Optimization of our previously described pyrrolopiperidone series led to the identification of a new benzamide sub-series, which exhibits consistently high potency in biochemical and cell-based assays throughout the series. Strong inhibition of LPS-induced production of the cytokine TNFα is coupled to the regulation of HSP27 phosphorylation, indicating that the observed cellular effects result from the inhibition of MK2. X-ray crystallographic and computational analyses provide a rationale for the high potency of the series.  相似文献

HLA-A2 protects from EBV+ classical Hodgkin lymphoma (cHL) in Western Europe, but it is unknown whether this protective effect also exists in the Chinese population. We investigated the association of HLA-A2 and specific common and well documented HLA-A2 subtypes with EBV stratified cHL patients (n = 161) from the northern part of China. Quantitative-PCR and sequence-based subtyping was performed to identify HLA-A2 positive samples and their subtypes. 67 (42%) of the cHL patients were EBV+. There were no significant differences in percentages of HLA-A2 positivity between cHL and controls (65% vs 66%) and between EBV+ and EBV− cHL patients (70% vs 61%). The frequency distribution of HLA-A2 subtypes was significantly different between EBV stratified cHL subgroups and controls. This difference was most striking for the HLA-A*02:07 type with a frequency of 38% in EBV+ cHL, 8% in EBV− cHL and 20% in controls. Significant differences were also observed for the HLA-A*02:07, HLA-A2 (non-02:07) and the A2-negative typings between EBV+ cHL vs controls (p = 0.028), EBV− cHL vs controls (p = 0.045) and EBV+ vs EBV− cHL cases (p = 2×10−5). In conclusion, HLA-A*02:07 is a predisposing allele for EBV+ cHL and a protective allele for EBV− cHL in the northern Chinese population.  相似文献

Numerous studies have focused on song in songbirds as a signal involved in mate choice and intrasexual competition. It is expected that song traits such as song rate reflect individual quality by being dependent on energetic state or condition. While seasonal variation in bird song (i.e., breeding versus non-breeding song) and its neural substrate have received a fair amount of attention, the function and information content of song outside the breeding season is generally much less understood. Furthermore, typically only measures of condition involving body mass are examined with respect to song rate. Studies investigating a potential relationship between song rate and other indicators of condition, such as physiological measures of nutritional condition, are scant. In this study, we examined whether non-breeding song rate in male European starlings (Sturnus vulgaris) reflects plasma metabolite levels (high-density lipoproteins (HDL), albumin, triglycerides and cholesterol) and/or body mass. Song rate was significantly positively related to a principal component representing primarily HDL, albumin and cholesterol (and to a lesser degree plasma triglyceride levels). There was only a trend toward a significant positive correlation between song rate and body mass, and no significant correlation between body mass and the abovementioned principal component. Therefore, our results indicate that nutritional condition and body mass represent different aspects of condition, and that song rate reflects nutritional rather than body condition. Additionally, we also found that intra-individual song rate consistency (though not song rate itself) was significantly positively related to lutein levels, but not to body mass or nutritional condition. Together our results suggest that the relation between physiological measures of nutritional condition and song rate, as well as other signals, may present an interesting line of future research, both inside and outside the breeding season.  相似文献

