Binding of 125I-labelled epidermal growth factor (EGF) to C3H/2K cells and the effect of a tumor promotor, 12-O-tetradecanoyl phorbol-13-acetate (TPA) and of a tumor promotor antagonist, retinoic acid, on the binding was studied. Scatchard plot analysis of the binding showed the presence of two types of binding sites with different affinity to EGF. Treatment of the cells with retinoic acid for 1 h resulted in elevation of the affinity of both sites without changing their number per cell. Prolonged exposure to retinoic acid abrogated this elevation of the affinity and caused cycloheximide-sensitive increase of the number of the binding sites of both types. TPA inhibited binding of EGF to the cells by abolishing the binding to the high affinity sites, whereas retinoic acid, in the presence of TPA, enhanced it by increasing the number of the low affinity sites.  相似文献

Summary Seed protein of 581 local strains of mung bean, Vigna radiata (L.) Wilczek, collected from throughout Asia, were analyzed by SDS-polyacrylamide gel electrophoresis. Eight protein types were recognized based on the combination of four albumin bands and three globulin bands. The frequency of each protein type strain showed a clear geographical cline. The pattern of geographical distribution of the protein types reflected the regions of genetic diversity, and two dissemination pathways in mung bean were proposed. The region of genetic diversity in seed protein was western Asia (Afghanistan-Iran-Iraq area). Mung bean may have spread mainly to the east by two routes from India, where the domestication of mung bean is believed to have occurred. One route led to Southeast Asia; strains consisting of a few protein types with prominent protein type 1 were disseminated from India to the Southeast Asian countreis. Thus, the strain composition in Southeast Asia was very simple, with the strains being similar to one another. Another dissemination pathway may have been the route known as the Silk Road. Since protein type 7 and 8 strains could not be found throughout Southeast Asia, it is assumed that these strains spread from western Asia or India to China and Taiwan via the Silk Road, and not by the route from Southeast Asia.  相似文献

Lipid A that contains mainly 2,3-diamino-2,3-dideoxy-D-glucose, phosphate and fatty acids in the molar ratio 2:1:5-6 was found in Pseudomonas diminuta lipopolysaccharide. The lipid A was considered to have a diamino-sugar disaccharide structure that carries a nonglycosidic phosphomonoester group and amide-bound acyloxyacyl and 3-hydroxy fatty acyl groups. The lipopolysaccharide exhibited endotoxic activities including lethal toxicity, pyrogenicity, local Shwartzman activity, body weight-decreasing toxicity and Limulus activity. The free lipid A was also endotoxic.  相似文献

Severe fever with thrombocytopenia syndrome (SFTS) caused by a species Dabie bandavirus (formerly SFTS virus [SFTSV]) is an emerging hemorrhagic infectious disease with a high case-fatality rate. One of the best strategies for preventing SFTS is to develop a vaccine, which is expected to induce both humoral and cellular immunity. We applied a highly attenuated but still immunogenic vaccinia virus strain LC16m8 (m8) as a recombinant vaccine for SFTS. Recombinant m8s expressing SFTSV nucleoprotein (m8-N), envelope glycoprotein precursor (m8-GPC), and both N and GPC (m8-N+GPC) in the infected cells were generated. Both m8-GPC- and m8-N+GPC-infected cells were confirmed to produce SFTSV-like-particles (VLP) in vitro, and the N was incorporated in the VLP produced by the infection of cells with m8-N+GPC. Specific antibodies to SFTSV were induced in mice inoculated with each of the recombinant m8s, and the mice were fully protected from lethal challenge with SFTSV at both 103 TCID50 and 105 TCID50. In mice that had been immunized with vaccinia virus strain Lister in advance of m8-based SFTSV vaccine inoculation, protective immunity against the SFTSV challenge was also conferred. The pathological analysis revealed that mice immunized with m8-GPC or m8-N+GPC did not show any histopathological changes without any viral antigen-positive cells, whereas the control mice showed focal necrosis with inflammatory infiltration with SFTSV antigen-positive cells in tissues after SFTSV challenge. The passive serum transfer experiments revealed that sera collected from mice inoculated with m8-GPC or m8-N+GPC but not with m8-N conferred protective immunity against lethal SFTSV challenge in naïve mice. On the other hand, the depletion of CD8-positive cells in vivo did not abrogate the protective immunity conferred by m8-based SFTSV vaccines. Based on these results, the recombinant m8-GPC and m8-N+GPC were considered promising vaccine candidates for SFTS.  相似文献

We investigated the effects of previously identified quantitative trait loci (QTL) in an experimental backcross (BC) between Chinese Meishan pigs and commercial Duroc pigs. We performed marker‐assisted introgression of two QTL for intramuscular fat (IMF) content (IMF population) and three QTL for reproductive traits (reproduction population) from a donor Meishan pig into a recipient Duroc pig. At the fourth BC generation of the IMF population and third BC generation of the reproduction population, carrier animals were selected for the production of animals homozygous for the QTL. Our previous studies have shown that the presence of a Meishan allele on the IMF QTL is associated with low IMF values, and the Meishan allele on the reproductive QTL is associated with large litters. In this study, the presence of a Duroc allele at the IMF QTL on SSC9 resulted in a 0.27% increase in IMF (additive effect = 0.27 ± 0.08), whereas the presence of a Meishan allele at the IMF QTL on SSC7 resulted in a 0.34% increase in IMF (additive effect = ?0.34 ± 0.09). The presence of the Meishan allele at the IMF QTL on SSC7 thus had the opposite effect to our previous studies, that is, increased IMF. In the reproduction population, we observed no differences between the genotypes of the three QTL in regard to number of corpora lutea or litter size. Marker‐assisted introgression at these QTL is thus unlikely to result in an associated increase in litter size. These results show that it is possible to introgress alleles from other breeds into a selection population using molecular markers; any unexpected results might be associated with the genetic background.  相似文献

