BACKGROUND AND AIMS: Lotus japonicus 'Gifu' develops multiple axillary shoots in the cotyledonary node region throughout the growth of the plant. The origin, initiation and development of these axillary meristems were investigated. METHODS: Morphological, histological and mRNA in situ analyses were done to characterize the ontogeny of cotyledonary axillary shoot meristems in Lotus. Morphological characterization of a putative Lotus shoot branching mutant (super-accessory branches) sac, is presented. KEY RESULTS: By using expression of an L. japonicus STM-like gene as a marker for meristematic tissues, it was demonstrated that groups of cells maintained in the meristematic state at the cotyledonary axil region coincide with the sites where additional axillary meristems (accessory meristems) form. A Lotus shoot branching mutant, sac, is a putative Lotus branching mutant characterized by increased proliferation of accessory shoots in all leaf axils including the cotyledons. CONCLUSION: In Lotus, axillary shoot meristems continually develop at the cotyledonary node region throughout the growth of the plant. These cotyledonary primary and accessory axillaries arise from the position of a meristematic zone of tissue at the cotyledonary node axil region.  相似文献

Chromoblastomycosis is a chronic and progressive deep mycosis that is usually found in tropical and subtropical areas. Fonsecaea pedrosoi is considered its most frequent etiologic agent and causes a typical granulomatous inflammatory response, whose degree reflects the immune status of the host. Since macrophages play a fundamental role in the control of the infection, this study aimed at investigating the production of oxygen reactive specimens, the phagocytic capacity and the production of nitric oxide (NO) by macrophages employing in vitro assays and an in vivo model of chromoblastomycosis. Our results demonstrated that, during the infection, peritoneal macrophages show an increased phagocytic capacity and H2O2 production, but also a reduced ability to produce NO. Moreover, F. pedrosoi stimulated H2O2 production in vitro but not the synthesis of NO. The incubation of IFNγ and LPS-stimulated macrophages with melanin, obtained from the fungus, inhibited NO production. Examination of the liver and spleen of infected animals, at day 30 or 60 following inoculation, showed a progressive increase in the number and size of granulomas, indicating that macrophages are properly mobilized and activated. Our data suggest that the inability of the host to clear F. pedrosoi, leading to a chronic disease, is due, at least in part, to the inhibition of NO synthesis by macrophages by fungus-produced melanin.  相似文献

We study a single species in a chemostat, limited by two nutrients, and separate nutrient uptake from growth. For a broad class of uptake and growth functions it is proved that a nontrivial equilibrium may exist. Aim The purpose of the study was to assess the degree of floristic differentiation between tropical dry forest (TDF) and savanna occurring in a single landscape. This comparison provides information on the responses of vegetation to the prevailing environmental conditions, while it also allows us to make inferences about large‐scale events and processes, both biogeographical and evolutionary. Our approach included three levels of analysis: (1) taxonomic, (2) morphological and (3) vegetational. Location The seasonal dry tropical landscape in the Nizanda region, Oaxaca State, southern Mexico. The landscape comprises a complex vegetation mosaic in which tropical dry forest and savannas are the most conspicuous components. Methods Comparisons between TDF and savanna were based on inventories for these communities produced after 8 years of botanical survey. At the taxonomic level, the relative representation of taxa of different hierarchical levels in each community was examined. Morphological analyses required the classification of species on each of three criteria: (1) growth form, (2) life form and (3) growth habit. Vegetation level analysis was based on the frequencies of taxa in one hundred 100‐m2 composition plots with which matrices of binary data were constructed for species, genera and families. These were subjected to classification analysis with Ward's method and using Euclidean distances as the dissimilarity algorithm. Results The combined flora for both communities comprised 600 species, 375 genera and 94 families; between them they shared 31, 40 and 34 taxa, respectively. The corresponding Sørensen similarity values were 10%, 21% and 72%, respectively. Ranking genera and families according to their species richness displayed large differences between savanna and TDF. Large differences between these communities were observed for Acanthaceae, Cactaceae, Euphorbiaceae and Mimosaceae, whereas Fabaceae and Asteraceae had similar high ranks according to the species richness in the two systems. The growth form spectrum diverged between the two communities, with TDF having more trees, shrubs and climbers. Savanna was characterised by forbs and graminoid herbs. Growth habit spectra revealed a clear dominance of herbaceous and suffruticose plants in savanna, and of woody elements and epiphytes in TDF. Regarding Raunkiaer's life forms, savanna had relatively more hemicryptophytes, and TDF more phanerophytes. Classification analyses showed that savanna and TDF forest samples kept their identities, regardless of taxonomic level (species, genera and families) at which the analyses were performed. Main conclusions The TDF and savanna of Nizanda represent two floristic systems with a large degree of differentiation at all taxonomic levels and patterns of morphological attributes. This suggest that the two floristic sets have evolved independently for extended periods of time, despite their close proximity. One important implication of this floristic differentiation is the large joint contribution made by these communities to the regional flora.  相似文献

