Achievements obtained in infertility treatments over the past two decades have sparked interest in optimizing progesterone administration. Although progesterone is absorbed orally when ingested in micronized form, bioavailability is poor because of extensive liver metabolism. This explains why full predecidual transformation of the endometrium cannot be achieved with oral progesterone and is therefore ineffective for luteal support in in vitro fertilization (IVF). Progesterone administered non-orally can duplicate the endometrial changes normally seen in the menstrual cycle in women whose ovaries are inactive. Similar results have been reported with intramuscular (i.m.) injections and vaginal administration, although tissue levels are higher in the latter case. The recent development of a controlled and sustained release vaginal progesterone gel, Crinone(R) 8%, has made the vaginal route clinically practical by limiting the number of necessary applications to 1 per day. This regimen has been found at least as effective as intramuscular (i.m.) injections in women whose ovaries are inactive (donor egg IVF) and for luteal support in regular IVF. Hence, painful daily i.m. injections of progesterone in oil become unnecessary. The possibility of reducing the number of daily applications of vaginal progesterone to 1 per day, made possible by the sustained release gel Crinone, has opened new possibilities for long-term treatments, as in hormone replacement therapy (HRT). The low incidence of systemic side effects with use of the vaginal progesterone gel used for HRT in amenorrheic women, contrasts with findings related to use of synthetic progestins. Preliminary data suggest that vaginal progesterone can be instrumental in enhancing the notoriously poor long-term compliance of HRT.  相似文献

Humans excel in cooperative exchanges between unrelated individuals. Although this trait is fundamental to the success of our species, its evolution and mechanisms are poorly understood. Other social mammals also build long-term cooperative relationships between non-kin, and recent evidence shows that oxytocin, a hormone involved in parent–offspring bonding, is likely to facilitate non-kin as well as kin bonds. In a population of wild chimpanzees, we measured urinary oxytocin levels following a rare cooperative event—food sharing. Subjects showed higher urinary oxytocin levels after single food-sharing events compared with other types of social feeding, irrespective of previous social bond levels. Also, urinary oxytocin levels following food sharing were higher than following grooming, another cooperative behaviour. Therefore, food sharing in chimpanzees may play a key role in social bonding under the influence of oxytocin. We propose that food-sharing events co-opt neurobiological mechanisms evolved to support mother–infant bonding during lactation bouts, and may act as facilitators of bonding and cooperation between unrelated individuals via the oxytocinergic system across social mammals.  相似文献

Electrospinning is a fascinating technique to fabricate micro- to nano-scale fibers from a wide variety of materials. For biopolymers, molecular entanglement of the constituent polymers in the spinning dope was found to be an essential prerequisite for successful electrospinning. Rheology is a powerful tool to probe the molecular conformation and interaction of biopolymers. In this report, we demonstrate the protocol for utilizing rheology to evaluate the electrospinnability of two biopolymers, starch and pullulan, from their dimethyl sulfoxide (DMSO)/water dispersions. Well-formed starch and pullulan fibers with average diameters in the submicron to micron range were obtained. Electrospinnability was evaluated by visual and microscopic observation of the fibers formed. By correlating the rheological properties of the dispersions to their electrospinnability, we demonstrate that molecular conformation, molecular entanglement, and shear viscosity all affect electrospinning. Rheology is not only useful in solvent system selection and process optimization, but also in understanding the mechanism of fiber formation on a molecular level.  相似文献

