Abstract Oral immunization of suckling mice with Cryptosporidium parvum results in a humoral response to a limited set of antigens. Six-day-old BALB/c mice were each inoculated orally with 1 × 106 viable oocysts and subsequently administered oral inoculations of 2 × 106 viable oocysts at 30 and 60 days following the primary infection. After 45 days, mice were boosted with 1 × 106 oocysts orally, plus soluble extracts equivalent to 2 × 106 and 1 × 106 oocysts given intravenously and intraperitoneally, respectively. Four days later, splenic lymphocytes were fused to Ag8 myeloma cells. Using this method, we have been able to select for monoclonal antibodies that predominately recognize sporozoite surface and apical complex antigens. 相似文献

Background
Recent progress in insect transgenesis has been dramatic but existing transposon-based approaches are constrained by position effects and potential instability. Gene targeting would bring a number of benefits, however progress requires a better understanding of the mechanisms involved. Much can be learned in vitro since extrachromosomal recombination occurs at high frequency, facilitating the study of multiple events and the impact of structural changes among the recombining molecules. We have investigated homologous recombination in mosquito cells through restoration of luciferase activity from deleted substrates. Information on migration patterns is critical to using no-take migratory corridors and marine reserves to protect the spawning stock of commercially exploited species. Both active and passive acoustic tracking methods quantified movement of commercially and ecologically important blue crabs in the White Oak River estuary, NC, USA. We targeted post-mating female crabs migrating down-estuary to oceanic spawning grounds. Crabs travelled approximately 14.1 km mainly in deeper channels and over 12–26 days from mating areas to spawning grounds. No crabs were detected migrating down-estuary in the autumn and only 30% were detected migrating down-estuary in spring. None of the crabs detected near spawning grounds were detected or recaptured back up-estuary, suggesting that they either (i) do not return to the estuary after a one to two week period in the spawning area or (ii) were captured by fishermen. The results from this study demonstrate that (1) acoustic transmitters coupled with passive acoustic receivers provided reliable and valuable data on migration patterns of mature female blue crabs and (2) mature female blue crabs are capable of migrating primarily within deep channels to spawning grounds shortly after insemination. 相似文献

Binary diastereomeric (-) (1R,2S)-ephedrine salts of various mandelic acids obtained from 95% ethanol show considerable differences in solubility. Structures and some properties of the less-soluble (L) and more-soluble (M) solid phases of (-)-ephedrine with unsubstituted mandelic acid, 2-, 3-, and 4-monosubstituted halo (F, Cl, Br) mandelic acids, and 3- and 4-methylmandelic acids have been determined. Salts were found to be binary, without solvent of crystallization, and composed of double-layered arrays of alternating anions and cations linked by H-bonds normal to the layers. H-bonding links charged donors and acceptors usually along a crystallographic 2-fold screw axis. A striking discrimination is evident in that the (2R)-mandelate salts typically display a compact four-atom chain as the H-bonding repeating unit [+N—H…O(—C?—O)…H-N′, C21(4)] while the (2S)-mandelate salts adopt a more dimensionally variable six-atom chain repeating unit [+N—H…O—C?—O…H—N′, C22(6)]. Two distinct packing schemes display the shorter H-bonding chain of the (2R)-mandelates which always occurs with ephedrinium ions in the fully extended conformation. Slightly greater packing efficiency and H-bonding energies of the (2R)-mandelate salts correlates with increased fusion points, lower solubilities (95% ethanol), and higher heats of fusion relative to the phase adopted by their diastereoisomers. In contrast, (2S)-mandelate salts exhibit considerably more structural variability involving all three major ephedrinium conformations, and at least four distinct packing motifs. Mandelates with larger 3′-substituents (Cl, Br, methyl) show similar property discriminations, but these occur with an opposing trend, that is, between phases in which the less-soluble salts contain (2S)-mandelates. The cis (2a) and trans (2b) isomers of methyl 3-benzamido-2-piperidinone-6-carboxylate (Apca) were prepared and separated by fractional recrystallizations. Proton n.m.r. studies in dimethylsulfoxide solution indicate that the six-membered lactam ring adopts a distorted chair conformation with an equatorially oriented benzamido substituent in both 2a and 2b. The carboxyl function also is equatorially oriented in the trans isomer 2b, but is disposed axially in the cis isomer 2a. In the crystal structure, the six-membered lactam ring of 2a is clearly in a boat conformation with the benzamido and carboxyl functions attached to the two apex carbon atoms equatorially. The trans isomer, 2b, exists as two crystallographically independent, conformationally distinct molecules in one unit cell. The lactam ring in both molecules adopts a distorted chair conformation, as is the case in solution, with both the benazamido and carboxyl functions attached equatorially. The rotameric orientation for the endocyclic lactam differs between the two molecules. Both structures show evidence of C-H...O hydrogen bond formation intermolecularly in the solid state. This ability, along with the distinctive conformational features of Apca, may be exploitable in the design of unique features of polypeptides. 相似文献
