The autonomic nervous system regulates hormone secretion from the endocrine pancreas, the islets of Langerhans, thus impacting glucose metabolism. The parasympathetic and sympathetic nerves innervate the pancreatic islet, but the precise innervation patterns are unknown, particularly in human. Here we demonstrate that the innervation of human islets is different from that of mouse islets and does not conform to existing models of autonomic control of islet function. By visualizing axons in three dimensions and quantifying axonal densities and contacts within pancreatic islets, we found that, unlike mouse endocrine cells, human endocrine cells are sparsely contacted by autonomic axons. Few parasympathetic cholinergic axons penetrate the human islet, and the invading sympathetic fibers preferentially innervate smooth muscle cells of blood vessels located within the islet. Thus, rather than modulating endocrine cell function directly, sympathetic nerves may regulate hormone secretion in human islets by controlling local blood flow or by acting on islet regions located downstream.  相似文献

After the decrease of the relative importance of Triatoma infestans, a number of studies reported the occurrence of sylvatic triatomines dispersing actively to domestic environments in the dry western Chaco Region of Argentina. Anthropic modification of the landscape is mentioned as one of the main causes of the increase in domicile invasion. The aim of this study was to describe the occurrence and frequency of sylvatic triatomines invading rural houses, and to evaluate the effect of habitat fragmentation and other ecological factors on the invasion of rural houses in central Argentina. We hypothesized that the decrease in food sources and the loss of wild ecotopes, as a consequence of habitat fragmentation, increase the chances of invasion by triatomines. The entomological data was collected by community-based vector surveillance during fieldwork carried out between 2017–2020, over 131 houses located in fourteen rural communities in the northwest of Córdoba Province (central Argentina). We used generalized linear models to evaluate the effect of (i) the environmental anthropic disturbance in the study area, (ii) the composition and configuration of the landscape surrounding the house, (iii) the spatial arrangement of houses, (iv) and the availability of artificial refuges and domestic animals in the peridomicile, on house invasion by triatomines. We report the occurrence of seven species of triatomines invading rural houses in the study area -T. infestans, T. guasayana, T. garciabesi, T. platensis, T. delpontei, T. breyeri and P. guentheri-. Study data suggest that invasion by triatomines occurs with higher frequency in disturbed landscapes, with houses spatially isolated and in proximity to subdivided fragments of forest. The availability of domestic refuges in the peridomestic structures as well as the presence of a higher number of domestic animals increase the chances of invasion by triatomines.  相似文献

Modular variation of multivariate traits results from modular distribution of effects of genetic and epigenetic interactions among those traits. However, statistical methods rarely detect truly modular patterns, possibly because the processes that generate intramodular associations may overlap spatially. Methodologically, this overlap may cause multiple patterns of modularity to be equally consistent with observed covariances. To deal with this indeterminacy, the present study outlines a framework for testing a priori hypotheses of modularity in which putative modules are mathematically represented as multidimensional subspaces embedded in the data. Model expectations are computed by subdividing the data into arrays of variables, and intermodular interactions are represented by overlapping arrays. Covariance structures are thus modeled as the outcome of complex and nonorthogonal intermodular interactions. This approach is demonstrated by analyzing mandibular modularity in nine rodent species. A total of 620 models are fit to each species, and the most strongly supported are heuristically modified to improve their fit. Five modules common to all species are identified, which approximately map to the developmental modules of the mandible. Within species, these modules are embedded within larger "super-modules," suggesting that these conserved modules act as building blocks from which covariation patterns are built.  相似文献

The influence of arthropod availability on the formation of male little bustard breeding territories in central Spain was studied in two consecutive years. Arthropods (68,217 individuals) were sampled using pitfall trapping and male territories estimated by means of kernel functions. Hymenopterans (ants), beetles, mites and spiders were the main taxa found in the samples, while the highest contributions in terms of biomass came from hymenopterans, beetles and orthopterans. The only group that exhibited significant biomass differences between territory and non-territory locations was that of the beetles; this pattern was consistent between years. Carabidae was the main beetle family in terms of biomass, followed by other large beetles such as Tenebrionidae, Cleridae, Scarabeidae and Elateridae. The biomass of Carabidae that were larger than 15 mm was significantly higher within territories than outside them. The biomass of Cleridae was greater in fallows, while Elateridae were more abundant in arable fields. The total biomass of beetles that were larger than 15 mm was higher in arable fields than in fallows. Family richness differed between years, while diversity showed differences between agricultural fields (it was highest in fallows), although these variables did not differ between territory and non-territory locations. Results indicate that large beetles (particularly carabids) seem to be a valuable food resource that is defended by little bustard males during the highly energy-demanding breeding season, which is discussed here in relation to habitat selection, the mating system and its implications for the conservation of the species.  相似文献