Development of the pteridine pathway in the zebrafish, Danio rerio   总被引:1,自引:0,他引:1  
In the zebrafish, the peripheral neurons and the pigment cells are derived from the neural crest and share the pteridine pathway, which leads either to the cofactor tetrahydrobiopterin or to xanthophore pigments. The components of the pteridine pattern were identified as tetrahydrobiopterin, sepiapterin, 7-oxobiopterin, isoxanthopterin, and 2,4,7-trioxopteridine. The expression of GTP cyclohydrolase I activity during the first 24-h postfertilization, followed by 6-pyruvoyl-5,6,7,8-tetrahydropterin synthase and sepiapterin reductase, suggest an early supply of tetrahydrobiopterin for neurotransmitter synthesis in the neurons and for tyrosine supply in the melanophores. At 48-h postfertilization, sepiapterin formation branches off the de novo pathway of tetrahydrobiopterin synthesis. Sepiapterin, via 7,8-dihydrobiopterin and biopterin, serves as a precursor for the formation of 7-oxobiopterin, which may be further catabolized to isoxanthopterin and 2,4,7-trioxopteridine. Neither 7, 8-dihydrobiopterin nor biopterin is a substrate for xanthine oxidoreductase. In contrast, both of these compounds are oxidized at C-7 by a xanthine oxidase variant form, which is inactivated by KCN, but is insensitive to allopurinol. The oxidase and the dehydrogenase form of xanthine oxidoreductase as well as the xanthine oxidase variant have specific developmental patterns. It follows that GTP cyclohydrolase I, the formation of sepiapterin, and the xanthine oxidoreductase family control the pteridine pathway in the zebrafish.  相似文献

Long‐term monitoring is vital to understanding the efficacy of restoration approaches and how restoration may enhance ecosystem functions. We revisited restored oyster reefs 13 years post‐restoration and quantified the resident and transient fauna that utilize restored reefs in three differing landscape contexts: on mudflats isolated from vegetated habitat, along the edge of salt marsh, and in between seagrass and salt marsh habitat. Differences observed 1–2 years post‐restoration in reef development and associated fauna within reefs restored on mudflats versus adjacent to seagrass/salt marsh and salt marsh‐only habitats persisted more than 10 years post‐restoration. Reefs constructed on open mudflat habitats had the highest densities of oysters and resident invertebrates compared to those in other landscape contexts, although all restored reefs continued to enhance local densities of invertebrate taxa (e.g. bivalves, gastropods, decapods, polychaetes, etc.). Catch rates of juvenile fishes were enhanced on restored reefs relative to controls, but to a lesser extent than directly post‐restoration, potentially because the reefs have grown vertically within the intertidal and out of the preferred inundation regime of small juvenile fishes. Reef presence and landscape setting did not augment the catch rates of piscivorous fishes in passive gill nets, similar to initial findings; however, hook‐and‐line catch rates were greater on restored reefs than non‐reef controls. We conclude that ecosystem functions and associated services provided by restored habitats can vary both spatially and temporally; therefore, a better understanding of how service delivery varies among landscape setting and over time should enhance efforts to model these processes and restoration decision‐making.  相似文献

Summary Choline, a necessary compound in an artificial diet for phloem-feeding aphids, was determined quantitatively in the phloem exudates of 16 tree species. The method used was a combination of choline oxidase action and oxygen determination with an oxygen electrode. The crustacean cuticle is a hierarchically organised material composed of an organic matrix and mineral. It is subdivided into skeletal elements whose physical properties are adapted to their function and the eco-physiological strains of the animal. Using a variety of ultrastructural and analytical techniques we studied the organisation of the tergite cuticle of the sand burrowing beach isopod Tylos europaeus. The surface of the tergites bear epicuticular scales, sensilla and micro-tubercles. A distal layer of the exocuticle is characterised by a low density of organic fibres and the presence of magnesium-calcite. Surprisingly, the mineral forms regions containing polyhedral structures alternating with smooth areas. Between sub-domains within the distal exocuticle calcite varies in its crystallographic orientation. Proximal layers of the exocuticle and the endocuticle are devoid of calcite and the mineral occurs in the form of amorphous calcium carbonate (ACC). Using thin sections of mineralised cuticle we describe for the first time that ACC forms tubes around single protein-chitin fibrils.  相似文献

