Anchoring junctions are cell adhesion apparatus present in all epithelia and endothelia. They are found at the cell-cell interface (adherens junction (AJ) and desmosome) and cell-matrix interface (focal contact and hemidesmosome). In this review, we focus our discussion on AJ in particular the dynamic changes and regulation of this junction type in normal epithelia using testis as a model. There are extensive restructuring of AJ (e.g., ectoplasmic specialization, ES, a testis-specific AJ) at the Sertoli-Sertoli cell interface (basal ES) and Sertoli-elongating spermatid interface (apical ES) during the seminiferous epithelial cycle of spermatogenesis to facilitate the migration of developing germ cells across the seminiferous epithelium. Furthermore, recent findings have shown that ES also confers cell orientation and polarity in the seminiferous epithelium, illustrating that some of the functions initially ascribed to tight junctions (TJ), such as conferring cell polarity, are also part of the inherent properties of the AJ (e.g., apical ES) in the testis. The biology and regulation based on recent studies in the testis are of interest to cell biologists in the field, in particular their regulation, which perhaps is applicable to tumorigenesis. 相似文献

Glutamine synthetase from Escherichia coli was inactivated by chemical modification with arginine-specific reagents (Colanduoni, J. A., and Villafranca, J. J. (1985) Biochem. Biophys. Res. Commun. 126, 412-418). E. coli glutamine synthetase was also a substrate for an erythrocyte NAD:arginine ADP-ribosyltransferase. Transfer of one ADP-ribosyl group/subunit of glutamine synthetase caused loss of both biosynthetic and gamma-glutamyltransferase activity. The ADP-ribose moiety was enzymatically removed by an erythrocyte ADP-ribosylarginine hydrolase, resulting in return of function. The site of ADP-ribosylation was arginine 172, determined by isolation of the ADP-ribosylated tryptic peptide. Arginine 172 lies in a central loop that extends into the core formed by the 12 subunits of the native enzyme. The central loop is important in anchoring subunits together to yield the spatial orientation required for catalytic activity. ADP-ribosylation may thus inactivate glutamine synthetase by disrupting the normal subunit alignment. Enzyme-catalyzed ADP-ribosylation may provide a simple, specific technique to probe the role of arginine residues in the structure and function of proteins. 相似文献

Amongst the Gush of southwestern Kenya instances of rape are frequently reported; this was true also in the colonial period and yet it seems that those cases which come to the notice of the authorities are few compared with the numbers which actually occur. While certain features of the traditional culture seem to have encouraged violent sexual assault, because in a patrilocal society miscreants are likely to victimize their relatives, that is women who are sexually taboo, the crime is considered a sacrilegious matter to be dealt with in the family, not by the secular authorities. Given the sacrilegious nature of the act the Gush are convinced that such behavior is involuntary and that the criminal is not a criminal but an afflicted person, motivated by the malevolence of the ancestral spirits or of jealous neighbors and kin. About 447 species of coccidia have been named from the 1687 living, known species of rodents; 207 host species, 92 host genera, and 15 host families are represented; this is about 12% of the known species of rodents. About 4600 species of apicomplexan protozoa have been named. Assuming that the same proportion of the total number of apicomplexan species has been named as of the coccidian species, there must actually be about 38,333 species of apicomplexan protozoa. There are 5.4 times as many protozoan genera as of apicomplexan genera. Assuming that the number of species in each genus is the same for all the protozoa as it is for the Apicomplexa, there may actually be 206,998 species of protozoa. This may be too conservative an estimate. Based on other criteria, an estimate of over 20 million species could be made. 相似文献

Summary When the dnaB37 initiation mutant of Bacillus subtilis is returned to a permissive temperature following a period at 45° C, a synchronous round of DNA replication immediately ensues. Using this system we have been able to analyse the first fragments to be replicated while avoiding the use of thymine starvation or inhibitors of DNA replication. Such treatments are necessary to achieve even modest synchrony in germinating spores. Our results showed that the first fragment to be replicated was a 4kb BamHI-SalI restriction fragment, BS6. In contrast, when the analysis was performed out in the presence of novobiocin, an inhibitor of DNA gyrase, replication from BS6 was inhibited and the first fragment to be replicated was BS5, a 5.6 kb fragment located 1.7 kb to the right of BS 6. Replication from both putative origins was suppressed by rifamycin and was dependent upon dnaB. The results are discussed in relation to previous attempts to identify the first replicating fragment in germinating spores. We also discuss the possibility that B. subtilis contains two origins and suggest that either can act as the primary origin under certain conditions, or alternatively that both origins may act in concert in normal bidirectional replication, each site being required for the leading strand in each direction. 相似文献
