Summary

Biomphalaria straminea is a freshwater snail and one of the intermediate hosts of the trematode parasite which causes schistosomiasis in Brazil. The main stages of embryonic development were analyzed with confocal laser scanning microscopy (CLSM), using the fluorescent probe DiOC and Nile Red as a vital stain in in vivo preparations. In fixed preparations nuclei were stained by the Feulgen reaction or the fluorescent DNA probe, Höechst 33342. Results obtained from the analysis of embryos at the stages of early cleavage, morula, blastula, gastrula, early trocophore and veliger showed that these stages are similar to those described for Biomphalaria glabrata and Biomphalaria tenagophila. CLSM optical sections of early trochophore showed important morphological structures such as the blastopore, stomodeum and shell gland; and in early veliger, internal organs such as the esophagus, stomach and male and female ducts were also clearly identified.  相似文献

Homoploid hybrid speciation in animals is poorly understood, mainly because of the scarcity of well‐documented cases. Here, we present the results of a multilocus sequence analysis on the house sparrow (Passer domesticus), Spanish sparrow (P. hispaniolensis) and their proposed hybrid descendant, the Italian sparrow (P. italiae). The Italian sparrow is shown to be genetically intermediate between the house sparrow and Spanish sparrow, exhibiting genealogical discordance and a mosaic pattern of alleles derived from either of the putative parental species. The average variation on the Z chromosome was significantly reduced compared with autosomal variation in the putative parental species, the house sparrow and Spanish sparrow. Additionally, divergence between the two species was elevated on the Z chromosome relative to the autosomes. This pattern of variation and divergence is consistent with reduced introgression of Z‐linked genes and/or a faster‐Z effect (increased rate of adaptive divergence on the Z). FST‐outlier tests were consistent with the faster‐Z hypothesis: two of five Z‐linked loci (CHD1Z and PLAA) were identified as candidates for being subject to positive, divergent selection in the putative parental species. Interestingly, the two latter genes showed a mosaic pattern in the (hybrid) Italian sparrow; that is, the Italian sparrow was found to be fixed for Spanish sparrow alleles at CHD1Z and to mainly have house sparrow alleles at PLAA. Preliminary evidence presented in this study thus suggests that sex chromosomes may play a significant role in this case of homoploid hybrid speciation.  相似文献

We tested the regeneration niche hypothesis by comparing the germination ecology of two sympatric Diplusodon species differing in their geographic range and microhabitat. Diplusodon orbicularis is an endemic shrub achieving high densities on sandy soils, whereas Diplusodon hirsutus has a wider geographic range and establishes on rocky outcrops. Seeds were set to germinate under constant temperatures of 15, 20, 25, 30 and 35°C under 12 h light/12 h dark conditions. Light induced germination in both species, but germination under darkness was also observed. Optimum conditions for germination were 25°C/light for both species. The lack of differences in soil temperatures between rocky outcrops and sandy soils provides evidence of no temperature‐dependent microhabitat selection. Unexpectedly, germinability of the more widespread species was lower than that of the endemic congener. Higher germinability of D. orbicularis may account for higher densities when compared with D. hirsutus. Our results provide limited support for the role of germination in contributing to ecological breadth and geographic range.  相似文献

Abstract: Prior to geochemical analyses, fossil bones and teeth are often extracted from any surrounding lithified sediments using chemical techniques such as immersion in acid. As stable isotope analysis becomes more commonplace in palaeoecological investigations, it is important to consider what effects these chemical preparation techniques may have on any subsequent isotopic data and to constrain these effects as quantitatively as possible. This study aims to elucidate these effects, as it is vital that variability in a data set should not be introduced as a result of protocols used during sample preparation; in addition, it defines the most effective and viable method of carbonate removal for processing bulk fossil samples without causing alteration of their stable isotopic signatures. Various strengths of two weak acids commonly used during palaeontological preparation were tested to evaluate their effects on the δ15N and δ13Corg isotopic signatures of the vertebrae of a large Eocene fossil fish. Changes in the isotopic values occurred over time regardless of which acid was used, each causing a variable response in both δ15N and δ13Corg isotopic values. Without careful monitoring of the acidification process in a controlled environment, any resulting data could therefore confound interpretation. Based on these experiments, it is recommended that 2 m acetic acid be used for the pretreatment of fossils prior to the acquisition of N and C isotope data where carbonate removal is necessary.  相似文献

One of the main options for carbon mitigation identified by the IPCC is the sequestration of carbon in soils. In this paper we use statistical relationships derived from European long-term experiments to explore the potential for carbon sequestration in soils in the European Union. We examine five scenarios, namely (a) the amendment of arable soils with animal manure, (b) the amendment of arable soils with sewage sludge, (c) the incorporation of cereal straw into the soils in which it was grown, (d) the afforestation of surplus arable land through natural woodland regeneration, and (e) extensification of agriculture through ley-arable farming. Our calculations suggest only limited potential to increase soil carbon stocks over the next century by addition of animal manure, sewage sludge or straw (Þbl 15 Tg C y–1), but greater potential through extensification of agriculture (≈ 40 Tg C y–1) or through the afforestation of surplus arable land (≈ 50 Tg C y–1). We estimate that extensification could increase the total soil carbon stock of the European Union by 17%. Afforestation of 30% of present arable land would increase soil carbon stocks by about 8% over a century and would substitute up to 30 Tg C y–1 of fossil fuel carbon if the wood were used as biofuel. However, even the afforestation scenario, with the greatest potential for carbon mitigation, can sequester only 0.8% of annual global anthropogenic CO2-carbon. Our figures suggest that, although efforts in temperate agriculture can contribute to global carbon mitigation, the potential is small compared to that available through reducing anthropogenic CO2 emissions by halting tropical and sub-tropical deforestation or by reducing fossil fuel burning.  相似文献

