Exposure to chemicals absorbed by the skin can threaten human health. In order to standardise the predictive testing of percutaneous absorption for regulatory purposes, the OECD adopted guideline 428, which describes methods for assessing absorption by using human and animal skin. In this study, a protocol based on the OECD principles was developed and prevalidated by using reconstructed human epidermis (RHE). The permeation of the OECD standard compounds, caffeine and testosterone, through commercially available RHE models was compared to that of human epidermis and animal skin. In comparison to human epidermis, the permeation of the chemicals was overestimated when using RHE. The following ranking of the permeation coefficients for testosterone was obtained: SkinEthic > EpiDerm, EPISKIN > human epidermis, bovine udder skin, pig skin. The ranking for caffeine was: SkinEthic, EPISKIN > bovine udder skin, EpiDerm, pig skin, human epidermis. The inter-laboratory and intra-laboratory reproducibility was good. Long and variable lag times, which are a matter of concern when using human and pig skin, did not occur with RHE. Due to the successful transfer of the protocol, it is now in the validation process.  相似文献

Studies of elevational gradients in forests are particularly interesting for the considerable differences that can be observed over short distances, such as in vegetation and temperature. Different taxonomic groups display varying types of distribution patterns along elevational gradients, with unimodal distribution being recognised as the most common pattern. The distribution of species can be affected by a range of factors that include, biotic, spatial, climatic, historic and energetic. Small mammals represent an ideal model for studies about distribution and habitat use as they can be highly abundant, tend to have different diets and use space differently. The aims of this study are to build a comprehensive understanding of the community of small mammals of the Biological Reserve of Serra do Japi and to explore its distribution pattern along elevational gradients. We investigated the influence of biomass of arthropods, fruits and seeds and percentage of ground cover, canopy cover and vertical vegetation at richness and abundance of small mammals at three different elevations. To accomplish this, we used seventy‐two pitfall traps of 63 L to capture small mammals and distributed them equally across three elevations defined as low (880–899 m), intermediate (1046–1089 m) and high (1170–1189 m). Each elevation had three lines or replicas of traps. Throughout the study, we captured one hundred and fourteen individuals belonging to eleven species of small mammals. The presence of rare and endemic species demonstrates the importance of conservation and maintenance of the Biological Reserve of Serra do Japi. In regard to the distribution of species, despite the short gradient range, we found a unimodal pattern and a positive correlation between ground cover (fallen twigs and branches up to 1 m high) and richness and abundance of small mammals. Background

Pulmonary drug delivery is characterized by short onset times of the effects and an increased therapeutic ratio compared to oral drug delivery. This delivery route can be used for local as well as for systemic absorption applying drugs as single substance or as a fixed dose combination. Drugs can be delivered as nebulized aerosols or as dry powders. A screening system able to mimic delivery by the different devices might help to assess the drug effect in the different formulations and to identify potential interference between drugs in fixed dose combinations. The present study evaluates manual devices used in animal studies for their suitability for cellular studies.

Methods

Calu-3 cells were cultured submersed and in air-liquid interface culture and characterized regarding mucus production and transepithelial electrical resistance. The influence of pore size and material of the transwell membranes and of the duration of air-liquid interface culture was assessed. Compounds were applied in solution and as aerosols generated by MicroSprayer IA-1C Aerosolizer or by DP-4 Dry Powder Insufflator using fluorescein and rhodamine 123 as model compounds. Budesonide and formoterol, singly and in combination, served as examples for drugs relevant in pulmonary delivery.

Results and Conclusions

Membrane material and duration of air-liquid interface culture had no marked effect on mucus production and tightness of the cell monolayer. Co-application of budesonide and formoterol, applied in solution or as aerosol, increased permeation of formoterol across cells in air-liquid interface culture. Problems with the DP-4 Dry Powder Insufflator included compound-specific delivery rates and influence on the tightness of the cell monolayer. These problems were not encountered with the MicroSprayer IA-1C Aerosolizer. The combination of Calu-3 cells and manual aerosol generation devices appears suitable to identify interactions of drugs in fixed drug combination products on permeation.  相似文献

Many primates have to cope with a temporal scarcity in food availability that shapes their foraging strategies. Here we investigated the changes in diet, activity, and ranging behavior of a group of black-fronted titi monkeys (Callicebus nigrifrons) according to the availability of the main high-nutritional-density item in their diet and the foraging strategy adopted when this food is scarce. We monitored one habituated group using instantaneous scan sampling over 1 year (533 h of observation, 61 days) in a seasonal tropical forest fragment (245 ha). We simultaneously collected data on food availability with fruit traps. The titi monkeys consumed fleshy fruits, the main high-nutritional-density item of their diet, in accordance with its availability, and the availability of this item modulated the ingestion of vegetative plant parts, a relatively low-nutritional-density food. During high fleshy fruit availability, the titi monkeys consumed more fleshy fruits, flowers, and invertebrates. They also traveled more, but concentrated their activity in a central area of their home range. Conversely, during fleshy fruit scarcity, they increased the breadth of their diet, switching to one richer in seeds and vegetative plant parts, and with greater plant diversity. At the same time, they reduced most energy-demanding activities, traveling less and over shorter distances, but using their home range more broadly. Corroborating the optimal foraging theory, titi monkeys altered foraging strategies according to temporal food fluctuations and responded to low fleshy fruit availability by changing their diet, activity, and ranging behavior. The adoption of a low-cost/low-yield strategy allowed us to classify them as energy minimizers.  相似文献

Most aspects of the ecology and behavior of Callicebus nigrifrons are still unknown. The information available about this species is mainly based on a few studies that also focused on other Callicebus. We examined the feeding behavior and activity pattern of a free-ranging pair of C. nigrifrons between March and November 2007 in an area of semideciduous tropical forest of southeastern Brazil. The study site is located at the southern limit of the Tropical Zone and is characterized by pronounced seasonality. As observed for other Callicebus monkeys, fruits were the most consumed food resource, accounting for 53% of the diet, which was complemented mainly by leaves (16%) but also by invertebrates and flowers (10% of each). A great variety of plant families (28) and species (62) were included in the diet. The titis spent 35% of their time feeding, distributing the remaining time between resting (30%) and traveling (24%). Data presented here indicate that C. nigrifrons prefer high-quality food items (fruit pulp), adding low-quality food items (such as leaves) as the availability of the higher-quality foods decreases. The amount of time spent traveling and resting did not change between seasons, but the time invested in feeding increased during the lean period. The activity pattern was not related to fruit availability, but in months with lower temperatures, monkeys spent more time feeding. We suggest that the feeding ecology and activity pattern of C. nigrifrons reflect adaptations related to annual fluctuations in food availability and temperature, respectively.  相似文献