The feeding habits of four species of mesopelagic fishes from northern Chile are described: Triphoturus mexicanus, Diogenichtys atlanticus, Vinciguerria lucetia and Cyclothone acclinidens. Samples were captured in September 1988 between 18 degrees 25' and 19 degrees 09'S in the South West Pacific. The quantitative and qualitative analyses of the stomach content showed that the species have a zooplanktophagous opportunistic behavior, mainly predating on Crustaceans, especially on Copepods. The evaluation of the trophic spectrum and diversity of T. mexicanus and C. acclinidens suggests that these are nictoepipelagic species, while D. atlanticus and V. lucetia would be typical mesopelagic.  相似文献

In this work, we estimate the contributions of the different sources of N incorporated by two N2-fixing cyanobacterial blooms (Anabaena sp. and Microchaete sp.) in the rice fields of Valencia (Spain) during the crop cycles of 1999 and 2000, and evaluate the response of nitrogenase and C assimilation activities to changing irradiances. Our results show that, far from the generally assumed idea that the largest part of the N incorporated by N2-fixing cyanobacterial blooms in rice fields comes from N2 fixation, both cyanobacterial blooms incorporated about three times more N from dissolved combined compounds than from N2 fixation (only about 33–41% of the N incorporated came from N2 fixation). Our results on the photodependence of C and N2 fixation indicate that in both cyanobacterial blooms, N2 fixation showed a steeper initial slope (α) and was saturated with less irradiance than C fixation, suggesting that N2 fixation was more efficient than photosynthesis under conditions of light limitation. At saturating light, N2 fixation and C fixation differed depending on the bloom and on the environmental conditions created by rice plant growth. Carbon assimilation but not nitrogenase activity appeared photoinhibited in the Anabaena but not in the Microchaete bloom in August 1999, when the plants were tall and the canopy was important, and there was no limitation of dissolved inorganic carbon. The opposite was found in the Microchaete bloom of June 2000, when plants were small and produced little shade, and dissolved inorganic carbon was very low.  相似文献

We show here that the paaABCDE genes of the paa cluster responsible for phenylacetate degradation in Escherichia coli W encode a five-component oxygenase that hydroxylates phenylacetyl-coenzyme A (CoA), the first intermediate of the pathway. The primary structure of the subunits of bacterial phenylacetyl-CoA oxygenases revealed that these enzymes constitute the prototype of a new and distinct group of the large bacterial diiron multicomponent oxygenase family.  相似文献

There is increasing evidence that mitochondria play an important role in the control of cytosolic Ca2+ signaling. We show here that the main mitochondrial Ca2+-exit pathway, the mitochondrial Na+/Ca2+ exchanger, controls the pattern of cytosolic Ca2+ oscillations in non-excitable cells. In HeLa cells, the inhibitor of the mitochondrial Na+/Ca2+ exchanger CGP37157 changed the pattern of the oscillations induced by histamine from a high-frequency irregular one to a lower frequency baseline spike type, surprisingly with little changes in the average Ca2+ values of a large cell population. In human fibroblasts, CGP37157 increased the frequency of the baseline oscillations in cells having spontaneous activity and induced the generation of oscillations in cells without spontaneous activity. This effect was dose-dependent, disappeared when the inhibitor was washed out and was not mimicked by mitochondrial depolarization. CGP37157 increased mitochondrial [Ca2+] and ATP production in histamine-stimulated HeLa cells, but the effect on ATP production was only transient. CGP37157 also activated histamine-induced Ca2+ release from the endoplasmic reticulum and increased the size of the cytosolic Ca2+ peak induced by histamine in HeLa cells. Our results suggest that the mitochondrial Na+/Ca2+ exchanger directly modulates inositol 1,4,5-trisphosphate-induced Ca2+ release and in that way controls cytosolic Ca2+ oscillations.  相似文献
