As a result of complex physical, chemical and biological processes, a major fraction of the trace metals introduced into the aquatic environment is found associated with the bottom sediments, distributed among a variety of physico-chemical forms. As these different metal forms will generally exhibit different chemical reactivities, the measurement of the total concentration of a particular metal provides little indication of potential interactions with the abiotic or biotic components present in the environment. In principle, the partitioning of sediment-bound metals could be determined both by thermodynamic calculations (provided equilibrium conditions prevail) and by experimental techniques. The modelling of sediment-bound metals is far less advanced than is that of dissolved species, primarily because the thermodynamic data needed for handling sediment-interstitial water systems are not yet available. The partitioning of a metal among various fractions obtained by experimental techniques (e.g., sequential extraction procedures) is necessarily operationally defined. These methods have, however, provided significant insight into the physico-chemical factors influencing the bioavailability of particulate trace metals; some of these factors are discussed. 相似文献

Daphnia pulicaria from three different populations were observed to express within-population variation in habitat-choice behavior in field assays. Individuals from different habitats (i.e., lake depths) were isolated and cultured as clonal lines under standard conditions. Habitat choices by clonal descendants were then estimated in the field, using replicate experimental columns. There was significant heritable and ontogenetic variation in habitat choice, but the heritable effect was small relative to the phenotypic variation of the original isolates. In a second set of experiments, D. pulicaria that were acclimatized to different habitats showed a strong tendency to choose the habitat to which they had been acclimatized. These data suggest that a given genotype can use a wide range of habitats, given appropriate acclimatization. Although genetic variation is significant, we hypothesize that natural selection on correlated ecological traits is more likely to maintain patterns of genotypic segregation among habitats in Daphnia. 相似文献

We studied minute ventilation, breathing pattern, end-tidal CO2 partial pressure (PACO2), and tracheal occlusion pressure in cats anesthetized with ketamine (40 and 80 mg/kg) before and after CO2 inhalation. Before CO2 administration ventilation was reduced and PACO2 increased relative to unanesthetized cats at both ketamine doses. Breathing pattern was of the "apneustic" type, being characterized by 1) prolonged inspiratory duration and relatively short expiratory time and 2) markedly curvilinear (convex upward) inspiratory volume-time profile. The latter reflected a similar curvilinearity in the tracheal occlusion pressure waveform. During CO2 inhalation, the ventilatory response to CO2 was similar to that in unanesthetized cats in spite of a depressed tracheal occlusion pressure response. This discrepancy was due to the fact that in the presence of a convex upward inspiratory volume-time profile, the shortening of inspiratory duration with increasing CO2 results in a marked increase of mean inspiratory flow, and hence the ventilatory response to CO2 remains high. 相似文献

Fatty acid binding protein 7 (FABP7), abundant in the embryonic brain, binds with the highest affinity to docosahexaenoic acid (DHA) and is expressed in the early stages of embryogenesis. Here, we have examined the consequences of the exposure to different DHA levels and of the in utero depletion of FABP7 on early rat brain development. Neurodevelopment was evaluated through the contents of two proteins, connexin 43 (Cx43) and cyclin‐dependent kinase 5 (CDK5), both involved in neuroblast proliferation, differentiation, and migration. The dams were fed with diets presenting different DHA contents, from deficiency to supplementation. DHA brain embryos contents already differed at embryonic day 11.5 and the differences kept increasing with time. Cx43 and CDK5 contents were positively associated with the brain DHA levels. When FABP7 was depleted in vivo by injections of siRNA in the telencephalon, the enhancement of the contents of both proteins was lost in supplemented animals, but FABP7 depletion did not modify phospholipid compositions regardless of the diets. Thus, FABP7 is a necessary mediator of the effect of DHA on these proteins synthesis, but its role in DHA uptake is not critical, although FABP7 is localized in phospholipid‐rich areas. Our study shows that high contents of DHA associated with FABP7 are necessary to promote early brain development, which prompted us to recommend DHA supplementation early in pregnancy. © 2015 Wiley Periodicals, Inc. T‐cell receptor (TCR) signaling is essential for the function of T cells and negatively regulated by the E3 ubiquitin–protein ligases CBL and CBLB. Here, we combined mouse genetics and affinity purification coupled to quantitative mass spectrometry to monitor the dynamics of the CBL and CBLB signaling complexes that assemble in normal T cells over 600 seconds of TCR stimulation. We identify most previously known CBL and CBLB interacting partners, as well as a majority of proteins that have not yet been implicated in those signaling complexes. We exploit correlations in protein association with CBL and CBLB as a function of time of TCR stimulation for predicting the occurrence of direct physical association between them. By combining co‐recruitment analysis with biochemical analysis, we demonstrated that the CD5 transmembrane receptor constitutes a key scaffold for CBL‐ and CBLB‐mediated ubiquitylation following TCR engagement. Our results offer an integrated view of the CBL and CBLB signaling complexes induced by TCR stimulation and provide a molecular basis for their negative regulatory function in normal T cells. 相似文献