We have sequenced most of the coding region of the gene Dopa decarboxylase (Ddc) in 24 fruitfly species. The Ddc gene is quite informative about Drosophila phylogeny. Several outstanding issues in Drosophila phylogeny are resolved by analysis of the Ddc sequences alone or in combination with three other genes, Sod, Adh, and Gpdh. The three species groups, melanogaster, obscura, and willistoni, are each monophyletic and all three combined form a monophyletic group, which corresponds to the subgenus Sophophora. The Sophophora subgenus is the sister group to all other Drosophila subgenera (including some named genera, previously considered outside the Drosophila genus, namely, Scaptomyza and Zaprionus, which are therefore downgraded to the category of subgenus). The Hawaiian Drosophila and Scaptomyza are a monophyletic group, which is the sister clade to the virilis and repleta groups of the subgenus Drosophila. The subgenus Drosophila appears to be paraphyletic, although this is not definitely resolved. The two genera Scaptodrosophila and Chymomyza are older than the genus Drosophila. The data favor the hypothesis that Chymomyza is older than Scaptodrosophila, although this issue is not definitely resolved. Molecular evolution is erratic. The rates of nucleotide substitution in 3rd codon position relative to positions 1 + 2 vary from one species lineage to another and from gene to gene. The rapid generation of H(2)O(2) by Cd(2+)-treated plant cells was investigated in cultured tobacco (Nicotiana tabacum L.) BY-2 cells. The starting point for the generation of H(2)O(2) has been located at the cell plasma membrane using cytochemical methods. Treatment of the cells with diphenyleneiodonium (DPI) and imidazol, both inhibitors of the neutrophil NADPH oxidase, prevented the generation of H(2)O(2) induced by Cd(2+). These data suggest the involvement of an NADPH oxidase-like enzyme leading to H(2)O(2) production through O(2)(*-) dismutation by superoxide dismutase enzymes. To investigate the implication of Ca(2+) channels in a Cd(2+)-induced oxidative burst, different inhibitors of Ca(2+) channels were used. Only La(3+) totally inhibited the generation of H(2)O(2) induced by Cd(2+). However, verapamil and nifedipine, inhibitors of Ca(2+) channels, were not effective. Calmodulin or a Ca(2+)-dependent protein kinase is also implicated in the signal transduction sequence, based on the results obtained with two types of calmodulin antagonists, fluphenazine and N-(-6-amino-hexyl)-5-chloro-1-naphthalenesulphonamide (W-7) and staurosporine, an inhibitor of protein kinases. However, neomycin, an inhibitor of the phosphoinositide cycle, did not inhibit the generation of H(2)O(2) induced by Cd(2+), suggesting mainly an induction of the oxidative burst mediated by calmodulin and/or calmodulin-dependent proteins.  相似文献

The use of unconventional yeast species in human-driven fermentations has attracted a lot of attention in the last few years. This tool allows the alcoholic beverage industries to solve problems related to climate change or the consumer demand for newer high-quality products. In this sense, one of the most attractive species is Saccharomyces kudriavzevii, which shows interesting fermentative traits such as the increased and diverse aroma compound production in wines. Specifically, it has been observed that different isolates of this species can produce higher amounts of higher alcohols such as phenylethanol compared with Saccharomyces cerevisiae. In this work, we have shed light on this feature relating it to the S. kudriavzevii aromatic amino acid anabolic pathway in which the enzyme Aro4p plays an essential role. Unexpectedly, we observed that the presence of the S. kudriavzevii ARO4 variant reduces phenylethanol production compared with the S. cerevisiae ARO4 allele. Our experiments suggest that this can be explained by increased feedback inhibition, which might be a consequence of the changes detected in the Aro4p amino end such as L26Q24 that have been under positive selection in the S. kudriavzevii specie.  相似文献

Heteroduplex analysis (HA) has proven to be a robust tool for mutation detection. HA by capillary array electrophoresis (HA-CAE) was developed to increase throughput and allow the scanning of large multiexon genes in multicapillary DNA sequencers. HA-CAE is a straightforward and high-throughput technique to detect both known and novel DNA variants with a high level of sensitivity and specificity. It consists of only three steps: multiplex-PCR using fluorescently labeled primers, heteroduplex formation and electrophoresis in a multicapillary DNA sequencer. It allows, e.g., the complete coding and flanking intronic sequences of BRCA1 and BRCA2 genes from two patients (approximately 25 kb each) to be scanned in a single run of a 16-capillary sequencer, and has enabled us to detect 150 different mutations to date (both single nucleotide substitutions, or SNSs, and small insertions/deletions). Here, we describe the protocol developed in our laboratory to scan BRCA1, BRCA2, MLH1, MSH2 and MSH6 genes using an ABI3130XL sequencer. This protocol could be adapted to other instruments or to the study of other large multiexon genes and can be completed in 7-8 h.  相似文献
