Tissue engineering techniques particularly using electrospun scaffolds have been intensively used in recent years for the development of small diameter vascular grafts. However, the development of a completely successful scaffold that fulfills multiple requirements to guarantee complete vascular regeneration remains challenging. In this study, a hydrophilic and compliant polyurethane namely Tecophilic (TP) blended with gelatin (gel) at a weight ratio of 70:30 (TP(70)/gel(30)) was electrospun to fabricate a tubular composite scaffold with biomechanical properties closely simulating those of native blood vessels. Hydrophilic properties of the composite scaffold induced non‐thrombogenicity while the incorporation of gelatin molecules within the scaffold greatly improved the capacity of the scaffold to serve as an adhesive substrate for vascular smooth muscle cells (SMCs), in comparison to pure TP. Preservation of the contractile phenotype of SMCs seeded on electrospun TP(70)/gel(30) was yet another promising feature of this scaffold. Phospholipid transfer protein (PLTP) is a key protein involved in biogenesis and remodeling of plasma HDL. Several neuroprotective properties have been ascribed to HDL. We reported earlier that liver X receptor (LXR) activation promotes cellular cholesterol efflux and formation of HDL-like particles in an established in vitro model of the blood-brain barrier (BBB) consisting of primary porcine brain capillary endothelial cells (pBCEC). Here, we report PLTP synthesis, regulation, and its key role in HDL metabolism at the BBB. We demonstrate that PLTP is highly expressed and secreted by pBCEC. In a polarized in vitro model mimicking the BBB, pBCEC secreted phospholipid-transfer active PLTP preferentially to the basolateral (“brain parenchymal”) compartment. PLTP expression levels and phospholipid transfer activity were enhanced (up to 2.5-fold) by LXR activation using 24(S)-hydroxycholesterol (a cerebral cholesterol metabolite) or TO901317 (a synthetic LXR agonist). TO901317 administration elevated PLTP activity in BCEC from C57/BL6 mice. Preincubation of HDL3 with human plasma-derived active PLTP resulted in the formation of smaller and larger HDL particles and enhanced the capacity of the generated HDL particles to remove cholesterol from pBCEC by up to 3-fold. Pre-β-HDL, detected by two-dimensional crossed immunoelectrophoresis, was generated from HDL3 in pBCEC-derived supernatants, and their generation was markedly enhanced (1.9-fold) upon LXR activation. Furthermore, RNA interference-mediated PLTP silencing (up to 75%) reduced both apoA-I-dependent (67%) and HDL3-dependent (30%) cholesterol efflux from pBCEC. Based on these findings, we propose that PLTP is actively involved in lipid transfer, cholesterol efflux, HDL genesis, and remodeling at the BBB.  相似文献

Background

Leiomyosarcoma of the inferior vena cava is a rare tumor that presents in an insidious manner with non-specific symptoms. Given its rarity, there are no consensus guidelines to its management. The aim of this study was to report the clinical experience in the management of patients presenting to our institution during a 12 year period.

Patients and Methods

Four patients with leiomyosarcomas of the inferior vena cava were managed at our institution during the period reviewed. Patient details were identified through a search of the pathology department computerized database, and case notes were retrospectively reviewed to obtain details of presentation and management.

Results

There were 3 females and 1 male with a mean age of 59 years. All tumors were identified within 2 months of first symptoms. Three of the 4 had localized tumors whilst 1 patient had lung metastases at presentation. The three patients with resectable tumors underwent radical surgical excision of the tumor, and two patients had postoperative radiotherapy. One patient died of recurrence at 7 months, and another at 30 months. The third patient is currently well and disease free at 16 months. The fourth patient with metastatic disease was treated with chemotherapy alone and survived 36 months.

Conclusion

Leiomyosarcoma of the inferior vena cava is an uncommon tumor that presents with non-specific symptoms. At the time of presentation, tumors are usually large and resection is challenging but probably offers the best opportunity for long-term survival.  相似文献

We study the optical bistability and multistability in a defect structure doped with polaritonic materials and three-level nanoparticles. It is realized that the threshold of optical bistability can be manipulated by some controllable parameters such as Rabi frequency, line width of upper level, and thickness of defect structure. Due to dense doping of three-level nanoparticles, the dipole-dipole interactions (DDI) between nanoparticles become important. Therefore, the DDI has been considered as an interesting mechanism for transition from optical bistability to multistability. The line width effect of upper level and thickness of defect structure on threshold of optical multistability has also been investigated. We hope that our proposed model may be useful for developing the future all-optical devices in nanoscales.

Divergent evolution between Western Rock Nuthatch Sitta neumayer and Eastern Rock Nuthatch Sitta tephronota is widely recognized as the original case study of character displacement. However, in their contact zone in the Zagros Mountains, Iran, the morphological differences important for niche segregation between the two species remain unknown. We investigated microhabitat use and morphological adaptations of the two species, predicting that morphological adaptations to different habitats in these two nuthatches have led to spatial segregation. Seventy‐seven birds were captured and measured in the contact zone and allopatric zone in Iran. Twenty‐two primary variables related to flight apparatus, functional foot apparatus and feeding apparatus were measured and 11 ratios of primary variables were calculated as secondary variables due to their importance in habitat use. We also measured environmental variables related to geological features, mineral substrates and vegetation cover at a random sample of 100 of the locations where a nuthatch was observed. Results of morphometric and habitat analyses indicated that, in addition to trophic niche segregation, the two nuthatch species also differ in their microhabitat use and show differences in morphological features accordingly. In many case studies of character displacement, much more interest has been focused on the morphological differences in feeding apparatus than on those relating to habitat use. We suggest that future studies of character displacement should pay more attention to spatial niche segregation between sympatric species rather than trophic niche segregation.  相似文献

