A systematic investigation of the S3 sub-pocket activity requirements was conducted. It was observed that linear and sterically small side chain substituents are preferred in the S3 sub-pocket for optimal renin inhibition. Polar groups in the S3-sub-pocket were not well tolerated and caused a reduction in renin inhibitory activity. Further, compounds with clog P's < or = 3 demonstrated a dramatic reduction in CYP3A4 inhibitory activity.  相似文献

Herein, we report on the identification of three potent glycine and related amino acid-based series of FXa inhibitors containing a neutral P1 chlorophenyl pharmacophore. A X-ray crystal structure has shown that constrained glycine derivatives with optimized N-substitution can greatly increase hydrophobic interactions in the FXa active site. Also, the substitution of a pyridone ring for a phenylsulfone ring in the P4 sidechain resulted in an inhibitor with enhanced oral bioavailability.  相似文献

Millepora species are conspicuous members of shallow coral reefs where they occupy a variety of substrata and produce morphologically complex skeletons. This study focuses on the roles of growth on vertical and horizontal surfaces and the production of encrusting bases and branches (a “sheet-tree” morphology) for the success of the Millepora alcicornis on coral reefs. The effects of inclination were investigated by comparing the size and growth rates of M. alcicornis on vertical and horizontal surfaces at 3–5 m depth, in St. John, US Virgin Islands. The consequences of morphological complexity were investigated by comparing polyp density, chlorophyll content and biomass between encrusting bases and branches; the role of branches in asexual reproduction was also quantified. Colonies on vertical surfaces had larger encrusting bases, longer perimeters and lower densities of branches compared to those on horizontal surfaces. Growth rates also varied significantly between surfaces, largely because colonies on horizontal surfaces shrank in area while those on vertical surfaces increased in area, albeit slowly. Branches were not specialized in comparison to encrusting bases in terms of the density of dactylozooids and gastrozooids, chlorophyll content and biomass, but they were effective asexual propagules. During one storm, 79% of the branches were removed from colonies of M. alcicornis, and 4% attached to the substratum to produce new colonies at a density of ≈0.5 colonies.m-2. Anecdotal observations suggest that such storms rarely damaged encrusting bases on vertical surfaces, but often destroyed those on horizontal surfaces. Thus, the encrusting bases on vertical surfaces are likely to be large because of greater age rather than faster growth, while those on horizontal surfaces are likely to be small because they are relatively young and short lived. These findings suggest that the success of M. alcicornis is a result, in part, of the beneficial consequences of their “sheet-tree” morphology, that supports: (a) slow growth and resistance to wave damage of encrusting bases on vertical surfaces, and (b) the use of branches as asexual propagules. Nitrate in the unsaturated zone between the soil surface and the water table was studied in agroforestry Parklands in north western Senegal by examination of samples obtained by hand auger. Depending on location, water tables existed at depths between 10 and 35m below ground. Previous studies of groundwater in this region had found that large concentrations of nitrate were unconnected with anthropogenic activity. The objective of this study was to determine whether nitrogen fixing vegetation had a role in groundwater nitrate accumulation and whether roots of trees were located deeply enough to access the nitrate. Accordingly, sample profiles were augered close to stems of nitrogen fixing trees, non-nitrogen fixing trees and also in adjacent areas that were unaffected by tree presence. These adjacent areas were typically open pasture or cultivated fields. Tree fine roots were quantified in the samples and examined for the presence of mycorrhizas. Similarly, sand/soil samples were examined and tested for the presence of nitrogen fixing rhizobia that were capable of forming functional nodules on appropriate host plants. Concentrations of nitrate were greatest in soils beneath nitrogen fixing trees and nitrate was more plentiful in profiles augered beneath nitrogen fixing crops than it was elsewhere suggesting that N-fixation was the source of the nitrate. The concentrrations of nitrate that were found in the unsaturated zone were greatly in excess of the WHO recommended limit for nitrate in drinking water. High NO3-N/Cl ratios confirm insitu production of nitrate, and indicate that this is a natural baseline occurrence related to N-fixation. The nitrate is moving down the profile and impacts the groundwater unless it can be intercepted by plant roots. NO3-N amounts in solution in the soil profile varied between 75 and 1000kg ha–1 beneath trees and between 120 and 400kg ha–1 in areas outwith tree crowns. Although these quantities of N occupy the lower end of the range of N values obtained in north American deserts, they comprise a considerable dryland resource where amounts of organic fertilizer are limited and where cost prohibits the use of commercial fertilizers. Roots of both nitrogen fixing and non-nitrogen fixing trees were deep enough to access the nitrate but the small amounts of available water at intermediate depths suggest that large scale uptake of nitrate will only be possible in the wetter zones located close to the water table. Shallow roots tended to be more heavily colonized by mycorrhizas than deeper roots but mycorrhizas were recovered from roots located 22m below ground. Tree roots and rhizobia had similar patterns of distribution. They were commonest close to the soil surface, less frequent at intermediate depths and tended to increase in frequency close to the water table.  相似文献

