The preparation and evaluation of novel aryl urea analogs as broad-spectrum antibacterial agents is described. Numerous compounds showed low micromolar minimum inhibitory concentrations (MIC) against both Gram-positive and Gram-negative bacteria. Evidence for spontaneous mappings between the dimensions of number and length, time and length, and number and time, has been recently described in preverbal infants. It is unclear, however, whether these abilities reflect the existence of privileged mappings between certain quantitative dimensions, like number, space and time, or instead the existence of a magnitude system underlying the representation of any quantitative dimension, and allowing mappings across those dimensions. Four experiments, using the same methods from previous research that revealed a number-length mapping in eight-month-old infants, investigated whether infants of the same age establish mappings between number and a different, non-spatial continuous dimension: level of brightness. We show that infants are able to learn and productively use mappings between brightness and number when they are positively related, i.e., larger numbers paired with brighter or higher contrast levels, and fail when they are inversely related, i.e., smaller numbers paired with brighter or higher contrast levels, suggesting that they are able to learn this mapping in a specific direction. However, infants not only do not show any baseline preference for any direction of the number-brightness mapping, but fail at transferring the discrimination from one dimension (number) to the other (brightness). Although infants can map multiple dimensions to one another, the number-length mapping may be privileged early in development, as it is for adults. 相似文献

BACKGROUND: Axon migrations are guided by extracellular cues that can act as repellants or attractants. However, the logic underlying the manner through which attractive and repulsive responses are determined is unclear. Many extracellular guidance cues, and the cellular components that mediate their signals, have been implicated in both types of responses. RESULTS: Genetic analyses indicate that MIG-10/RIAM/lamellipodin, a cytoplasmic adaptor protein, functions downstream of the attractive guidance cue UNC-6/netrin and the repulsive guidance cue SLT-1/slit to direct the ventral migration of the AVM and PVM axons in C. elegans. Furthermore, overexpression of MIG-10 in the absence of UNC-6 and SLT-1 induces a multipolar phenotype with undirected outgrowths. Addition of either UNC-6 or SLT-1 causes the neurons to become monopolar. Moreover, the ability of UNC-6 or SLT-1 to direct the axon ventrally is enhanced by the MIG-10 overexpression. We also demonstrate that an interaction between MIG-10 and UNC-34, a protein that promotes actin-filament extension, is important in the response to guidance cues and that MIG-10 colocalizes with actin in cultured cells, where it can induce the formation of lamellipodia. CONCLUSIONS: We conclude that MIG-10 mediates the guidance of AVM and PVM axons in response to the extracellular UNC-6 and SLT-1 guidance cues. The attractive and repulsive guidance cues orient MIG-10-dependant axon outgrowth to cause a directional response. 相似文献

The Cu(II) and Ag(I) complexes, [Cu(phendio)3](ClO4)24H2O and [Ag(phendio)2]ClO4 (phendio = 1,10-phenanthroline-5,6-dione), are prepared in good yield by reacting phendio with the appropriate metal perchlorate salt. The X-ray crystal structure of the Ag(I) complex shows it to have a pseudo tetrahedral structure. `Metal-free' phendio and the Cu(II) and Ag(I) phendio complexes strongly inhibit the growth of the fungal pathogen Candida albicans, and are more active than their 1,10-phenanthroline analogues. The simple Ag(I) salts, AgCH3CO2, AgNO3 and AgClO4.H2O display superior anti-fungal properties compared to analogous simple Cu(II) and Mn(II) salts, suggesting that the nature of the metal ion strongly influences activity. Exposing C. albicans to `metal-free' phendio, simple Ag(I) salts and [Ag(phendio)2]ClO4 causes extensive, non-specific DNA cleavage. `Metal-free' phendio and [Ag(phendio)2]ClO4 induce gross distortions in fungal cell morphology and there is evidence for disruption of cell division. Both drugs also exhibit high anti-cancer activity when tested against cultured mammalian cells. 相似文献

Robust, polymorphic microsatellite DNA markers (simple sequence repeats—SSRs) are valuable tools for a range of tree conservation
and breeding applications. SSRs are routinely used in the study of population genetic structure and diversity, pedigree reconstruction
and genetic linkage mapping. Their abundance in the genome, co-dominant inheritance and potential for cross-species amplification
make microsatellites highly prized markers. This paper characterises 22 novel genomic polymorphic microsatellite loci for
Sitka spruce (Picea sitchensis (Bong.) Carr.). Amplification of DNA from Sitka spruce material was carried out both with a set of unrelated trees to obtain
diversity statistics for each locus, and with the progeny of a full-sib family to test simple Mendelian inheritance. Observed
heterozygosity ranged from 0.38 to 0.91 and allele number per locus ranged from 6 to 21, with a mean of 12.2. In addition,
the primer pairs were tested with DNA from Norway spruce (P. abies) and white spruce (P. glauca) to investigate their potential for cross-species amplification and ten loci amplified in all three species. The results
from these genomic microsatellites are compared to data generated from microsatellites derived from Picea EST libraries. In summary, this novel, highly polymorphic markers represent a significant addition to the rapidly expanding
Picea genomics tool-box.

