Mechanisms of plasmid-mediated antibiotic resistances in Vibrio parahaemolyticus

The mechanisms of drug resistance of clinical isolate, Vibrio (V.) parahaemolyticus ST550, resistant to chloramphenicol (CP), aminoglycoside antibiotics (AGs) and beta-lactam antibiotics were investigated. The mechanisms of resistance to CP, AGs and beta-lactam antibiotics were dependent on chloramphenicol acetyltransferase (CAT), aminoglycoside-3"-adenylyltransferase AAD(3") and aminoglycoside-3'-phosphotransferase APH(3') and TEM type penicillinase, respectively.     

Suppressive effect of human natural killer cells on pokeweed mitogen-induced B cell differentiation

The suppressive effect of human natural killer (NK) cells on B cell differentiation induced by pokeweed mitogen (PWM) was investigated. By using Percoll discontinuous density gradient centrifugation, peripheral blood nonphagocytic and nonadherent mononuclear cells were divided into low and high density fractions for which NK cells (Large granular lymphocytes, LGL) and T cells were enriched, respectively. These fractionated mononuclear cells were co-cultured with purified autologous B cells in the presence of PWM, and were examined for their helper and suppressor activities on differentiation of B cells to immunoglobulin-(IgM and IgG) producing cells by a highly sensitive reversed hemolytic plaque assay. The T cell-enriched high density fractions provided help for B cell differentiation to levels higher than that of unfractionated mononuclear cells. On the other hand, the NK-enriched low density fractions did not show helper activity, and when added to the culture of B cells plus helper T cells, they markedly suppressed B cell differentiation. This suppressive activity, as well as the NK cytotoxicity of the NK-enriched fractions, was abrogated by treatment of the cells with monoclonal antibody against human NK cells (HNK-1), but not against T cells (OKT3) in the presence of complement. NK cells also suppressed PWM-driven B cell differentiation in the presence of T4+ (helper/inducer T) but not T8+ (cytotoxic/suppressor T) cells; however, they showed no inhibition of soluble factor-induced B cell differentiation assayed in the absence of helper T cells. It is thus concluded that human peripheral blood NK cells exhibit an ability to suppress PWM-driven B cell differentiation, possibly by acting through the effect on helper T cells but not directly on B cells.     

The occurrence of polysialogangliosides including ganglio-N-tetraose series in adult bovine nasal cartilage

We extracted glycolipids from adult bovine nasal cartilage and purified some glycolipids by DEAE-Sephadex A-25 and Iatrobeads column chromatography. Cartilage contained 20 nmol of lipid bound sialic acid per gram wet tissue. The relative content of mono, di, tri, and tetrasialo gangliosides were 14%, 40%, 28% and 18%, respectively, as sialic acid content. We characterized some by examining carbohydrate composition, methylation analysis, sialidase treatment and mild acid hydrolysis. The ganglio-N-tetraose series, including GDla, GDlb, GTla, GTlb and GQlb, was identified as one of the major ganglioside groups of this cartilage.

Zusammenfassung Die spektrale Transmission wurde mit dem UMSP (UniversalMicro-Spektralphotometer) im Bereich von 550–300 nm am fixierten (Carnoy) und frischen dioptrischen Apparat einzelner Ommatidien von Aeschna cyanea Müll. (Odonata) gemessen.Die Transmission der Kristallkegel und der frischen Cornea ist zwischen 550–370 nm nahezu konstant; bei kürzeren Wellenlängen des UV-Bereiches sinkt die Transmission etwas. Erst bei Wellenlängen, die kürzer als 330 nm sind, wird der Transmissionsabfall signifikant.

The spectral transmission of the dioptric apparatus of aeschna

Summary Spectral transmission was measured by means of a Zeiss Universalmicro-spectralphotometer between 550 and 300 nm on fixed (Carnoy) and also on fresh cornea and crystalline cones of single ommatidia of Aeschna cyanea Müll. (Odonata).Both crystalline cones and fresh corneas (in contrast to fixed corneas) have nearly constant transmission from 550 to 370 nm; in uv-range with shorter wavelengths the transmission decreases slightly. Only at wavelengths shorter than 330 nm is the absorption significant.

Oxidized and reduced nicotinamide adenine dinucleotide phosphate levels of plants hardened and unhardened against chilling injury

Pea plants (Pisum sativum L. var. Alaska) subjected to low temperature (5°) in the light acquired resistance against chilling injury.

Unhardened plants maintained high NADP and low NADPH levels during illumination at 25° but hardened plants had low NADP and high NADPH levels in the light. When the unhardened plants were transferred to the dark room at 25°, their NADPH levels decreased immediately. On the other hand, hardened plants maintained a high NADPH level for a few hours even in the dark.
