In a permanent cell line derived from Drosophila embryos, cytoplasmic actin is produced as an unstable precursor, which is subsequently converted to a stable form. This conversion results in a reduction in isoelectric point, with no apparent change in molecular weight. The conversion involves an enzymatic acetylation, and results in an insensitivity to aminopeptidase digestion, suggesting N-terminal blockage. Both the acetylated and unacetylated actins can participate in the assembly of F-actin, but with different efficiencies.This work was supported by a grant from the NIH (GM 22866).  相似文献

Correlations between mean age at smoltiftcation (MSA) of sea trout post-smolts and length of growing season are presented. The first year of the two-year smolt phase is the more important but the second is also significant as is the year of migration (the third). Strongest correlations are for MSA and the total number of growing days in all three years. A continuous record of calculated MSA from 1916–71 suggests there should have been relatively larger bursts of sea trout production in the 1940s and between 1955 and 1965. The number of trout migrating from one catchment correlates significantly with the number of growing days in the two-year smolt phase.  相似文献

This paper describes a mathematical model for the enzymatic hydrolysis and fermentation of cellulose by Trichoderma reesei. The principal features of the model are the assumption of two forms of cellulose (crystalline and amorphous), two sugars (cellobiose and glucose), and two enzymes (cellulase and β-glucosidase). An inducer–repressor–messenger RNA mechanism is used to predict enzyme formation, and pH effects are included. The model consists of 12 ordinary differential equations for 12 state variables and contains 38 parameters. The parameters were estimated from four sets of experimental data by optimization. The results appear satisfactory, and the computer programs permit simulation of a variety of system changes.  相似文献

Pulse-labeled oocyte proteins were found to have a maximum average half-life of 73 h. In general, larger peptides underwent degradation at a faster rate than smaller peptides. In this respect, oocytes are similar to most other cells. Microinjected 125I-labeled bovine serum albumin (BSA) was degraded over a 40 h period with a half-life of 20–30 h, regardless of the method of protein labeling, culture medium employed, size of oocyte microinjected, or hormonal history of the oocyte. The last two results, if applicable to oocyte proteins in general, imply that protein catabolism is constant throughout the later stages of oogenesis and that growth is primarily regulated by a stimulation of anabolism. Individual proteins microinjected into oocytes undergo rates of degradation consistent with turnover rates obtained in other systems. Sequestered 125I-labeled BSA is only partially (40%) degraded, which indicates that, unlike microinjected 125I-labeled BSA, it has access to a cytoplasmic compartment (yolk platelets?) within which it is relatively stable.  相似文献

Kresch (1977) presented the results of calculations which determined the cross-sectional shapes assumed by flexible, elastic tubes for varying transmural pressures. Extensions of these results are presented here in the form of graphs of the cross-sectional area as a function of the transmural pressure. Since the circumferential arc length, theX-axis intercept and theY-axis intercept were necessarily computed, and are of interest, graphs of these are also presented. Each graph is a composite of several curves. Each curve in the graph shows the behavior of the parameter as a function of the relative pressure (P) for a specific value ofR, a parameter related to the wall thickness. The curves are shown for both positive and negative values ofP. The most negative value ofP shown is the point where the walls touch and the most positive value shown is the point where the equations fail to converge. The cross sections were assumed to be elliptical when the transmural pressure was zero. Graphs for three of these initial shapes are shown: nearly circular, major-to-minor axis ratio equal to 2, and an intermediate case. For the nearly circular case, the curves divide into two regimes: one where stretching clearly predominates and another where bending clearly predominates. For the more eccentric initial cross-sections, the distinction between the two regimes cannot be made in this simple manner.  相似文献

We describe the N epsilon-acetimidylation of horse heart cytochrome c with retention of biological activity, the cleavage of the modified protein by CNBr, the separation of the fragments, and their further side-chain protection. We describe the manipulation of the amino acid sequences of the fragments by stepwise semisynthetic methods. We have prepared fragments corresponding to residues 66-78 and 66-79 of the protein, as well as the [Asp66] analogue of fragment 66-79. We have prepared the natural sequence and the [o-fluoro-Phe82] analogue of the fragment corresponding to residues 81-104 of the protein, and the [N epsilon-trifluoroacetyl-Lys79], the [N epsilon-dinitrophenyl-Lys79] and the [S-acetamidomethyl-Cys79] analogues of fragment 79-104, and the [N epsilon-Cbz-Lys81] analogue of fragment 80-104. We have coupled back the fragments of natural sequence to form a semisynthetic fragment corresponding to residues 66-104 of the protein. Modified fragments were also coupled to give analogues of the 66-104-residue sequence. In every case the homoserine residue representing methionine-80 was removed from the C-terminus of the 66-80-residue fragment and replaced by methionine on the N-terminus of the 81-104 residue fragment during the preparation of the fragments for coupling. The semisynthetic fragments are ready for specific deprotection and further coupling. We have coupled one such fragment to the (1-65)-peptide to produce semisynthetic [Hse65]cytochrome c. The product has satisfactory characteristics on chemical analysis, and on assay of its biological activity.  相似文献

The adhesive interaction between the ectosymbionts of the corn cob configuration (CCC), a naturally occurring bacterial consortium in human dental plaque, was studied. In vitro association was produced in mixed cultures consisting ofBacterionema matruchotii and streptococci resemblingStreptococcus sanguis previously isolated from human CCC. Phase-contrast, selective immunofluorescence, and scanning electron microscopy showed that in this aggregative system, a tuftlike polar structure typical of the coccal epibiont mediated binding to the core filament. This aggregative model provides evidence that a specialized structure on the cell surface of epiphytic organisms may participate in interbacterial adherence.  相似文献