According to a previously proposed coping model (De Waal [1989] Zoo Biol. 1(suppl):141–148), gregarious primates will react to crowding by adjusting their behavior in order to limit the amount of aggression displayed. Depending on the duration of the crowding, this can be achieved by the use of different strategies. Chimpanzees (Pan troglodytes) react to a medium‐term duration of spatial crowding by an active tension‐reduction mechanism whereby allogrooming markedly increases while aggression rises only slightly. Under comparable circumstances, a similar increase in grooming is found in bonobos (Pan paniscus). However, to test the coping model for bonobos, data on aggressive behavior during crowding are indispensable. This study provides such information by comparing the aggressive repertoire of a well‐established bonobo colony during a crowded period in winter with those that occurred during an uncrowded control period in summer. Given that during winter the aggression rate differed between food and nonfood contexts, we accounted for a potential effect of food context on aggression. During the crowded period, a significantly higher total frequency of aggression was found than during the control period. However, this increase was small in relation to the reduction in space when compared to similar experiments in other species. Together with observations of increased grooming under crowded conditions, our data confirm the hypothesis that bonobos cope with the increase in tension created by crowding (medium‐term duration) by applying a tension‐reduction strategy. Our data further show that although not all aggressive behaviors appear to be influenced by a high‐density condition, bonobos do not specifically limit their aggressive behaviors to mild, nonprovocative aggressions in a crowded environment. The inward rectifier current generated by Kir2.1 ion channel proteins is primarily responsible for the stable resting membrane potential in various excitable cell types, like neurons and myocytes. Tight regulation of Kir2.1 functioning prevents premature action potential formation and ensures optimal repolarization times. While Kir2.1 forward trafficking has been addressed in a number of studies, its degradation pathways are thus far unknown. Using three different lysosomal inhibitors, NH4Cl, chloroquine and leupeptin, we now demonstrate involvement of the lysosomal degradation pathway in Kir2.1 breakdown. Upon application of the inhibitors, increased steady state protein levels are detectable within few hours coinciding with intracellular granular Kir2.1 accumulation. Treatment for 24 h with either chloroquine or leupeptin results in increased plasmamembrane originating inward rectifier current densities, while current-voltage characteristics remain unaltered. We conclude that the lysosomal degradation pathway contributes to Kir2.1 mediated inward rectifier current regulation.  相似文献

Although heterospecific vocal imitation is well documented inpasserines, the evolutionary correlates of this phenomenon arepoorly known. Here, we studied interspecific variation in vocalmimicry in a comparative study of 241 European songbirds. Wetested whether vocal mimicry is a mode of repertoire acquisitionor whether it resulted from imperfect song learning. We alsoinvestigated the effect of the degree of contact with the vocalenvironment (with species having larger ranges, abundance, orbeing long lived having a higher degree of mimicry) and a possiblelink with cognitive capacity (an overall larger brain in specieswith mimicry). Finally, we determined the potential evolutionaryrole of vocal mimicry in different interspecific contexts, predictingthat mimicry may affect the intensity of brood parasitism, predation,or degree of hybridization. While controlling for research effortand phylogenetic relationships among taxa, we found that effectsizes for intersong interval, brain size, breeding dispersal,abundance, age-dependent expression of repertoires, and predationrisk reached a level that may indicate evolutionary importance.Vocal mimicry seems to be a consequence of song continuity ratherthan song complexity, may partially have some cognitive componentbut may also be dependent on the vocal environment, and mayattract the attention of predators. However, estimates of sexualselection and interspecific contacts due to brood parasitismand hybridization varied independently of vocal mimicry. Therefore,mimicry may have no function in female choice for complex songsand may be weakly selected via interspecific associations. Thesefindings provide little evidence for vocal mimicry having evolvedto serve important functions in most birds.  相似文献

Avian eggs contain substantial amounts of maternal yolk androgens, which have been shown to modulate offspring phenotype. The first studies on the functional consequences of maternal yolk androgens have focused on early life stages and their role in sibling competition. However, recent longitudinal studies reported long-lasting effects of maternal yolk androgens on offspring phenotype, mostly concerning traits that are sensitive to androgens. This suggests that maternal yolk androgens could play an important role in sexual selection, since the expression of many male sexual characters is testosterone-dependent. Using male canaries as a model, we examined the consequences of an experimental elevation of yolk testosterone concentrations on early development as well as long-lasting effects particularly on song, which is one of the most important sexual characters in male songbirds. Elevated yolk testosterone concentrations inhibited male growth, possibly in interaction with an existent ectoparasite exposure. Males hatched from testosterone-treated eggs (T-males) did not have enhanced competitive skills, in contrast to previous studies. The elevation of yolk testosterone concentrations delayed song development but did not affect adult song phenotype. This is intriguing, as yolk testosterone possibly induced developmental stress, which is known to reduce song quality. We hypothesize that yolk testosterone has either no direct effect on adult song phenotype, or that positive effects are merged by the negative effects of developmental stress. Finally, females mated with T-males invested more in their clutch indicating that females either assess T-males as more attractive (differential allocation hypothesis) or compensated for lower offspring viability (compensation hypothesis).  相似文献

