Mdm2 harnesses the p53 tumor suppressor, yet loss of one Mdm2 allele in Mdm2(+/-) mice has heretofore not been shown to impair tumor development. Here we report that Mdm2 haplo-insufficiency profoundly suppresses lymphomagenesis in E micro -myc transgenic mice. Mdm2(+/-)E micro -myc transgenics had greatly protracted rates of B cell lymphoma development with life spans twice that of wild-type transgenic littermates. Im paired lymphoma development was associated with drastic reductions in peripheral B cell numbers in Mdm2(+/-)E micro -myc transgenics, and primary pre-B cells from Mdm2(+/-)E micro -myc transgenics and Mdm2(+/-) littermates were extremely susceptible to spontaneous apoptosis. Loss of p53 rescued all of the effects of Mdm2 haplo-insufficiency, indicating they were p53 dependent. Furthermore, half of the lymphomas that ultimately emerged in Mdm2(+/-)E micro -myc transgenics harbored inactivating mutations in p53, and the majority overcame haplo-insufficiency by overexpressing Mdm2. These results support the concept that Mdm2 functions are rate limiting in lymphomagenesis and that targeting Mdm2 will enhance p53-mediated apoptosis, compromising tumor development and/or maintenance. 相似文献

With the worldwide prevalence of cardiovascular diseases, much attention has been focused on simulating the characteristics of the human heart to better understand and treat cardiac disorders. The purpose of this study is to build a finite element model of the left atrium (LA) that incorporates detailed anatomical features and realistic material characteristics to investigate the interaction of heart tissue and surgical instruments. This model is used to facilitate the design of an endoscopically deployable atrial retractor for use in minimally invasive, robotically assisted mitral valve repair. Magnetic resonance imaging (MRI) scans of a pressurized explanted porcine heart were taken to provide a 3D solid model of the heart geometry, while uniaxial tensile tests of porcine left atrial tissue were conducted to obtain realistic material properties for noncontractile cardiac tissue. A finite element model of the LA was constructed using ANSYS Release 9.0 software and the MRI data. The Mooney-Rivlin hyperelastic material model was chosen to characterize the passive left atrial tissue; material constants were derived from tensile test data. Finite element analysis (FEA) models of a CardioVations Port Access retractor and a prototype endoscopic retractor were constructed to simulate interaction between each instrument and the LA. These contact simulations were used to compare the quality of retraction between the two instruments and to optimize the design of the prototype retractor. Model accuracy was verified by comparing simulated cardiac wall deflections to those measured by MRI. FEA simulations revealed that peak forces of approximately 2.85 N and 2.46 N were required to retract the LA using the Port Access and prototype retractors, respectively. These forces varied nonlinearly with retractor blade displacement. Dilation of the atrial walls and rigid body motion of the chamber were approximately the same for both retractors. Finite element analysis is shown to be an effective tool for analyzing instrument/tissue interactions and for designing surgical instruments. The benefits of this approach to medical device design are significant when compared to the alternatives: constructing prototypes and evaluating them via animal or clinical trials. 相似文献

Background
The two sympatric species of Tunisian desert ants, Cataglyphis bicolor and C. mauritanica, do not exhibit any differences in their foraging ecology, e.g. in food preferences and in their spatial and temporal activity patterns. Here we show that instead the two species markedly differ in their life histories.Results
We analysed mtDNA of specimens that were collected along a 250-km transect. C. bicolor exhibited a genetically unstructured population (with the genetic and geographic distances among colonies not being correlated). On the contrary the populations of the polygynous C. mauritanica were clearly structured, i.e. exhibited a strong correlation between genetic and geographic distances. This difference is in accordance with large queen dispersal distances due to far-reaching mating flights in C. bicolor and small queen dispersal distances due to colony foundation by budding in C. mauritanica. Furthermore, wherever we found populations of both species to coexist within the same habitat, the habitat was used agriculturally. Mapping nest positions over periods of several years showed that plowing dramatically decreased the nest densities of either species.Conclusion
We conclude that owing to its greater queen dispersal potential C. bicolor might be more successful in quickly re-colonizing disturbed areas, while the slowly dispersing C. mauritanica could later out-compete C. bicolor by adopting its effective nest-budding strategy. According to this scenario the observed sympatry of the two species might be an intermediate stage in which faster colonization by one species and more powerful exploitation of space by the other species have somehow balanced each other out. In conclusion, C. bicolor and C. mauritanica represent an example where environmental disturbances in combination with different life histories might beget sympatry in congeneric species with overlapping niches. 相似文献44.
Autophagy regulates cellular homeostasis through degradation of aged or damaged subcellular organelles and components. Interestingly, autophagy-deficient beta cells, for example Atg7-mutant mice, exhibited hypoinsulinemia and hyperglycemia. Also, autophagy response is diminished in heart of diabetic mice. These results implied that autophagy and diabetes are closely connected and affect each other. Although protein O-GlcNAcylation is up-regulated in hyperglycemia and diabetes, and O-GlcNAcylated proteins play an important role in metabolism and nutrient sensing, little is known whether autophagy affects O-GlcNAc modification and vice versa. In this study, we suppressed the action of mTOR by treatment of mTOR catalytic inhibitors (PP242 and Torin1) to induce autophagic flux. Results showed a decrease in global O-GlcNAcylation, which is due to decreased OGT protein and increased OGA protein. Interestingly, knockdown of ATG genes or blocking of lysosomal degradation enhanced protein stability of OGT. In addition, when proteasomal inhibitor was treated together with mTOR inhibitor, protein level of OGT almost recovered to control level. These data suggest that mTOR inhibition is a more efficient way to reduce protein level of OGT rather than that of CHX treatment. We also showed that not only proteasomal degradation regulated OGT stability but autophagic degradation also affected OGT stability in part. We concluded that mTOR signaling regulates protein O-GlcNAc modification through adjustment of OGT stability. 相似文献

