Ontogeny of the secondary fruit of navel orange (Citrus sinensis [L.] Osbeck) was studied using flower buds and fruit from groves in north central Florida. The secondary fruit, or navel, develops as a whorl of secondary-carpel primordia within the primary-carpel whorl when the flower bud is 1.5 to 2.0 mm in length. A complete secondary gynoecium with fused ovary but separate styles and stigmas develops within the primary gynoecium before anthesis. The stigma and style of secondary carpels are not as distinct as those of primary carpels. Three types of tissue protrusions from the secondary fruit into primary-fruit locules were abnormal placentae, free secondary carpels, and secondary-carpel outgrowths, the last being the most common. An abscission layer is present in the central axis of the primary fruit near the base of the secondary ovary. This layer is indistinguishable from adjacent tissues prior to commencement of secondary fruit abscission. Parenchyma cells in the abscission layer, however, contain large numbers of starch grains during abscission. Secondary and primary fruit have similar sigmoid growth patterns; however, onset of the cell-enlargement stage is approximately 2 wk later in secondary fruit. 相似文献

The generation of aroma compounds by carotenoid cleavage in the 9–10 position was studied, due to the importance of these compounds in the flavor and fragrance industry. The bioconversion of the carotenoid lutein to C13 norisoprenoids utilizing a microbial mixture composed of Trichosporon asahii and Paenibacillus amylolyticus was carried out by a fermentation process. Applying an experimental design methodology, the effects of nutritional factors on the production of aroma compounds present in the tobacco profile were studied. After an assessment of the significance of each nutritional factor, the levels of the variables yielding the maximum response were calculated. Glucose, tryptone, and yeast extract exerted a strong negative effect over the objective function, with glucose being the strongest. Lutein possessed a positive effect over the tobacco aroma production, while sodium chloride and trace elements showed no influence over the process. The yield attained after culture medium manipulation was almost ten-fold higher, compared with the base medium; and the aroma mixture was characterized as: 7,8-dihydro--ionol (95.2%), 7,8-dihydro--ionone (3.7%), and -ionone (1.1%). 相似文献

The effect of trehalose on folding and stability of the small ribosomal protein S6 was studied. Non-disruptive point mutations distributed along the protein structure were analyzed to characterize the stabilizing effect of trehalose and map the folding pathway of S6. On average, the stability of the wild-type and S6 mutants increases by 3 kcal/mol M trehalose. Despite the non-specific thermodynamic stabilization mechanism, trehalose particularly stabilizes the less destabilized mutants. Folding/unfolding kinetics shows clearly that trehalose induces the collapse of the unfolded state to an off-pathway intermediate with non-native diffuse contacts. This state is similar to the collapsed state induced by high concentrations of stabilizing salts, as previously reported. Although it leads to the accumulation of this off-pathway intermediate, trehalose does not change the compactness of the transition state ensemble. Furthermore, the productive folding pathway of S6 is not affected by trehalose as shown by a Phi-value analysis. The unfolded state ensemble of S6 should be more compact in the presence of trehalose and therefore destabilized due to decreased conformational entropy. Increased compaction of the unfolded state ensemble might also occur for more stable mutants of S6, thus explaining the synergistic effect of trehalose and point mutations on protein stabilization. 相似文献

Thyrotropin-releasing hormone is inactivated in the extracellular space by a membrane-bound peptidase, pyroglutamyl aminopeptidase II (PPII), a member of the M1 family of zinc metallopeptidases. The functional significance of multiple PPII RNA species expression is unknown. We detected, in rat tissues, a RNA species derived from an alternative processing at the exon 14-intron 14 boundary. The alternatively processed RNA encoded a shorter version of PPII (PPII*), lacking part of the C-terminal domain. PPII* was expressed in COS-7 (or C6 glioma) cells but it did not exhibit any PPII activity. Co-transfection of PPII and increasing amounts of PPII* expression vectors resulted in a dose-dependent reduction in PPII activity and the formation of covalent PPII-PPII* heterodimers. PPII* is therefore a powerful dominant-negative isoform of PPII, and heterodimerization may be its mechanism of action. Natural expression of shortened versions of M1 aminopeptidases may constitute a new mode of regulation of their activity. 相似文献

One of the fundamental problems of cell biology is the understanding of complex regulatory networks. Such networks are ubiquitous in cells and knowledge of their properties is essential for the understanding of cellular behavior. In earlier work (Kholodenko et al. (PNAS 99: 12841), it was shown how the structure of biological networks can be quantified from experimental measurements of steady-state concentrations of key intermediates as a result of perturbations using a simple algorithm called "unravelling". Here, we study the effect of experimental uncertainty on the accuracy of the inferred structure (i.e. whether interactions are excitatory or inhibitory) of the networks determined using the unravelling algorithm. We show that the accuracy of the network structure depends not only on the noise level but on the strength of the interactions within the network. In particular, both very small and very large values of the connection strengths lead to large uncertainty in the inferred network. We describe a powerful geometric tool for the intuitive understanding of the effect of experimental error on the qualitative accuracy of the inferred network. In addition, we show that the use of additional data beyond that needed to minimally constrain the network not only improves the accuracy of the inferred network, but also may allow the detection of situations in which the initial assumptions of unravelling with respect to the network and the perturbations have been violated. In this study it was compared the MAS-100 and the Andersen air samplers' performances and a similar trend in both instruments was observed. It was also evaluated the microbial contamination levels in 3060 samples of offices, hospitals, industries, and shopping centers, in the period of 1998 to 2002, in Rio de Janeiro city. Considering each environment, 94.3 to 99.4% of the samples were the allowed limit in Brazil (750 CFU/m3). The industries' results showed more important similarity among fungi and total heterotrophs distributions, with the majority of the results between zero and 100 CFU/m3. The offices' results showed dispersion around 300 CFU/m3. The hospitals' results presented the same trend, with an average of 200 CFU/m3. Shopping centers' environments showed an average of 300 CFU/m3 for fungi, but presented a larger dispersion pattern for the total heterotrophs, with the highest average (1000 CFU/m3). It was also investigated the correlation of the sampling period with the number of airborne microorganisms and with the environmental parameters (temperature and air humidity) through the principal components analysis. All indoor air samples distributions were very similar. The temperature and air humidity had no significant influence on the samples dispersion patterns. 相似文献

We report the effect of Stryphnodendron adstringens on the trypanosomatid Herpetomonas samuelpessoai. The parasites were grown at 28 degrees C in a chemically defined medium containing crude extract and fractions at concentrations from 100 to 5000 microg/ml obtained from S. adstringens. Concentrations of 500, 1000, 2500, and 5000 microg/ml both crude extract and semi-purified fraction progressively inhibited the protozoans' growth. At a concentration of 100 microg/ml, crude extract or a semi-purified (F3) fraction did not affect the growth of the protozoans. The F3-9 - F3-12 sub-fractions, at a concentration of 1000 microg/ml, also showed increased inhibitory activity on H. samuelpessoai. The IC50 of the crude extract and the F3 fraction were 538 and 634 microg/ml, respectively. Ultrastructural and enzymatic alterations in the trypanosomatids were also evaluated. H. samuelpessoai cultivated in the presence of IC50 crude extract showed considerable ultrastructural alterations, such as marked mitochondrial swelling with a large number of cristae and evident Golgi complex vesiculation, as observed by transmission electron microscopy. Cells exposed to 538 microg/ml of crude extract at 28 degrees C for 72 h, showed decreased activity of the enzyme succinate cytochrome c reductase, a typical mitochondrion marker, as compared to untreated cells. 相似文献
