2% (in the 0–0.25 m soil layer) and 18.1% (in the 0.75–1.0 m soil layer). Mean root diameter was 0.6 mm but most of the roots were less than 0.7 mm in diameter. Changes in plant and soil water potentials reflected plant growth characteristics and climatic patterns. The overall relationship between the difference in water potential between soil and leaf and transpiration was linear, with the slope equal to average plant resistance (0.0165 MPa/(cm3 m-1 h-1 10-3). Different regression parameters were obtained for the various measurement days. The water potential difference was inversely related to transpiration at high leaf stomatal resistance and at high values of VPD. Total resistance decreased with transpiration flux in a linear relationship (r=−0.68). Different slope values were obtained for the different measurement days. Estimated soil resistance was much lower than the observed total resistance to water flow. The change from vegetative growth to pod filling was accompanied by an increase in plant resistance. The experimental results support previous findings that resistance to water flow through plants is not constant but is influenced by plant age, growth stage and environmental conditions. A more complex model than Ohm's law analogy may be necessary for describing the dynamic flow system under field conditions. The genetic diversity of Cistus ladanifer ssp. ladanifer (Cistaceae) growing on ultramafic and non-ultramafic (basic and schists) soils in the NE of Portugal was studied in order to identify molecular markers that could distinguish the metal-tolerant ecotypes of this species. Random Amplified Polymorphic DNA (RAPD) markers were used in order to estimate genetic variation and differences between populations. The RAPD dataset was analysed by means of a cluster analysis and an analysis of molecular variance (AMOVA). Our results indicate a significant partitioning of molecular variance between ultramafic and non-ultramafic populations of Cistus ladanifer, although the highest percentage of this variance was found at the intra-population level. Mantel's test showed no relationship between inter-population genetic and geographic distances. A series of RAPD bands that could be related to heavy metal tolerance were observed. The identification of such markers will enable the use of Cistus ladanifer in phytoremediation procedures.  相似文献

We tried to elucidate the effects of a brief and severe model of food restriction on insulin sensitivity in female rats, focusing on key proteins involved in the insulin signalling pathway in skeletal muscle and adipose tissue after 5, 10 and 15 days of food restriction. Using euglycemic clamp, we detected that food-restricted rats are significantly less sensitive to insulin action than control rats. However, the time of restriction promotes a progressive increase on insulin sensitivity. The analysis of the insulin signalling pathway showed a tissue-specific regulation of several proteins involved in insulin signalling. In skeletal muscle, insulin receptor substrate 1 and Glut4 are up-regulated at the end of the food restriction period, just the opposite of what we found in adipose tissue. In conclusion, a 50% reduction of food intake modulates insulin sensitivity through a tissue-specific regulation of the insulin signalling pathway in the main target tissues for this hormone.  相似文献

In lightning-induced fire risk prediction models, the number of potential predictors is usually high, with some redundancy among them. It is therefore important to select the best subset of predictors that obtain models with the greatest discrimination capacity. With this aim in mind, the logistic generalized linear model was used to estimate lightning-induced fire occurrence using a case study of the province of León (northwest Spain). A bootstrap-based test was used to obtain the optimal number of predictors and to model this optimal number of predictors displaying the largest area under the receiver operating characteristics curve. The results show that of the 16 variables initially considered, only three were necessary to obtain the model with the best discriminatory capacity for estimating lightning-induced fire occurrence. Moreover, this model can be considered equivalent to another nine alternative models with three covariates. Both the optimal and the equivalent models are useful in the spatially explicit assessment of fire risk, the planning and coordination of regional efforts to identify areas at greatest risk, and the design of long-term wildfire management strategies. The methodology used for this case study can be applied to other wildfire risk assessment situations where multiple and interconnected covariates are available.  相似文献