More than half of the cultivation‐induced carbon loss from agricultural soils could be restored through improved management. To incentivise carbon sequestration, the potential of improved practices needs to be verified. To date, there is sparse empirical evidence of carbon sequestration through improved practices in East‐Africa. Here, we show that agroforestry and restrained grazing had a greater stock of soil carbon than their bordering pair‐matched controls, but the difference was less obvious with terracing. The controls were treeless cultivated fields for agroforestry, on slopes not terraced for terracing, and permanent pasture for restrained grazing, representing traditionally managed agricultural practices dominant in the case regions. The gain by the improved management depended on the carbon stocks in the control plots. Agroforestry for 6–20 years led to 11.4 Mg ha?1 and restrained grazing for 6–17 years to 9.6 Mg ha?1 greater median soil carbon stock compared with the traditional management. The empirical estimates are higher than previous process‐model‐based estimates and indicate that Ethiopian agriculture has greater potential to sequester carbon in soil than previously estimated. 相似文献

Background:
Meta-analyses of continuous outcomes typically provide enough information for decision-makers to evaluate the extent to which chance can explain apparent differences between interventions. The interpretation of the magnitude of these differences — from trivial to large — can, however, be challenging. We investigated clinicians’ understanding and perceptions of usefulness of 6 statistical formats for presenting continuous outcomes from meta-analyses (standardized mean difference, minimal important difference units, mean difference in natural units, ratio of means, relative risk and risk difference).Methods:
We invited 610 staff and trainees in internal medicine and family medicine programs in 8 countries to participate. Paper-based, self-administered questionnaires presented summary estimates of hypothetical interventions versus placebo for chronic pain. The estimates showed either a small or a large effect for each of the 6 statistical formats for presenting continuous outcomes. Questions addressed participants’ understanding of the magnitude of treatment effects and their perception of the usefulness of the presentation format. We randomly assigned participants 1 of 4 versions of the questionnaire, each with a different effect size (large or small) and presentation order for the 6 formats (1 to 6, or 6 to 1).Results:
Overall, 531 (87.0%) of the clinicians responded. Respondents best understood risk difference, followed by relative risk and ratio of means. Similarly, they perceived the dichotomous presentation of continuous outcomes (relative risk and risk difference) to be most useful. Presenting results as a standardized mean difference, the longest standing and most widely used approach, was poorly understood and perceived as least useful.Interpretation:
None of the presentation formats were well understood or perceived as extremely useful. Clinicians best understood the dichotomous presentations of continuous outcomes and perceived them to be the most useful. Further initiatives to help clinicians better grasp the magnitude of the treatment effect are needed.Health professionals increasingly rely on summary estimates from systematic reviews and meta-analyses to guide their clinical decisions and to provide information for shared decision-making. Meta-analyses of clinical trials typically provide the information necessary for decision-makers to evaluate the extent to which chance can explain apparent intervention effects (i.e., statistical significance). However, interpreting the magnitude of the treatment effect — from trivial to large — particularly for continuous outcome measures, can be challenging.Such challenges include decision-makers’ unfamiliarity with the instruments used to measure the outcome. For instance, without further information, clinicians may have difficulty grasping the importance of a 5-point difference on the Short-Form Health Survey-36 (SF-36) or a 1-point difference on a visual analogue scale for pain.1 Second, trials often use different instruments to measure the same construct. For instance, investigators may measure physical function among patients with arthritis using 1 of 5 instruments (the Western Ontario and McMaster Universities Arthritis Index using either a visual analogue or Likert scale; the Arthritis Impact Measurement Scale; the SF-36 Physical Function; or the Lequesne index).2,3Authors have several options for pooling results of continuous outcomes. When all trials have used the same instrument to measure outcomes such as physical function or pain, the most straightforward method is to present the mean difference in natural units between the intervention and control groups. When trialists have used different instruments to measure the same construct, authors of systematic reviews typically report differences between intervention and control groups in standard deviation units, an approach known as the standardized mean difference (SMD). This approach involves dividing the mean difference in each trial by the pooled standard deviation for that trial’s outcome.4For meta-analyses of outcomes measured using different instruments, presenting results as an SMD is the longest standing and most widely used approach and is recommended in the Cochrane handbook for systematic reviews of interventions.4 Limitations of this approach include, however, statistical bias toward decreased treatment effects,5,6 the possibility that decision-makers will find the measure difficult to interpret7,8 and the possibility that the same treatment effect will appear different depending on whether the study population had similar results in the measure of interest (i.e., if homogeneous, a small standard deviation) or varied greatly in the measure of interest (i.e., if heterogeneous, a large standard deviation).9,10Several research groups have proposed alternative statistical formats for presenting continuous outcomes from meta-analyses that they postulate clinicians will more easily interpret.6–8,11–16 The Grading of Recommendations Assessment, Development and Evaluation (GRADE) Working Group recently provided an overview of methods for presenting pooled continuous data.9,10 These alternatives (Appendix 1, available at www.cmaj.ca/lookup/suppl/doi:10.1503/cmaj.150430/-/DC1), although intuitively compelling, have seen limited use.We conducted a survey to determine clinicians’ understanding of the magnitude of treatment effect for 6 approaches to the presentation of continuous outcomes from meta-analyses, as well as their perceptions of the usefulness of each approach for clinical decision-making.
