Adipose tissue is closely associated with angiogenesis and vascular remodeling. Chemerin is involved in inflammatory reaction and vascular dysfunction. However, the mechanisms of chemerin participating in vascular remodeling and whether Growth arrest‐specific homeobox (Gax) can effectively intervene it remain obscured. Here, 3T3‐F442A preadipocytes were cultured, injected into athymic mice to model fat pads, and treated respectively with Ad‐chemerin, Ad‐Gax, or specific inhibitors in vitro and in vivo. MTT, flow cytometry, Western blotting, and imunohisto(cyto)‐chemistry analyses showed that chemerin enhanced the expression of FABP4 and VEGF, activated Akt/mTOR and ERK pathways, increased the cell percent of S phase, decreased the percent of G0‐G1 phase and apoptotic cells, and augmented neovascular density in fat pads. Inversely, Gax suppressed the expression of these adipogenic and vasifactive markers and these signaling proteins, decreased the percent of S phase cells, and increased those of G0‐G1 phase and apoptotic cells, and reduced the neovascular density. Our results indicate that chemerin‐CMKLR1 activates Akt/mTOR and ERK pathways and facilitates preadipocyte proliferation, adipogenesis, and angiogenesis. Contrarily, Gax weakens the effect of chemerin on preadipocyte biofunctions.  相似文献

We have recently shown that hyperosmotic stress activates p65/RelB NFkappaB in cultured cardiomyocytes with dichotomic actions on caspase activation and cell death. It remains unexplored how NFkappaB is regulated in cultured rat cardiomyocytes exposed to hyperosmotic stress. We study here: (a) if hyperosmotic stress triggers reactive oxygen species (ROS) generation and in turn whether they regulate NFkappaB and (b) if insulin-like growth factor-1 (IGF-1) modulates ROS production and NFkappaB activation in hyperosmotically-stressed cardiomyocytes. The results showed that hyperosmotic stress generated ROS in cultured cardiac myocytes, in particular the hydroxyl and superoxide species, which were inhibited by N-acetylcysteine (NAC). Hyperosmotic stress-induced NFkappaB activation as determined by IkappaBalpha degradation and NFkappaB DNA binding. NFkappaB activation and procaspase-3 and -9 fragmentation were prevented by NAC and IGF-1. However, this growth factor did not decrease ROS generation induced by hyperosmotic stress, suggesting that its actions over NFkappaB and caspase activation may be due to modulation of events downstream of ROS generation. We conclude that hyperosmotic stress induces ROS, which in turn activates NFkappaB and caspases. IGF-1 prevents NFkappaB activation by a ROS-independent mechanism.  相似文献

In the fiddler crab, Uca annulipes, males attract receptive females into their burrows by waving their greatly enlarged major claw. We have previously shown that males clustered around a female wave in close synchrony. Females may have a preference for leading signals and synchronised waving may arise as an epiphenomenon of competition between males to signal first. Indeed, the males in clusters that females approach and visit in their burrows are more likely to produce leading waves than are their neighbours. Here we document two other differences in the waving behaviour of visited males and their neighbours. First, visited males complete the downward component of the wave more rapidly than their neighbours. Second, the interval between the end of one wave and the start of the next is shorter for visited males. How can waving be synchronous if visited males wave faster than their neighbours? While only 9% (40/431) of waves by neighbours did not overlap those of the visited male, 22% (110/501) of visited male waves did not overlap the wave of a focal neighbour (111 visited male-neighbour dyads). Hence, while overlapping waves are nearly synchronous, visited males produce additional, ‘nonoverlapping’ waves that result in a higher wave rate than that of their neighbours.  相似文献

Lysosome-associated membrane protein (LAMP)-1, one of the major protein components of the lysosomal membrane, is upregulated in the human glioblastoma cell lines, U-373 MG and LN-Z308, which undergo cisplatin-induced apoptosis. These human brain tumor cell lines demonstrated apoptosis in response to cisplatin/nifedipine treatment. Both cell lines demonstrated an apoptotic response by more than one criterion. Apoptosis was demonstrated by DNA fragmentation techniques such as DNA laddering, ApopTag in situ labeling, and an ELISA-based method of detecting liberated oligosomes. These cells also had characteristic morphologic changes and upregulation of bax consistent with apoptosis. LAMP-1 expression at the protein and mRNA level was examined and found to increase with cisplatin/nifedipine treatment. LAMP-1 expression was examined using indirect immunofluorescent staining, Northern blot analysis and Western blot analysis. The finding of an augmentation of LAMP-1 in these cells induced to die is enigmatic. These findings raise the possibility of LAMP-1 involvement in the apoptotic process.  相似文献