The forelimb digital flexors of the horse display remarkable diversity in muscle architecture despite each muscle-tendon unit having a similar mechanical advantage across the fetlock joint. We focus on two distinct muscles of the digital flexor system: short compartment deep digital flexor (DDF(sc)) and the superficial digital flexor (SDF). The objectives were to investigate force-length behavior and work performance of these two muscles in vivo during locomotion, and to determine how muscle architecture contributes to in vivo function in this system. We directly recorded muscle force (via tendon strain gauges) and muscle fascicle length (via sonomicrometry crystals) as horses walked (1.7 m s(-1)), trotted (4.1 m s(-1)) and cantered (7.0 m s(-1)) on a motorized treadmill. Over the range of gaits and speeds, DDF(sc) fascicles shortened while producing relatively low force, generating modest positive net work. In contrast, SDF fascicles initially shortened, then lengthened while producing high force, resulting in substantial negative net work. These findings suggest the long fibered, unipennate DDF(sc) supplements mechanical work during running, whereas the short fibered, multipennate SDF is specialized for economical high force and enhanced elastic energy storage. Apparent in vivo functions match well with the distinct architectural features of each muscle. 相似文献

This report describes a lysozyme expressed at high levels in the stomach of
the hoatzin, the only known foregut-fermenting bird. Evolutionary
comparison places it among the calcium-binding lysozymes rather than among
the conventional types. Conventional lysozymes were recruited as digestive
enzymes twice in the evolution of mammalian foregut fermenters, and these
independently recruited lysozymes share convergent structural changes
attributed to selective pressures in the stomach. Biochemical convergence
and parallel amino acid replacements are observed in the hoatzin stomach
lysozyme even though it has a different genetic origin from the mammalian
examples and has undergone more than 300 million years of independent
evolution.

Comparisons were made between the infestation levels of the honey bee tracheal miteAcarapis woodi (Rennie) in newly emerged honey bees (Apis mellifera L.) exposed for 12 h during the daytime or nighttime in mite-infested bee colonies. Bees exposed during the night harbored a significantly higher number of mites (718) when compared with the daytime bees (88 mites) (n=14 day/night cycles utilizing 33 colonies). On 4 days of an 8-day study, three test colonies were closed during the daytime to eliminate foraging flights. Thus equal numbers of bees were present in the colonies during the day and night sample periods. These 4 flightless days were compared to 4 free-flight days and mite dispersal rates were not significantly different. Additionally, the movement of bees on the combs of four glass-walled observation hives was quantified on 10 days at 0800, 1200, 1600, 2000, 2400 and 0400 h. Bee movement at 2400 and 0400 h was significantly lower than the other observation times. Movement of host bees may be one factor involved in the increased nighttime mite dispersals. These findings do not support the hypothesis that the absence of foraging bees during the day reduces the bee to bee contact time, thus reducing mite dispersals between host bees. Differential diurnal activity levels between host bees and mite parasites was demonstrated. The exact role of host-bee behavior and/or mite behavior in the nighttime dispersal patterns observed, remains for further investigation. 相似文献