This study measured the mean age, duration, and sequence of the emergence of permanent dentition in Nigerian children and compared the findings with other population groups. The cross‐sectional study involved 1,078 Nigerian children, aged 4–16 years old, from selected primary and secondary schools in the Ife Central local government area in Ile‐Ife, Osun State. In general, compared to boys, girls had an earlier mean age of emergence of all the permanent teeth. Children from high socioeconomic class had an earlier mean age of emergence for the maxillary incisors (6.43 and 7.58 years) and mandibular incisors (5.28 and 6.44 years) compared to children from middle and low socioeconomic classes, although socioeconomic effects were more mixed for premolars and molars. Compared to their counterparts in the USA, Australia, Belgium, and Iran, Nigerian children showed an earlier mean age of emergence of all the permanent teeth studied except for Pakistani boys, who had an earlier mean age of emergence of the maxillary premolars and second molar. Poorer economic status has been associated with delayed dental development; however, when compared to other populations, the Nigerian children in this study have earlier mean emergence ages than children from wealthier countries such as the USA and Australia. Arabinogalactan proteins (AGPs) are a family of extracellular plant proteoglycans implicated in many aspects of plant growth and development, including in vitro somatic embryogenesis (SE). We found that specific AGPs were produced by cotton (Gossypium hirsutum) calli undergoing SE and that when these AGPs were isolated and incorporated into tissue culture medium, cotton SE was promoted. When the AGPs were partly or fully deglycosylated, SE-promoting activity was not diminished. Testing of AGPs separated by reverse-phase high-performance liquid chromatography revealed that the SE-promoting activity resided in a hydrophobic fraction. We cloned a full-length complementary DNA (cotton PHYTOCYANIN-LIKE ARABINOGALACTAN-PROTEIN1 [GhPLA1]) that encoded the protein backbone of an AGP in the active fraction. It has a chimeric structure comprising an amino-terminal signal sequence, a phytocyanin-like domain, an AGP-like domain, and a hydrophobic carboxyl-terminal domain. Recombinant production of GhPLA1 in tobacco (Nicotiana tabacum) cells enabled us to purify and analyze a single glycosylated AGP and to demonstrate that this chimeric AGP promotes cotton SE. Furthermore, the nonglycosylated phytocyanin-like domain from GhPLA1, which was bacterially produced, also promoted SE, indicating that the glycosylated AGP domain was unnecessary for in vitro activity.Arabinogalactan proteins (AGPs) comprise a diverse group of plant proteoglycans (for review, see Fincher et al., 1993; Nothnagel, 1997; Seifert and Roberts, 2007; Ellis et al., 2010). They are structurally complex, generally consisting of a Pro-, Ala-, Ser-, and Thr-rich protein backbone that is extensively modified, principally by hydroxylation of Pro residues (to Hyp) and subsequent glycosylation through O-linkages with type II arabinogalactans (Tan et al., 2003; Shimizu et al., 2005). Many AGPs also have a C-terminal hydrophobic domain that is processed and replaced with a glycosylphosphatidylinositol (GPI) anchor, which acts to tether the molecule to the extracellular face of the plasma membrane (Schultz et al., 1998). AGPs are also defined by their ability to be bound and precipitated by the synthetic dye β-glucosyl Yariv reagent (β-GlcY) and related molecules (Yariv et al., 1967). These dyes have been useful in isolating, localizing, and quantifying AGPs.AGPs are grouped into three subclasses (Schultz et al., 2002): AGPs have an N-terminal signal sequence, an arabinogalactosylated domain, and a hydrophobic C-terminal domain; “chimeric