Oral bacterial biofilms trigger chronic inflammatory responses in the host that can result in the tissue destructive events of periodontitis. However, the characteristics of the capacity of specific host cell types to respond to these biofilms remain ill-defined. This report describes the use of a novel model of bacterial biofilms to stimulate oral epithelial cells and profile select cytokines and chemokines that contribute to the local inflammatory environment in the periodontium. Monoinfection biofilms were developed with Streptococcus sanguinis, Streptococcus oralis, Streptococcus gordonii, Actinomyces naeslundii, Fusobacterium nucleatum, and Porphyromonas gingivalis on rigid gas-permeable contact lenses. Biofilms, as well as planktonic cultures of these same bacterial species, were incubated under anaerobic conditions with a human oral epithelial cell line, OKF4, for up to 24h. Gro-1α, IL1α, IL-6, IL-8, TGFα, Fractalkine, MIP-1α, and IP-10 were shown to be produced in response to a range of the planktonic or biofilm forms of these species. P. gingivalis biofilms significantly inhibited the production of all of these cytokines and chemokines, except MIP-1α. Generally, the biofilms of all species inhibited Gro-1α, TGFα, and Fractalkine production, while F. nucleatum biofilms stimulated significant increases in IL-1α, IL-6, IL-8, and IP-10. A. naeslundii biofilms induced elevated levels of IL-6, IL-8 and IP-10. The oral streptococcal species in biofilms or planktonic forms were poor stimulants for any of these mediators from the epithelial cells. The results of these studies demonstrate that oral bacteria in biofilms elicit a substantially different profile of responses compared to planktonic bacteria of the same species. Moreover, certain oral species are highly stimulatory when in biofilms and interact with host cell receptors to trigger pathways of responses that appear quite divergent from individual bacteria.  相似文献

The impact of a sewage point source on the bacterial densities in an intertidal mud flat in Boston Harbor, Mass., was investigated. The area, Savin Hill Cove, acts as a receiving basin for a combined storm and sewage outlet (CSO). Preliminary examination of sediments and overlying water at high tide demonstrated that fecal coliforms were present in sediments at abundances 2 to 4 orders of magnitude higher than in the overlying water column. The following bacterial counts were determined from sediments along a sampling transect extending 460 m from the CSO: total bacteria by epifluorescent microscopy, heterotrophic bacteria by plate counts on nutrient-rich and nutrient-poor media, fecal coliforms and enterococci by membrane filtration, and Vibrio parahaemolyticus by a most-probable-number technique with a resuscitation step. Median sediment grain size, average tidal exposure, carbon/nitrogen ratio, and total organic carbon were also measured. All bacterial indices, except for V. parahaemolyticus, declined significantly with distance from the outfall. Multiple regression analysis indicated that tidal exposure (low tides) may affect densities of total bacteria. Fecal coliforms and enterococci were still present in appreciable numbers in sediments as far as 460 m away from the CSO. In contrast, V. parahaemolyticus densities did not correlate with the other bacterial counts nor with any of the environmental parameters examined. These results indicate that intertidal sediments which adjoin point sources of pollution are severely contaminated and should be considered as potentially hazardous reservoirs of sewage-borne diseases.  相似文献

Summary Food density, degree of hunger prior to foraging, and food processing difficulty were varied in order to determine their effects on the diet diversity of captive Peromyscus leucopus foraging for buried seeds. No relationship was found between diet diversity and food density. However, diet diversity exhibited a significantly positive relationship with hunger, and a significantly negative relationship with seed processing time. Endogenous material present in heat-denatured extracts of rat brain that inhibited the binding of [3H]-isopropyl-4-(2,1,3-benzoxadiazol-4-yl)-1,4-dihydro-5-metho xyca rbonyl-2,6-dimethyl-3-pyridinecarboxylate ([3H]-PN200-110) to calcium channels in brain membranes was purified. Spectrophotometric analysis of material purified by strong anion-exchange and reverse-phase chromatography showed an absorption maximum at 266 nm at pH 7.0 that shifted to 245 nm at pH 2.0. This pH-dependent spectral shift was indistinguishable from that of ascorbic acid. Samples of the purified extract contained ascorbic acid; however, the inhibition of binding by purified material was always greater than the inhibition seen with equivalent concentrations of ascorbate, implying the presence of additional inhibitory factors. Attempts to detect and identify such inhibitory substances by chromatography showed that inhibition activity was coincident with the presence of ascorbate, and the inhibitory activity of purified material was abolished after treatment with ascorbic acid oxidase. Iron enhanced the inhibition produced by ascorbate, and chemical analysis of purified preparations revealed the presence of iron. Studies comparing the potency of the purified material with that of a mixture of ascorbate plus iron showed that the content of ascorbate and iron in the purified brain extract is sufficient to explain the observed inhibition of binding of [3H]PN200-110.  相似文献

A multianalyte immunoassay for simultaneous detection of three analytes (hTSH, hCG and beta-Gal) has been demonstrated using DNA-labeled antibodies and polymerase chain reaction (PCR) for amplification of assay response. The labeled antibodies were prepared by covalently coupling uniquely designed DNA oligonucleotides to each of the analyte-specific monoclonal antibodies. Each of the DNA oligonucleotide labels contained the same primer sequences to facilitate co-amplification by a single primer pair. Assays were performed using a two-antibody sandwich assay format and a mixture of the three DNA-labeled antibodies. Dose-response relationships for each analyte were demonstrated. Analytes were detected at sensitivities exceeding those of conventional enzyme immunoassays by approximately three orders of magnitude. Detection limits for hTSH, beta-Gal and hCG were respectively 1 x 10(-19), 1 x 10(-17) and 1 x 10(-17) mol. Given the enormous amplification afforded by PCR and the existing capability to differentiate DNA based on size or sequence differences, the use of DNA-labeled antibodies could provide the basis for the simultaneous detection of many analytes at sensitivities greater than those of existing antigen detection systems. These findings in concert with previous reports suggest this hybrid technology could provide a new generation of ultra-sensitive multianalyte immunoassays.  相似文献