Recently, trans-disubstituted oxo-aryl-piperidines have been identified as small molecule nonpeptide renin inhibitors for the modulation of hypertension. Herein, we report on the discovery and preparation of a new class of novel cis-disubstituted amino-aryl-piperidines as a mixture of enantiomers that are potent in vitro renin inhibitors and also, possess in vivo antihypertensive activity in a double transgenic mouse model.  相似文献

Pompe's disease is caused by a deficiency of the lysosomal enzyme acid alpha-glucosidase (GAA). GAA is synthesized as a 110-kDa precursor containing N-linked carbohydrates modified with mannose 6-phosphate groups. Following trafficking to the lysosome, presumably via the mannose 6-phosphate receptor, the 110-kDa precursor undergoes a series of complex proteolytic and N-glycan processing events, yielding major species of 76 and 70 kDa. During a detailed characterization of human placental and recombinant human GAA, we found that the peptides released during proteolytic processing remained tightly associated with the major species. The 76-kDa form (amino acids (aa) 122-782) of GAA is associated with peptides of 3.9 kDa (aa 78-113) and 19.4 kDa (aa 792-952). The 70-kDa form (aa 204-782) contains the 3.9- and 19.4-kDa peptide species as well as a 10.3-kDa species (aa 122-199). A similar set of proteolytic fragments has been identified in hamster GAA, suggesting that the multicomponent character is a general phenomenon. Rabbit anti-peptide antibodies have been generated against sequences in the proteolytic fragments and used to demonstrate the time course of uptake and processing of the recombinant GAA precursor in Pompe's disease fibroblasts. The results indicate that the observed fragments are produced intracellularly in the lysosome and not as a result of nonspecific proteolysis during purification. These data demonstrate that the mature forms of GAA characterized by polypeptides of 76 or 70 kDa are in fact larger molecular mass multicomponent enzyme complexes.  相似文献

After several decades of disturbance, many coral reefs in the Caribbean are dominated by macroalgae. One process affecting this transition is coral-macroalgal competition, yet few studies have addressed the mechanisms involved. In this study, we investigated competition between the tall and bushy macroalga Sargassum hystrix (J. Agardh) and the branching coral Porites porites (Pallas) on a shallow reef in Jamaica. Experiments were designed to expose coral branches to different treatments to test the role of shading and abrasion by Sargassum on coral growth and polyp expansion. Corals exposed to Sargassum grew significantly more slowly (80% reduction) than controls, but this effect was absent when corals were caged to prevent physical contact with macroalgae. Light levels were reduced in both the algal and cage treatments, but shading apparently had little effect on the growth of corals in cages. Short-term measurements of integrated net water flow did not detect variation among treatments. In algal-mimic treatments, where clear plastic strips could touch but not shade the corals, growth rates were 25% lower than controls, but this effect was not statistically significant. Thus, the growth of corals in contact with Sargassum was reduced by abrasion and, to a lesser extent, by factors unique to living macroalgae. Analysis of polyp expansion showed that polyps were more frequently retracted when corals were in contact with macroalgae or algal-mimics compared to controls or cage treatment; the frequency of polyp contraction was correlated positively with growth. Together, these results suggest that abrasion-mediated polyp retraction is one of the primary mechanisms of competition utilized by tall (ca. 17 cm) macroalgae against scleractinian corals.  相似文献

Although most physiological traits scale allometrically in unitary organisms, it has been hypothesized that modularity allows for isometric scaling in colonial modular taxa. Isometry would allow increases in size without functional constraints, and is thought to be of central importance to the success of a modular design. Yet, despite its potential importance, scaling in these organisms has received little attention. To determine whether scleractinian corals are free of allometric constraints, we quantified metabolic scaling, measured as aerobic respiration, in small colonies (< or =40 mm in diam.) of the scleractinian Siderastrea siderea. We also quantified the scaling of colony surface area with biomass, since the proposed isometry is contingent upon maintaining a constant ratio of surface area to biomass (or volume) with size. Contrary to the predicted isometry, aerobic respiration scaled allometrically on biomass with a slope (b) of 0.176, and colony surface area scaled allometrically on biomass with a slope of 0.730. These findings indicate that small colonies of S. siderea have disproportionately high metabolic rates and SA:B ratios compared to their larger counterparts. The most probable explanations for the allometric scaling of aerobic respiration are (1) a decline in the SA:B ratio with size such that more surface area is available per unit of biomass for mass transfer in the smallest colonies, and (2) the small size, young age, and disproportionately high growth rates of the corals examined. This allometric scaling also demonstrates that modularity, alone, does not allow small colonies of S. siderea to overcome allometric constraints. Further studies are required to determine whether allometric scaling is characteristic of the full size range of colonies of S. siderea.  相似文献