Unialgal cultures of the macroscopic, vesicular, coenocytic gametophyte (Halicystis parvula Schmitz) of Derbesia tenuissima (DeNotaris) Crouan fr. were grown under various environmental regimes to elucidate the cytology of gamete formation and the factors controlling synchronous gamete formation and release. No synchrony of nuclear division was observed in vegetative plants or during the early stages of gamete formation. In the later stages of gamete formation in plants in a light-dark cycle, nuclear divisions within any gametangium were synchronous, and the stages of gamete formation were synchronous for the population. This synchrony was not as great for plants in continuous light. Gametes of plants in a light-dark cycle were released explosively immediately following the dark-to-light transition. Release was random and much less forceful for plants in continuous light. After a certain stage of gamete formation, gamete release was timed to occur after a particular interval of darkness, but release could be triggered by light during the last portion of this interval. The length of the dark interval was shorter for male plants than for females, but the period of light sensitivity was longer for females. Formation of gametangia by series of isolated plants was also synchronous and sometimes periodic under certain conditions. Intervals between gametangia on the same plant varied from 2 to 14 days but were usually 4 or 5 days (unlike plants in nature, which show a bi- or tri-weekly periodicity). Male and female plants did not differ in synchrony or periodicity. Different media affected the number of gametangia formed over a period of time but not the synchrony of formation. Under some conditions changing the medium had a stimulating or synchronizing effect. Non-repeated temperature changes also synchronized gamete formation. Optimum temperature for continued gamete formation was about 21 C. Regular daily light and temperature variation together maintained synchronous and periodic gamete formation in populations of isolated plants. Reproduction diminished and became less synchronous at constant temperature either in continuous light or under a light-dark schedule, although in the light-dark regime steps in the formation of any given gametangium remained synchronous with the light-dark cycle. Length of times between gametangial formation on individual plants showed a tendency to occur in multiples of the usual period lengths; e.g., plants sometimes tend to “skip” intervals, thus maintaining the synchrony of the population. These results suggest that interaction between daily environmental cycles and an endogenous physiological cycle may maintain periodic reproduction.  相似文献

All of the cells of the upper (adaxial) epidermis of the leaves ofOxalis carnosa are transformed into large bladders, while in the lower epidermis the bladder cells are interrupted by “normal” cells with stomata. The epidermal bladders contain a high concentration of free oxalic acid (pH approx. 1). Water-relations parameters of these epidermal bladder cells have been determined using the pressure probe. Original cell turgor (P0) of the closely packed bladders of theupper epidermis was P0=0.7 to 2.9 bar ( \(\overline {P_0 } = 1.7 \pm 0.5 bar\) ; mean±SD;N=25 cells) and lower than that in the club-shaped bladders of thelower epidermis (P0=1.3 to 3.7 bar; \(\overline {P_0 } = 2.5 \pm 0.7 bar\) ;N=25 cells). Large differences in the elastic modulus (ε) and the hydraulic conductivity (Lp) of the two different types of cells were observed. For the lower epidermal bladders, ε=18 to 166 bar and was similar to that of other higher plant cells. Also, for these cells it was found that ε was increasing with both, cell turgor and cell volume. By contrast, ε of the cells of the upper epidermis was by one order of magnitude smaller (ε=1.9 to 17.0 bar) and no dependence of ε on cell volume could be detected. The Lp values of the cell membranes were also different (lower epidermis: \(\overline {Lp} = (2.3 \pm 1.6) \cdot 10^{ - 5} cm s^{ - 1} bar^{ - 1}\) ; upper epidermis: \(\overline {Lp} = (3.8 \pm 2.4) \cdot 10^{ - 6} cm s^{ - 1} bar^{ - 1}\) ). These differences seem to be too large to be caused by errors in determining the exchange area for water (A) between cells and adjacent tissue. The half-times of water exchange between bladders and leaf (T1/2) were, on average, somewhat longer for the upper than for the lower epidermis (lower epidermis: T1/2=7 to 38 s; upper epidermis: T1/2=22 to 213 s), but the differences in the T1/2 values were not as distinct as for ε and Lp. This is because of the compensatory effects of ε, Lp and the different ratios of volume to exchange area. Since the bladders make up about 75% of the entire volume of the leaf, it is assumed that the rate of response of the leaf to changes in the water potential should be similar to that of the bladder cells. The results are discussed in terms of a possible function of the bladders in the leaf.  相似文献