Our earlier studies have shown that the Vibrio harveyi flavin reductase FRP undergoes a monomer-dimer equilibrium, and luciferase forms a functional complex with the FRP monomer but not significantly with the dimer. This work is aimed at further investigating the nature and regulation of FRP subunit interactions by computation and site-directed mutagenesis approaches. In silico mutations of a number of residues were performed, and energetic analyses led us to target residue E99, which interacts directly with R113 and R225 from the second subunit of the FRP homodimer, for detailed investigation. E99 was found non-essential to the binding of either the FMN cofactor or the substrates. However, in comparison with the native enzyme, the E99K variant was shown to have an enhanced subunit dissociation as evident from a 44-fold higher Kd for the monomer-dimer equilibrium. The critical role of E99 in the formation of the FRP dimer has thus been demonstrated.  相似文献

Current anesthetics, especially the inhaled ones, have troublesome side effects and may be associated with durable changes in cognition. It is therefore highly desirable to develop novel chemical entities that reduce these effects while preserving or enhancing anesthetic potency. In spite of progress toward identifying protein targets involved in anesthesia, we still do not have the necessary atomic level structural information to delineate their interactions with anesthetic molecules. Recently, we have described a protein target, apoferritin, to which several anesthetics bind specifically and in a pharmacodynamically relevant manner. Further, we have reported the high resolution X-ray structure of two anesthetic/apoferritin complexes (Liu, R.; Loll, P. J.; Eckenhoff, R. G. FASEB J. 2005, 19, 567). Thus, we describe in this paper a structure-based approach to establish validated shape pharmacophore models for future application to virtual and high throughput screening of anesthetic compounds. We use the 3D structure of apoferritin as the basis for the development of several shape pharmacophore models. To validate these models, we demonstrate that (1) they can be used to effectively recover known anesthetic agents from a diverse database of compounds; (2) the shape pharmacophore scores afford a significant linear correlation with the measured binding energetics of several known anesthetic compounds to the apoferritin site; and (3) the computed scores based on the shape pharmacophore models also predict the trend of the EC50 values of a set of anesthetics. Therefore, we have now obtained a set of structure-based shape pharmacophore models, using ferritin as the surrogate target, which may afford a new way to rationally discover novel anesthetic agents in the future.  相似文献

Heat stress is a major limiting factor of grapevine production and quality. Acclimation and recovery are essential to ensure plant survival, and the recovery mechanisms can be independent of the heat response mechanisms. An experimental set up with and without acclimation to heat followed by recovery [stepwise acclimation and recovery (SAR) and stepwise recovery (SR), respectively] was applied to two grapevine varieties, Touriga Nacional (TN), and Trincadeira (TR), with different tolerance to abiotic stress. Major differences were found between leaves of SAR and SR, especially after recovery; in SAR, almost all parameters returned to basal levels while in SR they remained altered. Acclimation led to a swifter and short‐term antioxidative response, affecting the plant to a lesser extent than SR. Significant differences were found among varieties: upon stress, TN significantly increased ascorbate and glutathione reduction levels, boosting the cell's redox‐buffering capacity, while TR needed to synthesize both metabolites, its response being insufficient to keep the redox state at working levels. TR was affected by stress for a longer period and the up‐regulation pattern of antioxidative stress genes was more obvious. In TN, heat shock proteins were significantly induced, but the canonical heat‐stress gene signature was not evident probably because no shutdown of the housekeeping metabolism was needed.  相似文献

We present the most comprehensive pan‐European assessment of future changes in cropland and grassland soil organic carbon (SOC) stocks to date, using a dedicated process‐based SOC model and state‐of‐the‐art databases of soil, climate change, land‐use change and technology change. Soil carbon change was calculated using the Rothamsted carbon model on a European 10 × 10′ grid using climate data from four global climate models implementing four Intergovernmental Panel on Climate Change (IPCC) emissions scenarios (SRES). Changes in net primary production (NPP) were calculated by the Lund–Potsdam–Jena model. Land‐use change scenarios, interpreted from the narratives of the IPCC SRES story lines, were used to project changes in cropland and grassland areas. Projections for 1990–2080 are presented for mineral soil only. Climate effects (soil temperature and moisture) will tend to speed decomposition and cause soil carbon stocks to decrease, whereas increases in carbon input because of increasing NPP will slow the loss. Technological improvement may further increase carbon inputs to the soil. Changes in cropland and grassland areas will further affect the total soil carbon stock of European croplands and grasslands. While climate change will be a key driver of change in soil carbon over the 21st Century, changes in technology and land‐use change are estimated to have very significant effects. When incorporating all factors, cropland and grassland soils show a small increase in soil carbon on a per area basis under future climate (1–7 t C ha?1 for cropland and 3–6 t C ha?1 for grassland), but when the greatly decreasing area of cropland and grassland are accounted for, total European cropland stocks decline in all scenarios, and grassland stocks decline in all but one scenario. Different trends are seen in different regions. For Europe (the EU25 plus Norway and Switzerland), the cropland SOC stock decreases from 11 Pg in 1990 by 4–6 Pg (39–54%) by 2080, and the grassland SOC stock increases from 6 Pg in 1990 to 1.5 Pg (25%) under the B1 scenario, but decreases to 1–3 Pg (20–44%) under the other scenarios. Uncertainty associated with the land‐use and technology scenarios remains unquantified, but worst‐case quantified uncertainties are 22.5% for croplands and 16% for grasslands, equivalent to potential errors of 2.5 and 1 Pg SOC, respectively. This is equivalent to 42–63% of the predicted SOC stock change for croplands and 33–100% of the predicted SOC stock change for grasslands. Implications for accounting for SOC changes under the Kyoto Protocol are discussed.  相似文献
