Interest in a single gene etiology for obesity, as assessed by the body mass index (BMI), has been spurred recently by reports of a putative recessive major gene for extreme values, which accounts for as much as 40% of the variance. The major gene hypothesis was evaluated here in the Québec Family Study, a random sample of 375 French-Canadian volunteer families. This report represents one component in a more complete investigation of obesity in these families. In contrast to the recent studies, a major gene hypothesis for BMI was not verified here. Although there was a major effect, it did not conform to a Mendelian pattern of transmission. A multifactorial component (i.e., polygenic and/or common environmental factors) accounted for 42% of the phenotypic variance. In addition, evidence of heterogeneity between the generations was found. The heterogeneity was traced to the major non-Mendelian component (which accounted for 0.01% of the variance in parents and over 40% in offspring) rather than to the multifactorial one. These results would suggest that a simple recessive gene mixed model may not be sufficient to explain the familial distribution of the BMI. Several factors which may have contributed to these results include temporal trends and surrogate effects such as those related to variation in body composition and energy balance components. The globin derived from the monomer Component IV hemoglobin of the marine annelid,Glycera dibranchiata, has been completely sequenced, and the resulting information has been used to create a structural model of the protein. The most important result is that the consensus sequence of Component IV differs by 3 amino acids from a cDNA-predicted amino acid sequence thought earlier to encode the Component IV hemoglobin. This work reveals that the histidine (E7), typical of most heme-containing globins, is replaced by leucine in Component IV. Also significant is that this sequence is not identical to any of the previously reportedGlycera dibranchiata monomer hemoglobin sequences, including the sequence from a previously reported crystal structure, but has high identity to all. A three-dimensional structual model for monomer Component IV hemoglobin was constructed using the published 1.5 å crystal structure of a monomer hemoglobin fromGlycera dibranchiata as a template. The model shows several interesting features: (1) a Phe31 (B10) that is positioned in the active site; (2) a His39 occurs in an interhelical region occupied by Pro in 98.2% of reported globin sequences; and (3) a Met41 is found at a position that emerges from this work as a previously unrecognized heme contact. 相似文献

Polyacrylamide gel electrophoresis was used to investigate the seed proteins of 36 accessions belonging toSolanum sect.Solanum (Solanaceae). These accessions represented 20 species, of four differing ploidy levels, and included infraspecific morphological variants. The resultant band patterns tended to reflect the morphological differences and genetical isolation displayed by many of the species. The most variable band patterns were encountered in the taxa with the greatest infraspecific variation, while many of the more morpho-genetically distinct taxa seemed to have species-specific band patterns.Good matches were found between the band patterns of artificial hybrids, those of their known parents, and mixtures of the parental protein extracts. This illustrates the potential use of such a technique for pinpointing possible genome donors of natural hybrids, and especially of polyploids. A chemically synthesized branched tetranucleotide, G3'p5'A [2'p5'G]3'p5'C corresponding to the consensus sequence at the branch point in introns undergoing RNA splicing, was used as a hapten to elicit antibranch antibodies. Binding assays with 32P-labeled hapten and unlabeled structurally related haptens indicated that the antibodies are highly specific for the branch structure and have some specificity for the A2'p5'G sequence at the branch point, but have essentially none for a variety of other 2'p5' or 3'p5' dinucleotides or for the linear trinucleotide G3'p5'A3'p5'C. Eggshell thickness is often expressed by means of an index based on the length, breadth and weight of the shell. The effect of the blow-hole and egg eccentricity on Ratcliffe's shell thickness index was investigated in a sample of 585 eggs from six raptor species. Corrections for the size of the hole and egg eccentricity are proposed, as is a combined formula to correct both sources of error at the same time. It is shown that by using these formulae, considerable improvements in estimates of shell thinning are obtained. To examine the interactions between age and photoperiod on reproduction and spleen weights, we exposed adult male and female rice rats of various ages to photoperiods of 16:8-h light-dark photoperiods (16L:8D) or 12L:12D. After 10 wk, animals were killed and the following data were recorded: weights of testes, seminal vesicles, uterus, ovaries, body, and spleen and, in addition, vaginal patency. Young adult males displayed a greater degree of testicular and seminal vesicle regression in short photoperiods than did older males; the testes of most older males did not regress in response to short photoperiods. Spleen weight was unresponsive to short photoperiods in all males, but was affected by age. Females, however, exhibited reproductive organ regression and decreased vaginal patency in response to short photoperiods at all ages examined. Body weights were affected by photoperiod in young females, and, as in males, photoperiod had no effect on spleen weights. These data suggest that the reproductive response to photoperiod in adult male rice rats declines with age, whereas in adult females it does not. 相似文献
