Predation on ringed seals ( Phoca hispida ) was examined in Barrow Strait between March and May 1984 to 1986. Polar bears were the most important predator. Evidence of bear predation was observed at 18–30% of the ringed seal subnivean structures we located. Ten to 24% of predation attempts were successful, with pups making up 75% to 100% of the seals killed. Bears killed an average of 0.08 to 0.51 seals/km2, which comprised 8 to 44% of the estimated annual pup production. Bears were successful on average in 11.3% of their attempts to kill pups hidden inside birth lairs. On southeast Baffin Island where snow was soft and pups were exposed, bears were successful in 33.5% of their attempts to kill a seal. Negative correlations were found between mean snow depth and predation by polar bears ( r = -0.896, P = 0.04, n = 5) in 1985, and between snow depth and the number of predation attempts ( r = -0.613, P = 0.02, n = 14) in 1986.  相似文献

Two multipotential embryonal carcinoma (EC) cell lines, 1003 and 1009, can be induced to form preferentially neural derivatives in vitro. Synthesis of specific proteins during neural differentiation was followed by two-dimensional gel electrophoresis. The comparison of protein patterns obtained with neural and non-neural derivatives of these EC cell lines indicates that two changes are specific for the neural pathway: (i) the appearance of a new beta-tubulin isoform and (ii) the accumulation of the brain isozyme of creatine phosphokinase already present in small amounts in EC stem cells. These changes were found to take place early in the course of differentiation and to occur even when neurite outgrowth was prevented.  相似文献

A straightforward protocol for the site-specific incorporation of a 19F label into any protein in vivo is described. This is done using a plasmid containing an orthogonal aminoacyl-tRNA synthetase/tRNA(CUA) that incorporates L-4-trifluoromethylphenylalanine in response to the amber codon UAG. This method improves on other in vivo methods because the 19F label is incorporated into only one location on the protein of interest and that protein can easily be produced in large quantities at low cost. The protocol for producing 19F-labeled protein is similar to expressing protein in Escherichia coli and takes 4 d to obtain pure protein starting from the appropriate vectors.  相似文献

Ecosystem engineers are species that affect others through the provision of habitat rather than consumptive resources. The extent to which ecosystem engineers can indirectly affect entire food webs, however, is poorly understood. Epiphytic tank bromeliads (Bromeliaceae) are ecosystem engineers that are common throughout the Neotropics, and are associated with a variety of predatory arthropods. Here, we examine if bromeliads, by increasing predator densities, indirectly benefit their support tree through reduction in herbivorous insects and leaf damage. We observed and manipulated bromeliad densities in Costa Rican orange orchards, and measured impacts on leaf damage and arboreal and bromeliad invertebrate communities in two different seasons. Our results show that bromeliads are associated with predatory and herbivorous invertebrates but not leaf damage. Bromeliads were correlated with increased densities of their associated predators, especially ants and hunting spiders, but we could not confirm a causal link. Associations with bromeliads changed over time, with seasonal shifts interfering with responses to our manipulations. Bromeliads had a reduced association with predators in the dry season. Moreover, a null association between bromeliads and herbivorous invertebrates in the dry season unexpectedly became positive in the wet season. In summary, we have only limited evidence that bromeliads indirectly promote the top‐down control of arboreal herbivores; instead, our manipulations suggest that bromeliads increase herbivore densities in the wet season. This research suggests that although bromeliads may act as ecosystem engineers, indirectly influencing the invertebrate food web on support trees, their effects are trophically complex and seasonally dependent. Two putative populations of hooded seals (Cystophora cristata) occur in the North Atlantic. The Greenland Sea population pup and breed on the pack ice near Jan Mayen ('West Ice') while the Northwest Atlantic population is thought to pup in the Davis Strait, in the Gulf of St. Lawrence (the 'Gulf'), and off southern Labrador or northeast Newfoundland (the 'Front'). We used microsatellite profiling of 300 individuals at 13 loci and mitochondrial DNA sequencing of the control region of 123 individuals to test for genetic differentiation between these four breeding herds. We found no significant genetic differences between breeding areas, nor evidence for cryptic nor higher level genetic structure in this species. The Greenland Sea breeding herd was genetically most distant from the Northwest Atlantic breeding areas; however, the differences were statistically nonsignificant. Our data therefore suggest that the world's hooded seals comprise a single panmictic genetic population.  相似文献

Signal-induced apoptosis is a normal phenomenon in which cells respond to changes in their environment through a cascade of intracellular biochemical changes culminating in cell death. However, it is not clear at what point in this process the cell becomes committed to die. An early biochemical change characteristic of cells undergoing apoptosis is the loss of plasma membrane asymmetry, such that high levels of phosphatidylserine become exposed on the outside cell surface. These cells can be recognized by staining with Annexin V, which binds to phosphatidylserine with high affinity. To investigate the mechanisms controlling signal-induced apoptosis we have examined the response of a B cell lymphoma to crosslinking of the membrane immunoglobulin (mIg) receptor. We have found that many of the cells that stain positive for Annexin V are viable and can resume growth and reestablish phospholipid asymmetry once the signal is removed. These results indicate that Annexin V staining, and thus loss of membrane asymmetry, precedes commitment to apoptotic death in this system.  相似文献

The long-term stability of high-level expression is the mostimportant factor to consider when choosing cell lines for the expression of recombinant proteins. Declining volumetricyields in large-scale fermentation can be caused by changes affecting the cell population as a whole such as loss in viability, depletion of nutrients or accumulation of metabolites affecting cell growth. Alternatively, geneticinstability may lead to the outgrowth of a less productive,metabolically favored sub-population. Currently a variety ofparameters are measured to monitor the condition of cells infermenters including glucose uptake, lactate accumulation andoxygen consumption; in addition, periodic viable cell countsallow the determination of the growth rate and viability of the population. All of these methods measure the condition ofthe cell population as a whole and changes must involve a significantly large proportion of the total culture in orderto be detectable. Here we report on a method that allows theevaluation of the productivity of individual cells. Using the gel microdrop secretion assay, we detected the appearance ofa sub-population of cells with lower productivity. Subsequentanalysis of the culture confirmed the existence of lower productivity cells with a lower vector copy number. Therefore,the single cell secretion assay proved to be a rapid method todetect and isolate a low productivity variant of the producer cell line.  相似文献
