The effect of melittin, an activator of phospholipase A2, on proliferation and death of rat thymocytes in a broad concentration range was studied. Cell proliferation was estimated by the accumulation of colchicin metaphases, necrotic death was determined from lysis and staining of cells with trypan blue, and apoptosis was assessed from the type of DNA fragmentation, the amount of fragmented DNA, and the percentage of cells with subdiploid DNA. It was shown that low melittin concentrations (below 5 μg/ml) stimulate thymocyte proliferation. At high melittin concentrations, thymocytes die by the primary necrosis type. Throughout the concentration range studied, melittin does not produce apoptosis in thymocytes. Conversely, high melittin concentrations even inhibit thymocyte apoptosis in the control and after irradiation. An inhibitor of RNA synthesis actinomycin D does not affect thymocyte death in the presence of melittin. It is concluded that the activation of phospholipase A2 can induce necrosis but not apoptosis and thus is not a necessary step in the signaling cascade that initiates apoptosis in thymocytes.  相似文献

The time course of poststimulatory adaptation of the inferior colliculus central nucleus (ICC) of CBB6F1 hybrid mice to sound sequences, specifically, series of four tonal stimuli presented at intervals of 0, 2, 4, 10, 20, 50, 100, 200, 500, 700, 1000, and 1500 ms were studied. Assessment of the adaptation of the entire neuronal population have shown that, at an interstimulus interval of 0–200 ms, the response to the first tone in a series is significantly stronger than those to the second to fourth tones, the strengths of the latter three responses not differing significantly from one another. If the interstimulus interval is 500 ms or longer, the response to none of the tones in a series differs significantly in strength from the others. The role of adaptation of midbrain neurons to the grouping of components of bioacoustic stimuli is discussed.  相似文献

A study was made of the effect of ionizing radiation, phospholipase A2, and low concentrations of mediators (for instance, acetylcholine and gamma-aminobutyric acid) on the content of cyclic nucleotides and phosphorylation of membrane proteins of Helix pomatia nervous ganglia. Ionizing radiation was shown to decrease considerably the levels of cAMP and cGMP which correlated with the diminution of membrane phosphorylation. Phospholipase A2 and low doses of mediators produced a modifying effect on the cyclic nucleotide content.  相似文献

Tumor necrosis factor receptor-associated protein-1 (TRAP1) is a mitochondrial (MITO) antiapoptotic heat-shock protein. The information available on the TRAP1 pathway describes just a few well-characterized functions of this protein in mitochondria. However, our group's use of mass-spectrometric analysis identified TBP7, an AAA-ATPase of the 19S proteasomal subunit, as a putative TRAP1-interacting protein. Surprisingly, TRAP1 and TBP7 colocalize in the endoplasmic reticulum (ER), as demonstrated by biochemical and confocal/electron microscopic analyses, and interact directly, as confirmed by fluorescence resonance energy transfer analysis. This is the first demonstration of TRAP1's presence in this cellular compartment. TRAP1 silencing by short-hairpin RNAs, in cells exposed to thapsigargin-induced ER stress, correlates with upregulation of BiP/Grp78, thus suggesting a role of TRAP1 in the refolding of damaged proteins and in ER stress protection. Consistently, TRAP1 and/or TBP7 interference enhanced stress-induced cell death and increased intracellular protein ubiquitination. These experiments led us to hypothesize an involvement of TRAP1 in protein quality control for mistargeted/misfolded mitochondria-destined proteins, through interaction with the regulatory proteasome protein TBP7. Remarkably, expression of specific MITO proteins decreased upon TRAP1 interference as a consequence of increased ubiquitination. The proposed TRAP1 network has an impact in vivo, as it is conserved in human colorectal cancers, is controlled by ER-localized TRAP1 interacting with TBP7 and provides a novel model of the ER-mitochondria crosstalk.  相似文献

The ability of three substituted quinones, 2,5-dibromo-3-methyl-6-isopropyl-p-benzoquinone (DBMIB), 2,6-dichloro-p-benzoquinone (DCBQ), and tetramethyl-p-benzoquinone (duriquinone) to quench the excited states of chlorophyll (Chl) molecules in Photosystem I (PSI) was studied. Chl fluorescence emission measured with isolated PSI submembrane fractions was reduced following the addition of exogenous quinones. This quenching progressively increased with rising concentrations of the exogenous quinones according to the Stern-Volmer law. The values of Stern-Volmer quenching coefficients were found to be 3.28 x 10(5) M(-1) (DBMIB), 1.31 x 10(4) M(-1) (DCBQ), and 3.7 x 10(3) M(-1) (duroquinone). The relative quenching capacities of the various exogenous quinones in PSI thus strictly coincided to those found for the quenching of Fo level of Chl fluorescence in isolated thylakoids, which is emitted largely by Photosystem II (PSII) [Biochim. Biophys. Acta (2003) 1604, 115-123]. Quenching of Chl excited states in PSI submembrane fractions by exogenous quinones slowed down the rate of P700, primary electron donor of PSI, photooxidation measured at limiting actinic light irradiances thus revealing a reduced photochemical capacity of absorbed quanta. The possible involvement of non-photochemical quenching of excited Chl states by oxidized phylloquinones, electron acceptors of PSI, and oxidized plastoquinones, mobile electron carriers between PSII and the cytochrome b(6)/f complex, into the control of photochemical activity of PSI is discussed.  相似文献

A complex of clinical and laboratory examinations of patients with bacterial chronic prostatitis was carried out. The patients were divided into two groups: 55 patients treated by the method of immunotherapy and 20 patients (controls) undergoing basic therapy. Immunotherapy was carried out with the use of polycomponent vaccine VP-4 containing Staphylococcus aureus, Klebsiella pneumoniae, Proteus vulgaris and Escherichia coli antigens. Vaccinal therapy was indicated in cases with infection focus associated with opportunistic bacteria, a prolonged torpid course of prostatitis and when different schemes of etiotropic therapy proved to be ineffective. The vaccine was introduced in 4 or 5 subcutaneous injections at an interval of 5-7 days. The study revealed that in cases of chronic prostatitis accompanied by microecological and immune disturbances the use of complex therapy allowed to achieve good clinical and laboratory results in 91% of the patients. In the patients undergoing immunotherapy the elimination of opportunistic bacteria from the urogenital tract and the restoration of affected microflora occurred.  相似文献

Studies were carried out in 2000–2007 near Medusa Bay (73°21′N, 80°32′ E) and along the Agapa River (from 70°11′N, 86°15′ E. down to the mouth 71°26′ N, 89°13′ E), in the northwestern and central parts of the Taimyr Peninsula. White-fronted goose nests are usually spread in the tundra or placed in 1–3 nest colonies near nests or staging points of snowy owls, peregrine falcons, or rough-legged buzzards. The intent of white-fronted geese to breed near birds of prey or owls increases sharply when arctic fox numbers are high. In the area near Medusa Bay, white-fronted geese nest much closer to peregrine falcon nests than in the area along the Agra River. At the latter location, white-fronted geese lose the competition to red-breasted geese, which are more numerous here. Bean geese, in spite of their greater size and ability to protect their nests against arctic foxes, really tend to breed near peregrine falcons or buzzards, where they manage to compete with red-breasted geese.  相似文献