Methods for the solubilization of an insect granulosis virus capsule from Trichoplusia ni were studied using turbidity measurements. The solubilization in the solvents employed was most strongly influenced by pH. Various alcohols and hydrogen bond cleaving agents also enhanced solubilization. The most effective solvent tested at pH′s close to neutrality was 60% n-propanol saturated with guanidine. HCl. The effectiveness of this solvent was minimal at pH 5.2. It was shown that the time courses of the solubilization were generally composed of three phases. A plot of the third phase levels versus pH indicated a normal distribution of those capsules or capsular fragments in resisting solubilization. Validity of the calculation of rate constants for the dissociation reaction in the second phase was discussed. Logarithm of the rate constant showed a linear relationship with the reciprocal of absolute temperature. The activation energy for the dissociation calculated from this relationship using the Arrhenius equation indicated that hydrophobic and hydrogen binding forces play the major roles for the stability of the crystalline structure of the capsular protein. Inic binding forces were estimated to be of minor importance.  相似文献

Resonance Raman spectra were observed for the threonine-301 to serine or valine mutant as well as the wild type of rabbit liver microsomal cytochrome P-450 [laurate(omega-1)-hydroxylase] (P-450(omega-1], which were prepared through site-directed mutagenesis. The high-spin marker resonance Raman (RR) bands became similarly stronger for all the P-450s examined in the oxidized form upon addition of laurate, and the RR spectra in the higher frequency region of the oxidized, reduced and CO-adduct forms did not distinctly differ among the P-450s examined. Nevertheless, the Fe-CO stretching mode (vFe-CO) of the CO adduct exhibited an upshift for the valine mutant, suggesting positional proximity of Thr-301 to bound CO like Thr-252 of P-450cam, in agreement with the expectation from the sequence analysis. The vFe-CO band was shifted to higher frequency upon binding of normal alkyl fatty acids with C10 or longer alkyl chain but little affected by binding of shorter fatty acids.  相似文献

It has been demonstrated that gamma delta T cells accumulating in early tumor lesions and those purified from spleen cells of tumor-bearing mice attenuate the activity of CTLs and NK cells. We, therefore, investigated whether depletion of gamma delta T cells from early lesions of tumors results in restoration of CTL and NK cell activities and subsequent regression of tumors. A daunomycin-conjugated anti-gamma delta TCR mAb UC7-13D5 (Dau-UC7) was prepared to efficiently deplete gamma delta T cells. An in vitro study revealed that Dau-UC7 specifically lysed gamma delta TCR+ cells and effectively inhibited splenic gamma delta T cells from tumor-bearing mice to produce cytotoxic cell-suppressive factors. Furthermore, intralesional injections of Dau-UC7 at an early stage of tumor development led to augmentation of tumor-specific CTL as well as NK cell activities and to the resultant regression or growth inhibition of the tumors. On analysis of cytokine profile, gamma delta T cells transcribed mRNAs for IL-10 and TGF-beta, but not IL-4 or IFN-gamma, suggesting the T regulatory 1-like phenotype. Finally, a blocking study with mAbs showed that the inhibitory action of gamma delta T cells on CTLs and NK cells was at least partly mediated by IL-10 and TGF-beta. These results clearly demonstrated the novel mechanism by which T regulatory 1-like gamma delta T cells suppress anti-tumor CTL and NK activities by their regulatory cytokines in early tumor formation.  相似文献

An enzyme which catalyzes the following esterase reaction was isolated from mouse serum: 12-O-tetradecanoyl phorbol 13-acetate (TPA) + H2O----phorbol 13-acetate + tetradecanoic acid. The recovery was 0.18% of total serum protein and 820-fold purification was achieved. The enzyme is composed of a single polypeptide chain with sugar moiety; its molecular weight was estimated to be 77,000. Its sugar content is 15%, the isoelectric point was 4.3, and the alpha-helix content was 15.3% . The activity is stable between pH 5 and 9 under 40 degrees C; it is insensitive to 2-mercaptoethanol and is not dependent on divalent cations. The optimal pH is around 7.5. The apparent Km for TPA is 6.6 X 10(-7)M. The hydrolysis of [3H]TPA is inhibited by phorbol diesters and phorbol 12-myristate, but not by phorbol and phorbol 13-acetate. The activity is inhibited to some extent by phosphatidylcholine, cholesterol, and lanosterol, but not by free fatty acids, fatty acid esters of glycerol, cholesterol esters, or cholestanol. The enzyme hydrolyzes ester linkages, but not peptide linkages of synthetic substrates. Esterase inhibitors and serine-reactive reagents affect the activity. Although sera from rodents displayed strong activity, such activity was not detected in human serum. Unlike lipoprotein lipase, the serum enzyme activity was not enhanced by treatment of the animal with heparin. These characteristics and the amino acid composition do not agree with any of the reported characteristics of known serum enzymes with esterase activity.  相似文献