AGPs” contain at least one arabinogalactosylated domain and a domain with an unrelated motif; while “hybrid AGPs” contain arabinogalactosylated as well as different Pro/Hyp-rich glycoprotein motifs.AGPs are implicated in many aspects of plant cell growth and development. Historically, it was not possible to assign roles to individual AGPs, as tests were conducted with unfractionated mixtures of AGPs. More recently, individual AGPs, mainly from Arabidopsis (Arabidopsis thaliana), have been studied using techniques such as mutant analysis and gene knockout/silencing, providing evidence for roles of individual AGPs in cell expansion, root and seed regeneration, the coordination of vascular development, both male and female gametogenesis, the development of cotton fibers, and as contributors to plant stem strength (Shi et al., 2003; van Hengel and Roberts, 2003; Acosta-García and Vielle-Calzada, 2004; Motose et al., 2004; Yang et al., 2007; Levitin et al., 2008; Coimbra et al., 2009; Li et al., 2010; MacMillan et al., 2010).Conditioned media from in vitro embryogenic cultures contain factors that can promote somatic embryogenesis (SE), implying the presence of secreted signaling molecules (de Vries et al., 1988). There is evidence that secreted AGPs, which are components of conditioned media, are involved in SE. For example, SE in carrot (Daucus carota) and spruce (Picea abies) cell cultures was promoted when AGPs from conditioned media were added exogenously (Kreuger and van Holst, 1993; Egertsdotter and von Arnold, 1995). Subsequent studies showed the association of particular AGP epitopes with SE-promoting activity and the involvement of AGPs in SE for several other species (Kreuger et al., 1995; McCabe et al., 1997; Toonen et al., 1997; Chapman et al., 2000; Saare-Surminski et al., 2000; Ben Amar et al., 2007). There is also evidence that SE-promoting AGPs may be cleaved by an endochitinase (Egertsdotter and von Arnold, 1988; Domon et al., 2000; van Hengel et al., 2001, 2002), but neither the identity of the individual AGP(s) involved in promoting SE nor the mechanism of action has been established.In this study, we focused on SE in cotton (Gossypium hirsutum ‘Coker 315’), which is a limiting step in cotton transformation, and the potential role of AGPs in this process. We show that cotton calli undergoing somatic embryogenesis secrete an AGP fraction that promotes SE when incorporated back into the growth medium. The leaf beetle Chrysomela aeneicollis occurs across Western North America, either at high elevation or in small, isolated populations along the coast, and thus has a highly fragmented distribution. DNA sequence data (three loci) were collected from five regions across the species range. Population connectivity was examined using traditional ecological niche modeling, which suggested that gene flow could occur among regions now and in the past. We developed geographically explicit coalescence models of sequence evolution that incorporated a two‐dimensional representation of the hypothesized ranges suggested by the niche‐modeling estimates. We simulated sequence data according to these models and compared them to observed sequences to identify most probable scenarios regarding the migration history of C. aeneicollis. Our results disagreed with initial niche‐modeling estimates by clearly rejecting recent connectivity among regions, and were instead most consistent with a long period of range fragmentation, extending well beyond the last glacial maximum. This application of geographically explicit models of coalescence has highlighted some limitations of the use of climatic variables for predicting the present and past range of a species and has explained aspects of the Pleistocene evolutionary history of a cold‐adapted organism in Western North America. 相似文献