In this study, we aimed to delineateevolutionarily significant units (ESUs) andmanagement units (MUs) for the Europeanbullhead in Flanders (Belgium). Therefore, wedetermined the genetic interrelationshipsbetween 11 bullhead populations, using lengthvariation at 7 polymorphic microsatellite lociand sequence variation in the d-loop of themitochondrial DNA (mtDNA). Despite therelatively small geographical scale of ourstudy, the analysis of the d-loop sequencesshows that the Flemish bullhead populationscontain 3 haplotype groups, which can beassigned to 3 previously described EuropeanmtDNA clades. Because of the importantdifferences between these clades, they may bedefined as evolutionarily significant units,which should be managed separately. Analysis ofmicrosatellite data reveals very high degreesof isolation between populations, with theexception of 3 pairwise comparisons whichinvolved adjacent populations. Our data suggestthat the 3 haplotype groups probably qualify asESUs, as they show phylogeographicdifferentiation for mtDNA variants as well assignificant divergence of allele frequencies atnuclear loci. However, one of these units,limited to a single population, may be ofCentral European origin. All populations of theScheldt basin meet the criteria for MUrecognition, since significantly differentmicrosatellite allele frequencies as well asprivate alleles are found. In contrast, geneticdifferentiation among the 3 populations of theMeuse basin is very low.  相似文献

Maternal hormones in bird eggs modulate not only offspring development, but recently it has also been shown that these effects can persist into adult life. A number of long-lasting effects concern traits of which the expression or development is modulated by androgens. This suggests that the nature of yolk hormone-mediated maternal effects may be organizational. Maternal androgens may therefore play an important role in sexual selection, since the expression of sexually selected male characters is often androgen-dependent.We experimentally manipulated the yolk androgen concentrations of European Starling (Sturnus vulgaris) eggs. Subsequently we followed 49 unrelated males from hatching until year of first reproduction. We investigated the expression of multiple sexually selected male characters (song, beak color and throat feather characteristics), taking into account whether a trait is androgen-dependent.Elevated levels of yolk androgens affected the length of the embryonic period, but did not modify the expression of either androgen-dependent or androgen-independent sexually selected male characters including song phenotype at adulthood. Thus the most important function of yolk androgens in starlings and possibly other bird species may relate to the early developmental period. The outcome of our study together with the results of our meta-analysis indicates that the effects of yolk androgens on sexually selected male characters may be comparatively small. Our results suggest that this may relate to the numerous other environmental and/or genetic factors influencing the expression of sexually selected male characters.  相似文献

In many temperate zone songbird species males only produce song during the breeding season, when plasma testosterone (T) levels are high. Males of some species sing throughout the year, even when T levels are low, indicating a dissociation between high T levels and song rate. Given that few studies have taken advantage of these species, we compare here song traits expressed under high versus low T concentrations and we study the role of testosterone in adult song learning in the European Starling, an open-ended learner in which repertoire size dramatically increases with age. We performed a detailed comparison of song complexity and song rate between fall and spring in 6-year-old intact male European starlings. In parallel, we investigated whether potential seasonal changes were regulated by the gonadally induced increase in plasma T, by comparing seasonal changes in intact and castrated males of the same age (castrated as juveniles during their first fall) and by subsequently experimentally elevating T in half of the castrated males. While song rate and stereotypy did not differ between intacts and castrates or between fall and spring, both groups increased their average song bout length from fall to spring, but only intact males increased their repertoire size, indicating that effects of seasonal T changes differ between song traits. Intact males overall displayed a larger song repertoire and a longer bout length than the castrates, and implantation with T caused a turnover in repertoire composition in castrates. However, as the castrates had never experienced high T levels and yet displayed a markedly higher repertoire size than that of typical yearling males, this suggests that the progressive increase of song repertoire with age in male starlings is not dependent on gonadal T, although it may be T-enhanced.  相似文献