Sex determination in fishes is often enigmatic, a situation that is often made even more complex by the fact that the process of sexual differentiation in many species may be influenced by environmental conditions. This situation is typified in zebrafish, a popular model organism. Despite the vast array of information available for the species, the genetic controls of sex are unknown. Further, environmental parameters, such as rearing densities, seem to exert an influence on the sex ratios of captive stocks. In an effort to dissect the genetic and environmental controls underlying the expression of sex in this species, we manipulated growth of pure-bred and out-crossed zebrafish by varying their food supply during development. Faster-growing zebrafish were more likely to be female than siblings that were fed less, and out-crossed broods had higher proportions of females than broods from pure-bred crosses. The dependence of sex ratio on feeding rate is readily understood in terms of adaptive sex allocation: zebrafish life history seems to confer the greater pay-off for large size on females. A similar male/female difference in the pay-off for hybrid vigor could similarly account for the female bias of out-crossed broods—and it could be a manifestation of Haldane’s rule.  相似文献

Wildfires in many western North American forests are becoming more frequent, larger, and severe, with changed seasonal patterns. In response, coniferous forest ecosystems will transition toward dominance by fire‐adapted hardwoods, shrubs, meadows, and grasslands, which may benefit some faunal communities, but not others. We describe factors that limit and promote faunal resilience to shifting wildfire regimes for terrestrial and aquatic ecosystems. We highlight the potential value of interspersed nonforest patches to terrestrial wildlife. Similarly, we review watershed thresholds and factors that control the resilience of aquatic ecosystems to wildfire, mediated by thermal changes and chemical, debris, and sediment loadings. We present a 2‐dimensional life history framework to describe temporal and spatial life history traits that species use to resist wildfire effects or to recover after wildfire disturbance at a metapopulation scale. The role of fire refuge is explored for metapopulations of species. In aquatic systems, recovery of assemblages postfire may be faster for smaller fires where unburned tributary basins or instream structures provide refuge from debris and sediment flows. We envision that more‐frequent, lower‐severity fires will favor opportunistic species and that less‐frequent high‐severity fires will favor better competitors. Along the spatial dimension, we hypothesize that fire regimes that are predictable and generate burned patches in close proximity to refuge will favor species that move to refuges and later recolonize, whereas fire regimes that tend to generate less‐severely burned patches may favor species that shelter in place. Looking beyond the trees to forest fauna, we consider mitigation options to enhance resilience and buy time for species facing a no‐analog future.  相似文献

In comparison to isometric muscle action models, little is known about the electromyographic (EMG) and mechanomyographic (MMG) amplitude and mean power frequency (MPF) responses to fatiguing dynamic muscle actions. Simultaneous examination of the EMG and MMG amplitude and MPF may provide additional insight with regard to the motor control strategies utilized by the superficial muscles of the quadriceps femoris during a concentric fatiguing task. Thus, the purpose of this study was to examine the EMG and MMG amplitude and MPF responses of the vastus lateralis (VL), rectus femoris (RF), and vastus medialis (VM) during repeated, concentric muscle actions of the dominant leg. Seventeen adults (21.8+/-1.7 yr) performed 50 consecutive, maximal concentric muscle actions of the dominant leg extensors on a Biodex System 3 Dynamometer at velocities of 60 degrees s(-1) and 300 degrees s(-1). Bipolar surface electrode arrangements were placed over the mid portion of the VL, RF, and VM muscles with a MMG contact sensor placed adjacent to the superior EMG electrode on each muscle. Torque, MMG and EMG amplitude and MPF values were calculated for each of the 50 repetitions. All values were normalized to the value recorded during the first repetition and then averaged across all subjects. The cubic decreases in torque at 60 degrees s(-1) (R2 = 0.972) and 300 degrees s(-1) (R2 = 0.931) was associated with a decline in torque of 59+/-24% and 53+/-11%, respectively. The muscle and velocity specific responses for the MMG amplitude and MPF demonstrated that each of the superficial muscles of the quadriceps femoris uniquely contributed to the control of force output across the 50 repetitions. These results suggested that the MMG responses for the VL, RF, VM during a fatiguing task may be influenced by a number of factors such as fiber type differences, alterations in activation strategy including motor unit recruitment and firing rate and possibly muscle wisdom.  相似文献

Eighteen adults performed isometric muscle actions of the leg extensors at 25, 50, 75, and 100% maximal voluntary contraction (%MVC) at leg flexion angles of 25, 50, and 75 degrees. The results indicated that isometric torque production increased as leg flexion angle increased (75 degrees > 50 degrees > 25 degrees). For each muscle tested (rectus femoris, vastus lateralis, and vastus medialis), the EMG amplitude increased up to 100%MVC at each leg flexion angle (25, 50, and 75 degrees). The MMG amplitude for each muscle, however, increased up to 100%MVC at 25 and 50 degrees of leg flexion, but plateaued from 75 to 100%MVC at 75 degrees of leg flexion. We hypothesize that the varied patterns for the MMG amplitude-isometric torque relationships were due to leg flexion angle differences in: (1) muscle stiffness, (2) intramuscular fluid pressure, or (3) motor unit firing frequency.  相似文献
