Continuum mechanical tools are used to describe the deformation, energy density, and material symmetry of a lipid bilayer with spontaneous curvature. In contrast to conventional approaches in which lipid bilayers are modeled by material surfaces, here we rely on a three-dimensional approach in which a lipid bilayer is modeling by a shell-like body with finite thickness. In this setting, the interface between the leaflets of a lipid bilayer is assumed to coincide with the mid-surface of the corresponding shell-like body. The three-dimensional deformation gradient is found to involve the curvature tensors of the mid-surface in the spontaneous and the deformed states, the deformation gradient of the mid-surface, and the transverse deformation. Attention is also given to the coherency of the leaflets and to the area compatibility of the closed lipid bilayers (i.e., vesicles). A hyperelastic constitutive theory for lipid bilayers in the liquid phase is developed. In combination, the requirements of frame indifference and material symmetry yield a representation for the energy density of a lipid bilayer. This representation shows that three scalar invariants suffice to describe the constitutive response of a lipid bilayer exhibiting in-plane fluidity and transverse isotropy. In addition to exploring the geometrical and physical properties of these invariants, fundamental constitutively associated kinematical quantities are emphasized. On this basis, the effect on the energy density of assuming that the lipid bilayer is incompressible is considered. Lastly, a dimension reduction argument is used to extract an areal energy density per unit area from the three-dimensional energy density. This step explains the origin of spontaneous curvature in the areal energy density. Importantly, along with a standard contribution associated with the natural curvature of the lipid bilayer, our analysis indicates that constitutive asymmetry between the leaflets of the lipid bilayer gives rise to a secondary contribution to the spontaneous curvature.  相似文献

Many animals exhibit seasonal changes in behavior and its underlying neural substrates. In seasonally breeding songbirds, the brain nuclei that control song learning and production undergo substantial structural changes at the onset of each breeding season, in association with changes in song behavior. These changes are largely mediated by photoperiod‐dependent changes in circulating concentrations of gonadal steroid hormones. Little is known, however, about whether changes in the electrophysiological activity of neurons accompany the dramatic morphological changes in the song nuclei. Here we induced seasonal‐like changes in the song systems of adult white‐crowned sparrows and used extracellular recording in acute brain slices from those individuals to study physiological properties of neurons in the robust nucleus of the arcopallium (RA), a pre‐motor nucleus necessary for song production. We report that: RA neurons from birds in breeding condition show a more than twofold increase in spontaneous firing rate compared to those from nonbreeding condition; this change appears to require both androgenic and estrogenic actions; and this change is intrinsic to the RA neurons. Soluble proteins that reside in the lumen of the endoplasmic reticulum are known to have at their carboxyterminus the tetrapeptides KDEL or HDEL. In yeast and mammalian cells, these tetrapeptides function as endoplasmic reticulum (ER)-retention signals. To determine the effect of an artificially-introduced KDEL sequence at the exact carboxyterminus of a plant secretory protein, we modified the gene of the vacuolar protein phytohemagglutinin-L (PHA) so that the amino-acid sequence would end in LNKDEL rather than LNKIL, and expressed the modified gene in transgenic tobacco with a seed-specific promoter. Analysis of the glycans of PHA showed that most of the control PHA had one endoglycosidase H-sensitive and one endoglycosidase H-resistant glycan, indicating that it had been processed in the Golgi complex. On the other hand, a substantial portion of the PHA-KDEL (about 75% at mid-maturation and 50% in mature seeds) had two endoglycosidase H-sensitive glycans. Phytohemagglutinin with two endoglycosidase H-sensitive glycans is normally found in the ER. Using immunocytochemistry we found that a substantial portion of the PHA-KDEL was present in the ER or accumulated in the nuclear envelope while the remainder was found in the protein storage vacuoles (protein bodies). We interpret these data to indicate that carboxyterminal KDEL functions as an ER retention-retardation signal and causes protein to accumulate in the nuclear envelope as well as in the ER.
