Bax is a pro-apoptotic member of the Bcl-2 family of proteins, which is present in the cytosol of various types of cells in full-length form (p21 Bax). During apoptosis, the N-terminal truncated version of Bax (p18 Bax) is often formed via cleavage of the p21 Bax by the calcium-dependent enzyme, calpain. p18 Bax is a membrane protein found primarily in the mitochondrial fraction of apoptotic cells. Although noticeable amounts of p18 Bax appear relatively late in apoptosis, it may still play a role in the apoptotic cascade. The role of p18 Bax in the apoptotic cascade, particularly, in the release of cytochrome c from mitochondria has not been studied. The goal of this study was to produce reasonable amounts of p18 Bax and study its effect on isolated mitochondria. The expression and purification of membrane proteins such as p18 Bax represents a substantial challenge due to insolubility. We report here that the apoptotic form of Bax, p18 Bax, is highly soluble in the absence of detergents upon fusion with maltose-binding protein (MBP). We describe a scheme for expression and simple metal-affinity based purification of MBP-p18Bax. The MBP-p18Bax triggers the release of cytochrome c from isolated mitochondria in a concentration-dependent, Bcl-2-sensitive manner. The MBP tag of the MBP-p18Bax can be cleaved off with 3C protease to produce pure p18 Bax, although the solubility of p18 Bax becomes very limited. The highly soluble, MBP-fused form of p18 Bax provides a convenient tool to study this apoptotic form of Bax protein.  相似文献

A mathematical model of the movement of olfactory cilia in different conditions was constructed. The realization of the model includes the development of a mechanical mathematical rheological model of the behavior of a continuous deformable medium, the development of the method of solution adapted to this problem, and obtaining a numerical solution, which takes into account different starting data. The mathematical modeling of the dynamic behavior of a deformable medium was performed using a system of equations for the dynamics of the deformable medium and the solution of the corresponding nonstationary system of equations in partial derivatives.  相似文献

The effectiveness of dot immunoanalysis for evaluating the dynamics of the synthesis of O-antigen, cholera toxin, neuraminidase, adhesin CFA1 in the process of the reactor cultivation of V. cholerae used for the production of oral chemical cholera vaccine is shown. The established regularities of the synthesis of the protective antigens of V. cholerae in the process of scaled-up cultivation are discussed.  相似文献

Processes occurring in a plasma flow generated by a magnetoplasma compressor (MPC) during the formation of the compression zone are discussed. The paper presents results of measurements of the spatial distribution of the electric current in the plasma flow, the temporal and spatial (along the flow) distributions of the plasma density, and the profiles of the velocity of individual flow layers along the system axis. The spatial distribution of the electromagnetic force in the flow is analyzed. It is shown that the plasma flow is decelerated when approaching the compression zone and reaccelerated after passing it. In this case, the plasma flow velocity decreases from ν = (2–3) × 107 cm/s at the MPC output to ν < 106 cm/s in the region of maximum compression and then again increases to 107 cm/s at a distance of 15–17 cm from the MPC output. In some MPC operating modes, a displacement of the magnetic field from the compression zone and the formation of toroidal electric current vortices in the plasma flow after passing the compression zone were detected.  相似文献

It is established that the Bacillus sp. C-14 strain is able to produce the biosurfactant of lipopeptide nature which decreases the surface and interfacial tension to 29.5 and 2.0 mH/m, respectively. The biosurfactant synthesis is shown to depend on the substrate nature, N:C ratio in the medium as well as on the presence of sodium citrate as a stimulator of lipogenesis.  相似文献

Aldehydes produced from carbohydrates by oxidation or acid hydrolysis may be visualized by application of aqueous thiosemicarbazide followed by Schmorl's ferricyanide reduction. The thiosemicarbazide reacts with the aldehydes by its hydrazine group, while its thiocarbamyl group remains active. The thiocarbamyl moiety is a strong reducing group that converts ferricyanide to ferrocyanide in Schmorl's reaction. The ferrocyanide is trapped immediately by the ferric salt, which deposits Prussian blue at the site of the aldehydes thereby demonstrating the location of the original substance.  相似文献

Commercial probiotic capsules that contain probiotic bacteria, kefir, koumiss and yogurt contain beneficial microorganisms that affect cholesterol levels and immune response, and are used for treatment of some diseases. We investigated using immunohistochemistry the effects of kefir, koumiss, yogurt and a commercial probiotic formulation on the expression levels of peroxisome proliferator-activated receptor-α (PPARα) and peroxisome proliferator-activated receptor-β/δ (PPAR-β/δ), which are members of the nuclear steroid hormone receptor superfamily in mouse kidney. Mice were assigned to five groups: group 1, commercial probiotic capsules; group 2, kefir; group 3, koumiss; group 4, yogurt; group 5, control. After oral administration for 15 days, body weights were recorded and kidney tissue samples were obtained. Hematoxylin & eosin staining and the streptavidin-biotin peroxidase complex (ABC) method were applied to tissue sections to examine histology and to determine the localization of PPARα and PPAR-β/δ in the kidneys. We found that the weights of the mice in the kefir, koumiss, yogurt and commercial probiotic capsules groups were increased compared to controls. No differences in kidney histology were observed in any of the experimental groups. Kefir, koumiss, yogurt and the commercial probiotic preparation increased PPARα and PPAR-β/δ expressions.  相似文献