1. Eutrophic acid lakes are not common. Delamere Lake in Cheshire, U.K. is shallow and acid (mean pH 4.5) with a very high phytoplankton crop (mean 290 μg chlorophyll a L?1), dominated by Dictyosphaerium pulchellum. Rotifers were dominant in the pelagic waters but small cladocerans (Alona guttata, Chydorus sphaericus and Scapholeberis mucronata) were occasional in the littoral waters. Chaoborus flavicans larvae were the top predators in this fishless lake. Two mesocosm experiments were carried out in which pH and Chaoborus populations were manipulated. 2. Progressively higher concentrations of D. pulchellum were maintained in the elevated pH treatments (pH 6 and 8; P < 0.001) with increased amounts of a Chlamydomonas species at the end of the experiment. Highest species richness was seen at ambient pH. Thus the low pH of Delamere Lake alone did not control the structure of the phytoplankton community. Keratella quadrata showed significantly higher abundance at pH 6 than in other pH treatments (P < 0.001). Species richness of rotifers was unaffected by pH. 3. Most Cladocera were C. sphaericus. Although never seen in the open lake, Daphnia pulex appeared in all the pH treatments. Low pH did not control small Cladocera abundance in Delamere Lake, but probably hampered reproduction in Daphnia. Negative correlations between chlorophyll a concentrations and Daphnia in the mesocosms (r2 = 0.215, P < 0.05), however, indicated the potential of large‐bodied daphniids in controlling phytoplankton. 4. Neither different combinations of Chaoborus instars (none, instars 1 and 2 and instars 3–5) nor different densities of instars 3–5 (0.15, 0.5 and 1.0 L?1) had a negative impact on Cladocera. Daphnia pulex remained unaffected in the experiment, perhaps because of its large size, and C. sphaericus because of its high reproductive rate compensating predatory losses. 5. Very low pH in Delamere Lake might suppress Daphnia by hampering its reproduction. Consequently, Daphnia may be vulnerable to invertebrate predation even at low predator density in the lake.  相似文献

The design and implementation of controlled environments to continuously culture and evolve viruses provides a means to track how their populations respond to natural and designed anti-viral agents. We have previously demonstrated how the growth of viruses in spreading plaques enables detection and characterization of their evolutionary dynamics. Using plaques of phage T7 growing on E. coli as a model system, we observe here that velocities of propagation can be readily controlled by the level of anti-viral antiserum incorporated into the propagation medium. Further, we develop a simple analytic expression for the radial velocity of propagation in terms of the microscopic rates of viral amplification, Fickian diffusion of the virions and their neutralization by antiserum. Our analysis captures the essential dependence of propagation velocity on antiserum concentration. This study provides an ex vivo foundation for exploring how medically relevant viruses escape suppression by the immune system. (c) 1997 John Wiley & Son, Inc. Summary Coexistence of peptides in the small intensely fluorescent cells was demonstrated by immunocytochemistry for met-enkephalin-Arg-Gly-Leu, vasoactive intestinal polypeptide, somatostatin, neuropeptide Y and dynorphin. In the extreme example, a single cell was immunoreactive to all 5 peptides examined. Four peptides coexisted in 8% and three peptides in 13% of SIF cells. In 10% of SIF cells no peptide immunoreactivity could be detected. The most prevalent peptide was met-enkephalin (in 46% of cells), then vasoactive intestinal polypeptide (45%), somatostatin (39%), neuropeptide Y (31%) and dynorphin (24%). Met-enkephalin and vasoactive intestinal polypeptide coexisted most commonly (25%).  相似文献

This paper describes the production and calibration of a miniature psychrometer treated with a specially developed porous coating. The investigation was conducted to determine localized patterns of rapidly changing temperature and relative humidity in dynamic flowing gas environments (e.g., with particular attention to future applications to the human respiratory system). The technique involved the use of dry miniature thermocouples and wetted miniature thermocouples coated with boron nitride to act as a wicking material. A precision humidity generator was developed for calibrating the psychrometer. It was found that, in most cases, the measured and expected (i.e., theoretically predicted) relative humidity agreed to within 0.5 to 1.0 percent relative humidity. Procedures that would decrease this discrepancy even further were pinpointed, and advantages of using the miniature psychrometer were assessed.  相似文献
