BackgroundCrimean-Congo haemorrhagic fever virus (CCHFV) is a highly pathogenic virus for which a safe and effective vaccine is not yet available, despite being considered a priority emerging pathogen. Understanding transmission patterns and the use of potential effective vaccines are central elements of the future plan against this infection.MethodsWe developed a series of models of transmission amongst livestock, and spillover infection into humans. We use real-world human and animal data from a CCHFV endemic area in Afghanistan (Herat) to calibrate our models. We assess the value of environmental drivers as proxy indicators of vector activity, and select the best model using deviance information criteria. Finally we assess the impact of vaccination by simulating campaigns targeted to humans or livestock, and to high-risk subpopulations (i.e, farmers).FindingsSaturation deficit is the indicator that better explains tick activity trends in Herat. Recent increments in reported CCHFV cases in this area are more likely explained by increased surveillance capacity instead of changes in the background transmission dynamics. Modelling suggests that clinical cases only represent 31% (95% CrI 28%-33%) of total infections in this area. Vaccination campaigns targeting humans would result in a much larger impact than livestock vaccination (266 vs 31 clinical cases averted respectively) and a more efficient option when assessed in courses per case averted (35 vs 431 respectively). Targeted vaccination of farmers is impactful and more efficient, resulting in 19 courses per case averted (95% CrI 7–62) compared to targeting the general population (35 courses 95% CrI 16–107)ConclusionsCCHFV is endemic in Herat, and transmission cycles are well predicted by environmental drivers like saturation deficit. Vaccinating humans is likely to be more efficient and impactful than animals, and importantly targeted interventions to high risk groups like farmers can offer a more efficient approach to vaccine roll-out.  相似文献

Interest in therapeutic kinase inhibitors continues to grow beyond success in oncology. To date, ATP-mimetic kinase inhibitors have focused primarily on monocyclic and bicyclic heterocyclic cores. We sought to expand on the repertoire of potential cores for kinase inhibition by exploring tricyclic variants of classical bicyclic hinge binding motifs such as pyrrolopyridine and pyrrolopyrazine. Herein we describe the syntheses of eight alternative tricyclic cores as well as in vitro screening results for representative kinases of potential therapeutic interest.  相似文献

For many marine invertebrates, the maximum size of an individual is influenced heavily by environmental factors and may be limited by energetic constraints. In this study, an energetic model developed originally for anemones was applied to the free-living scleractinian Fungia concinna (Verrill) from Moorea (French Polynesia) to test the hypothesis that energetic constraints limit the size of this solitary coral. The modified model assumed that photosynthesis was the primary source of metabolic energy, and that metabolic costs were represented by aerobic respiration; these sources and sinks of energy were compared using daily energy budgets that were analyzed using double logarithmic regressions of energy against coral size. With this approach, energy limitation is characterized by a scaling exponent for energetic cost (bcost) that is larger than the scaling exponent for energy intake (bintake). For the size range of F. concinna studied, bintake = 0.73 ± 0.09 and bcost = 0.46 ± 0.10, thereby demonstrating that large individuals accumulated an energetic surplus, even when the expenditure associated with host tissue and symbiont growth was included in the model. The surplus of energy that this coral acquires as it grows appears to be driven by the scaling of traits associated functionally with the scaling of respiration and photosynthesis. Specifically, tissue biomass displayed a strong positive allometry with respect to surface area (i.e., b > 1), and this constraint on surface area may be the mechanistic basis of the low scaling exponent for metabolic cost. In contrast, the capacity for autotrophy - defined indirectly as Symbiodinium population density and chlorophyll content - increased isometrically with surface area, and likely contributed to the higher scaling exponent for intake relative to cost. Our results suggest that growth in F. concinna is not limited strictly by energy, but instead maximum size must be determined by alternative physiological or ecological constraints.  相似文献

