Incubation of conditioned media derived from mammary epithelial cells and from fibroblasts with [gamma-32P]ATP revealed much higher intensities of labeled polypeptides in transformed cells compared to normal cells. Conditioned media from human mammary carcinoma cells have in common the presence of characteristic phosphoproteins with molecular masses of about 37,22 and 19.5 kDa. Ehrlich Ascites Mammary Carcinoma Cells secrete a dominant 32 kDa phosphoprotein. Normal fibroblasts secrete elevated levels of phosphoproteins after treatment with transforming growth factors. A phosphoprotein with molecular mass of about 37 kDa becomes secreted preferentially if cell-conditioned media were labeled in vivo. The results indicate that the phosphoproteins comprise a family of secretory polypeptides associated with early steps of transformation.  相似文献

The effect on the Ca-dependent potassium current, IK(Ca), of procedures that increase intracellular cAMP levels was studied in Aplysia neurons using three different pharmacological approaches. Exposure to cAMP analogues which were either resistant to or protected from phosphodiesterase hydrolysis caused an increase in IK(Ca) from 30 to 50% in 10 min. The degree of reversibility of this effect varied from complete with db cAMP to very little with pcpt cAMP. Exposure to cholera toxin, which stimulates the synthesis of endogenous cAMP, increased IK(Ca) 25% in 10 min and the effect was not reversible. Both approaches were effective in all seven neuron types studied. Application of serotonin plus phosphodiesterase inhibitor caused an increase in IK(Ca) in neuron R15 but not in the other neuron types. Application of pentylene tetrazole (PTZ) led to a decrease in IK(Ca). It is proposed that elevation of cyclic AMP mediates an increased sensitivity of the IK(Ca) channel to Ca ions.  相似文献

This paper probes the calculation of conformation-related basis spectra from infrared spectra (amide I′band) of reference proteins of known conformational composition and, with their aid, the computation of conformations from the amide I′ band of globular proteins using in both approaches a least-squares, curve-fitting computer program for the analysis of the spectra. The following results were obtained. The infrared basis spectra for the α-helix conformation, the β-(antiparallel-chain pleated sheet) conformation and the ρ-conformation were calculated and their physical reality was substantiated. The basis spectra were shown to be similar when the absorption contributions of the side chains of amino acids were either neglected or taken into account (uncorrected or corrected basis spectra). The mutual correlation of the basis spectra, quantified by the roots of the diagonal elements of the inverse matrix, was found to be low enough only for the β-conformation to allow a statistically reliable estimate of the β-conformation content of proteins. The comparison of the percentages of the β-conformation derived from x-ray structural analysis or calculated from infrared spectra showed the suitability of the basis spectra for the rough estimate of the β-conformation percentages of proteins. The results were not significantly different when using the uncorrected or corrected basis spectra.  相似文献

The Echinococcus granulosus diagnostic arc 5 was revealed by sera from an Alaskan and a Swiss patient with surgically confirmed E. multilocularis infections. The possibility of each patient harboring a concurrent infection with both parasites may be disregarded on the basis of radiologic, scintillographic and surgical data. This conclusion is compatible with ecologic and epidemiologic considerations. These observations suggest that the immunoelectrophoresis test based on arc 5 positivity is not E. granulosus-specific as originally described.  相似文献

Although talents and disabilities appear to run in families, direct links between genes and cognitive ability are difficult to establish. Investigators are currently searching for intermediate phenotypes with plausible links to both genome and cognome (the cognitive phenotype). Cortical anatomy could provide one such intermediate phenotype. Variation in cortical size, asymmetry and sulcal pattern is influenced by genetic variation in neurotrophic factors and can predict variation in verbal and mathematical talent. Anecdotal evidence suggests that individuals with a rare morphological variant of Sylvian fissure sometimes have superior visualization ability combined with verbal deficits. Documentation of such 'cognitive cortical syndromes' might prove as genetically informative as the identification of dysmorphic syndromes associated with mental retardation. A necessary prerequisite for the establishment of such syndromes is a reliable technique for the identification of cortical patterns. Recent technical advances in software for automatically labeling and measuring cortical sulci now provide the possibility of establishing standard measures for their shape, size and location. Such measures are a prerequisite for genetic studies of cortical patterns that could illuminate the neurodevelopmental pathways by which genes affect cognitive ability.  相似文献
