The investigation of longitudinal dispersion of tracer substances in unsteady flows has biomechanical application in the study of heat and mass transport within the bronchial airways during normal, abnormal, and artificial pulmonary ventilation. To model the effects of airway curvature on intrapulmonary gas transport, we have measured local gas dispersion in axially uniform helical tubes of slight pitch during volume-cycled oscillatory flow. Following a small argon bolus injection into the flow field, the time-averaged effective diffusion coefficient (Deff/Dmol) for axial transport of the contaminant was evaluated from the time-dependent local argon concentration measured with a mass spectrometer. The value of (Deff/Dmol) is extracted from the curve of concentration versus time by two techniques yielding identical results. Experiments were conducted in two helical coiled tubes (delta = 0.031, lambda = 0.022 or delta = 0.085, lambda = 0.060) over a range of 2 < alpha < 15, 3 < A < 15, where delta is the ratio of tube radius to radius of curvature, lambda is the ratio of pitch height to radius of curvature, alpha is the Womersley parameter or dimensionless frequency, and A is the stroke amplitude or dimensionless tidal volume. Experimental results show that, when compared to transport in straight tubes, the effective diffusivity markedly increases in the presence of axial curvature. Results also compare favorably to mathematical predictions of bolus dispersion in a curved tube over the ranges of frequency and tidal volume studied. 相似文献

Previous microsatellite analysis showed that two subpopulations of perch (Perca fluviatilis L) exist in Lake Constance. This raises questions of whether (i). Lake Constance was colonized by two populations that diverged in allopatry, or (ii). the two subpopulations diverged in sympatry. Sequence analysis of a 365 bp mtDNA fragment (5'-end of the D-loop) of perch from Lake Constance and adjacent waters revealed 10 haplotypes. We suggest colonization via the Danube river, based on the frequency and dispersion of haplotypes, and knowledge of the lake's palaeohydrological development. Pairwise FST-values using mitochondrial DNA sequences showed no significant population subdivision. Our study provides strong evidence that subpopulations of perch in Lake Constance have diverged in sympatry. 相似文献

Giardia lamblia infection of the human small intestine is a common protozoan cause of diarrheal disease worldwide. Although infection is luminal and generally self-limiting, and secretory Abs are thought to be important in host defense, other defense mechanisms probably affect the duration of infection and the severity of symptoms. Because intestinal epithelial cells produce NO, and its stable end products, nitrite and nitrate, are detectable mainly on the apical side, we tested the hypothesis that NO production may constitute a host defense against G. lamblia. Several NO donors, but not their control compounds, inhibited giardial growth without affecting viability, suggesting that NO is cytostatic rather than cytotoxic for G. lamblia. NO donors also inhibited giardial differentiation induced by modeling crucial environmental factors, i. e., encystation induced by bile and alkaline pH, and excystation in response to gastric pH followed by alkaline pH and protease. Despite the potent antigiardial activity of NO, G. lamblia is not simply a passive target for host-produced NO, but has strategies to evade this potential host defense. Thus, in models of human intestinal epithelium, G. lamblia inhibited epithelial NO production by consuming arginine, the crucial substrate used by epithelial NO synthase to form NO. These studies define NO and arginine as central components in a novel cross-talk between a luminal pathogen and host intestinal epithelium. 相似文献

Cathelicidin-related antimicrobial peptide (mCRAMP), the sole murine cathelicidin, is encoded by the gene Cnlp. We show that mCRAMP expression in the intestinal tract is largely restricted to surface epithelial cells in the colon. Synthetic mCRAMP had antimicrobial activity against the murine enteric pathogen Citrobacter rodentium, which like the related clinically important human pathogens enteropathogenic Escherichia coli and enterohemorrhagic E. coli, adheres to the apical membrane of intestinal epithelial cells. Colon epithelial cell extracts from Cnlp+/+ mice had significantly greater antimicrobial activity against C. rodentium than those of mutant Cnlp-/- mice that lack mCRAMP. Cnlp-/- mice developed significantly greater colon surface and crypt epithelial cell colonization, surface epithelial cell damage, and systemic dissemination of infection than Cnlp+/+ mice after oral infection with C. rodentium. Moreover, Cnlp+/+ mice were protected from oral infections with C. rodentium inocula that infected the majority of Cnlp-/- mice. Giardia lamblia is one of the most important causes of waterborne diarrheal disease worldwide, and giardiasis is the most common protozoan infection of the human small intestine. Symptomatic infection is characterized by diarrhea, abdominal pain, and malabsorption, leading to malnutrition and weight loss, particularly in children. The pathogen resides strictly in the lumen of the small intestine, and infection is typically not accompanied by significant mucosal inflammation. Clinical and experimental studies indicate that B cell-dependent host defenses, particularly IgA, are important for controlling and clearing Giardia infection, although B cell-independent mechanisms also contribute to this outcome. In contrast to antigiardial host defenses, much less is known about the pathophysiological mechanisms underlying the clinical symptoms of giardiasis, partly because of the current lack of suitable model systems. In addition to being an important human enteric pathogen, Giardia is an interesting model organism for gaining basic insights into genetic innovations that led to evolution of eukaryotic cells, since it belongs to the earliest diverging eukaryotic lineage known. The completion of the giardial genome project will increase understanding of the basic biology of the protozoan and will help us to better understand host pathogen-interactions as a basis for developing new vaccination and therapeutic strategies. 相似文献

The objective of this study was to evaluate the effect of Eqvalan (ivermectin) on stallion semen quality and freezability. Experiments were performed using 22 Freiberger stallions, randomly divided into a control and test group. Semen was collected once a week for 17 weeks from October 1997 to February 1998. Eqvalan was given orally to all stallions of the test group at a therapeutic dose of 0.2 mg ivermectin/kg. Besides measuring the scrotal width, ejaculates were collected to determine the volume, concentration, and the motility and morphology (normal sperm, major defects, vacuoles and acrosome defects). In addition, the motility and viability (fluorescence staining with propidium iodide/SYBR-14) were tested in all frozen-thawed semen samples. During the experimental period, stallions treated with Eqvalan had significantly better concentration (P < 0.0001) and motility (P < 0.0001) in fresh semen compared to control animals. After freezing-thawing, the motility (P < 0.0001) and viability (P = 0.0003) of semen also increased significantly for treated stallions. Regarding morphology, normal sperm (P < 0.0001), major defects (P = 0.0027) and vacuoles (P = 0.0236) were significantly better in the Eqvalan group. In addition to group differences we also observed a time effect on morphological parameters as shown by a decrease of normal sperm and an increase of major defects in both groups during the experiment. Our results demonstrate that a single oral application of Eqvalan did not negatively influence the quality and freezability of stallion semen in the nonbreeding season. Rather, it seems that Eqvalan has a favorable effect on stallion fertility as most sperm parameters examined were significantly improved in treated animals compared to control. 相似文献