During eukaryotic cell division, the sister chromatids of duplicated chromosomes are pulled apart by microtubules, which connect via kinetochores. The kinetochore is a multiprotein structure that links centromeres to microtubules, and that emits molecular signals in order to safeguard the equal distribution of duplicated chromosomes over daughter cells. Although microtubule‐mediated chromosome segregation is evolutionary conserved, kinetochore compositions seem to have diverged. To systematically inventory kinetochore diversity and to reconstruct its evolution, we determined orthologs of 70 kinetochore proteins in 90 phylogenetically diverse eukaryotes. The resulting ortholog sets imply that the last eukaryotic common ancestor (LECA) possessed a complex kinetochore and highlight that current‐day kinetochores differ substantially. These kinetochores diverged through gene loss, duplication, and, less frequently, invention and displacement. Various kinetochore components co‐evolved with one another, albeit in different manners. These co‐evolutionary patterns improve our understanding of kinetochore function and evolution, which we illustrated with the RZZ complex, TRIP13, the MCC, and some nuclear pore proteins. The extensive diversity of kinetochore compositions in eukaryotes poses numerous questions regarding evolutionary flexibility of essential cellular functions.  相似文献

ObjectivesTo assess the cost effectiveness of a meningococcal serogroup C conjugate vaccination campaign in 0-17 year olds.DesignCost effectiveness analysis from the perspective of the healthcare provider.SettingEngland and Wales.ResultsIn 1998-9, immediately before the introduction of meningococcal C vaccination, the burden of serogroup C disease was considerable, with an estimated 1137 cases in people aged 0-17 years and at least 72 deaths. The vaccination campaign is estimated to have cost between £126m ($180m, €207m) and £241m ($343m, €395m), depending on the price of the vaccine. Under base case assumptions the cost per life year saved from the vaccination campaign is estimated to be £6259. School based vaccination was more cost effective than general practice based vaccination because of lower delivery costs. Immunisation of infants aged under 1 year was the least cost effective component of the campaign because, although this maximises the life years gained, the three dose schedule required is more expensive than other methods of delivery. Estimates of the cost per life year saved were sensitive to assumptions on the future incidence of disease and the case fatality ratio.ConclusionsMeningococcal C vaccination is likely to be more cost effective in all age groups when the incidence of disease is high. It is also more cost effective when given to children aged 1-4 (by general practitioners) and to children and young people aged 5-17 years at school than when administered to infants under 12 months of age or young people aged 16-17 years who are not at school.

Although mixing patterns are thought to be important determinants of the spread of airborne infectious diseases, to our knowledge, there have been no attempts to directly quantify them for humans. We report on a preliminary study to identify such mixing patterns. A sample of 92 adults were asked to detail the individuals with whom they had conversed over the period of one, randomly assigned, day. Sixty-five (71%) completed the questionnaire, providing their age, the age of their contacts and the social context in which the contacts took place. The data were analysed using multilevel modelling. The study identified, and allowed the quantification of, contact patterns within this sample that may be of epidemiological significance. For example, the degree of assortativeness of mixing with respect to age was dependent not only on the age of participants but the number of contacts made. Estimates of the relative magnitude of contact rates between different social settings were made, with implications for outbreak potential. Simple questionnaire modifications are suggested which would yield information on the structure and dynamics of social networks and the intensity of contacts. Surveys of this nature may enable the quantification of who acquires infection from whom and from where.  相似文献

An energy budget was determined for the coral Porites porites living in a stressed environment for comparison of the energy inputs and expenditure with those of the same species living in an adjacent clear water fore reef environment. The stressed site was characterised by higher sedimentation and lower irradiances than at the fore reef site. Zooplankton ingestion was found to be an insignificant component of the energy intake: the coral is fully autotrophic under stress conditions. The integrated 24 h rate of photosynthetic energy production on a clear sunny day was 20% higher for stressed corals compared to fore reef corals. This was largely the result of photoadaptation which resulted in increased values for and decreased values for Ik in the hyperbolic tangent function equation for the photosynthesis versus irradiance curve. The energy investment in growth of animal tissue was lower in stressed corals. The percentage translocation of photosynthase to the animal tissue remained at about 78%, but the respiration rate of the animal tissue was reduced by 3 fold. These data combined with the high rate of photosynthetic production predict a net daily energy surplus of 67% in stressed corals compared with the 45% surplus in unstressed corals. Scope for growth is reduced under stress conditions.  相似文献
