The sequence of a cDNA from the giant gene of Drosophila shows that its product has a basic domain followed by a leucine zipper motif. Both features contain characteristic conserved elements of the b-ZIP family of DNA-binding proteins. Expression of the gene in bacteria or by in vitro translation yields a protein that migrates considerably faster than the protein extracted from Drosophila embryos. Treatment with phosphatase shows that this difference is due to multiple phosphorylation of the giant protein in the embryo. Ectopic expression of the protein in precellular blastoderm embryos produces abnormal phenotypes with a pattern of segment loss closely resembling that of Krüppel mutant embryos. Immunological staining shows that giant, ectopically expressed from the hsp70 promoter, represses the expression of both the Krüppel and knirps segmentation gap genes. The analysis of the interactions between Krüppel, knirps and giant reveals a network of negative regulation. We show that the apparent positive regulation of knirps by Krüppel is in fact mediated by a negative effect of Krüppel on giant and a negative effect of giant on knirps. giant protein made in bacteria or in embryos binds in vitro to the Krüppel regulatory elements CD1 and CD2 and recognizes a sequence resembling the binding sites of other b-ZIP proteins.  相似文献

The administration to albino rats of tannic acid as a retention enema (in doses of 0.2 g./kg. body weight and over) prolonged the duration of anesthesia induced by thiopental given immediately before, or 72 hours after, the tannic acid. This dose of tannic acid corresponds, on the basis of body weight, to a radiodiagnostic enema of 2 1. of 0.25% tannic acid in barium sulfate suspension given to a child weighing 25 kg. By excluding certain hypothermic effects of tannic acid, it was concluded that thiopental potentiation was probably due to impairment by the tannic acid of the liver''s ability to detoxify the barbiturate. The results suggest that a drug which is detoxified in the liver should be administered three to five days after a tannic acid-barium sulfate radiodiagnostic enema only with considerable caution.  相似文献

We compared the proportions of mammalian-type and reptilian-type nephrons in the kidneys of two species of passerine birds. The desert house sparrow (Passer domesticus) is relatively well adapted for water conservation, whereas the white-crowned sparrow (Zonotrichia leucophrys) is more mesic adapted. The two species do not differ in body mass, but the kidneys of P. domesticus are significantly smaller than those of Z. leucophrys. Associated with its smaller size, the house sparrow kidney has significantly fewer glomeruli (35,700 per kidney) than does the white-crowned sparrow kidney (53,000 per kidney). The medullary cones, which contain the loops of Henle of the mammalian-type nephrons, are significantly longer in house sparrows than in white-crowned sparrows (2.2 vs. 1.9 mm). The number of medullary cones, the number of nephrons per medullary cone, and, hence, the number of mammalian-type nephrons do not differ between the two species. The smaller number of nephrons in the kidney of the house sparrow therefore represents a smaller number of reptilian-type nephrons. Desert house sparrows have 18% mammalian-type nephrons, whereas white-crowned sparrows have 10% mammaliantype nephrons. The relative reduction of reptilian-type nephrons in P. domesticus may reduce the flow of dilute urine through the collecting ducts, thereby permitting a greater concentration gradient to be established along the length of the medullary cones.  相似文献