Among various factors influencing mood disorders, the impact of micronutrient deficiencies has attracted a great attention. Zinc deficiency is considered to play a crucial role in the onset and progression of mood disorders in different stages of life. The main objective of this study was to assess the correlation between serum zinc levels and mood disorders in high school female students. This cross-sectional study was conducted on a random sample of 100 representative high school female students. The participants completed 24-h food recall questionnaires to assess the daily zinc intakes. Serum zinc status was assessed using flame atomic absorption spectrometry, and zinc deficiency was defined accordingly. Mood disorders were estimated by calculating the sum of two test scores including Beck’s depression inventory (BDI) and hospital anxiety depression scale (HADS) tests. General linear model (GLM) and Pearson’s regression test were applied to show the correlation of serum zinc levels and mood disorder scores and the correlation between zinc serum levels and BDI scores, respectively. Dietary zinc intake was higher in subjects with normal zinc concentrations than that of zinc-deficient group (p = 0.001). Serum zinc levels were inversely correlated with BDI and HADS scores (p < 0.05). Each 10 μg/dL increment in serum zinc levels led to 0.3 and 0.01 decrease in depression and anxiety scores, respectively (p < 0.05). Serum zinc levels were inversely correlated with mood disorders including depression and anxiety in adolescent female students. Increasing serum levels of zinc in female students could improve their mood disorders.  相似文献

Wild progenitors of common wheat are a potential source of tolerance to biotic and abiotic stresses. We conducted a glasshouse pot experiment to study genotypic differences in response to drought stress in a collection of 180 accessions of Aegilops and Triticum along with one tolerant and one sensitive control variety. Several physiological traits and chlorophyll fluorescence parameters were evaluated. Our findings indicated that drought significantly reduced shoot fresh (59.45%) and dry (50.83%) weights, stomatal conductance (41.52%) and maximum photosynthetic capacity (41.06%), but increased initial fluorescence (28.10%). Drought stress also decreased the chlorophyll content, relative water content and maximum quantum efficiency by 14.90, 12.13 and 11.42%, respectively. Principal component analysis of the 182 individuals identified three components that explained 57.61 and 61.68% of the total variation in physiological and photosynthetic traits under control and stress conditions, respectively. When grouped into the 12 species tested, the three top components explained 78.22% of the total variation under drought. The means comparison, stress tolerance index and biplot analysis identified five accessions with superior tolerance to drought. Remarkably, four species of wild relatives—Ae. cylindrica (DC genome), Ae. crassa (DM genome), Ae. caudata (C genome) and T. urartu (Au genome)—responded well to drought stress with a lower percentage decline for most traits and high values for the first two components. The potential of these species offers further opportunities for analysis at the molecular and cellular levels to confront with drought stress through a physiological mechanism.  相似文献

An intercellular spreading strategy using herpes simplex virus type 1 (HSV-1) VP22 protein is employed to enhance DNA vaccine potency of Leishmania major amastin antigen in BALB/c mice model. We evaluated the immunogenicity and protective efficacy of plasmid DNA vaccines encoding amastin-enhanced green fluorescent protein (EGFP) and VP22-amastin-EGFP. Optimal cell-mediated immune responses were observed in BALB/c mice immunized with VP22-amastin-EGFP as assessed by cytokine gene expression analysis using real time RT-PCR. Vaccination with the VP22-amastin-EGFP fusion construct elicited significantly higher IFN-gamma response upon antigen stimulation of splenocytes from immunized mice compared to amastin as a sole antigen. Mice immunized by VP22-amastin-EGFP showed partial protection following infectious challenge with L. major, as measured by parasite load in spleens. These results suggest that the development of DNA vaccines encoding VP22 fused to a target Leishmania antigen would be a promising strategy to improve immunogenicity and DNA vaccine potency.  相似文献

Germ-line mutations in BRCA1 breast cancer susceptibility gene account for a large proportion of hereditary breast cancer families and show considerable ethnic and geographical variations. The contribution of BRCA1 mutations to hereditary breast cancer has not yet been thoroughly investigated in Middle Eastern and North African populations. In this study, 16 Tunisian high-risk breast cancer families were screened for germline mutations in the entire BRCA1 coding region and exon?Cintron boundaries using direct sequencing. Six families were found to carry BRCA1 mutations with a prevalence of 37.5%. Four different deleterious mutations were detected. Three truncating mutations were previously described: c.798_799delTT (916 delTT), c.3331_3334delCAAG (3450 delCAAG), c.5266dupC (5382 insC) and one splice site mutation which seems to be specific to the Tunisian population: c.212?+?2insG (IVS5?+?2insG). We also identified 15 variants of unknown clinical significance. The c.798_799delTT mutation occurred at an 18% frequency and was shared by three apparently unrelated families. Analyzing five microsatellite markers in and flanking the BRCA1 locus showed a common haplotype associated with this mutation. This suggests that the c.798_799delTT mutation is a Tunisian founder mutation. Our findings indicate that the Tunisian population has a spectrum of prevalent BRCA1 mutations, some of which appear as recurrent and founding mutations.  相似文献
