Periphyton nutrient limitation was assessed in Lake Okeechobee, a large, shallow, eutrophic lake in the southeastern U.S.A. Nutrient assays were performed to determine if the same nutrients that limit phytoplankton also limit periphyton growth in the lake. Nutrient diffusing clay substrates containing agar spiked with nitrogen, phosphorus, or both, along with nutrient-free controls, were incubated at four sites in the lake. Three sites were located in a pelagic–littoral interface (ecotone) and one site was located in the interior littoral region. Incubations lasted for 20–26 days, and were repeated on a quarterly basis between 1996 and 1997, to incorporate seasonal variability into the experimental design. The physical and chemical conditions at each site also were measured. Periphyton biomass (chlorophyll a and ash-free dry mass) was highest at the littoral and northern ecotone sites. At the littoral site, nitrogen limited biomass in four of five incubations, although the largest biomass differences between the treatments and controls (3 g cm–2 as chl) were probably not ecologically significant. Periphyton biomass at the western and southern ecotone sites was low compared to the other two sites. Increases in water column depth and associated declines in light penetration strongly correlated with periphyton growth and suggested that they may have limited growth most often at all three ecotone sites. Nitrogen also was found to limit periphyton growth approximately 20% of the time at the ecotone sites and phosphorus was found to limit growth once at the west site.  相似文献

In a time of unprecedented ecological change, understanding natural biophysical relationships between reef resilience and physical drivers is of increasing importance. This study evaluates how wave forcing structures coral reef benthic community composition and recovery trajectories after the major 2015/2016 bleaching event in the remote Chagos Archipelago, Indian Ocean. Benthic cover and substrate rugosity were quantified from digital imagery at 23 fore reef sites around a small coral atoll (Salomon) in 2020 and compared to data from a similar survey in 2006 and opportunistic surveys in intermediate years. Cluster analysis and principal component analysis show strong separation of community composition between exposed (modelled wave exposure > 1000 J m−3) and sheltered sites (< 1000 J m−3) in 2020. This difference is driven by relatively high cover of Porites sp., other massive corals, encrusting corals, soft corals, rubble and dead table corals at sheltered sites versus high cover of pavement and sponges at exposed sites. Total coral cover and rugosity were also higher at sheltered sites. Adding data from previous years shows benthic community shifts from distinct exposure-driven assemblages and high live coral cover in 2006 towards bare pavement, dead Acropora tables and rubble after the 2015/2016 bleaching event. The subsequent recovery trajectories at sheltered and exposed sites are surprisingly parallel and lead communities towards their respective pre-bleaching communities. These results demonstrate that in the absence of human stressors, community patterns on fore reefs are strongly controlled by wave exposure, even during and after widespread coral loss from bleaching events.

We demonstrate a major ecological change in a large lake ecosystem in response to a series of extreme weather events. Phytoplankton community dynamics in subtropical Lake Okeechobee are described from 2000 through early 2008 with emphasis on inter-relationships among phytoplankton populations and associated environmental variables in this large, shallow eutrophic lake. The lake experienced the physical effects of three hurricanes in 2004–2005, which caused massive sediment resuspension, near total elimination of submerged aquatic vegetation, elevated biologically available nutrients and total suspended solids, and lower water transparency. Patterns of long-term co-dominance by nitrogen (N)-fixing cyanobacteria and meroplanktonic diatoms abruptly changed to dominance by only meroplanktonic diatoms. The planktonic cyanobacteria genera Anabaena and Planktolyngbya both decreased approximately an order of magnitude in the post-hurricane period despite large surpluses of bioavailable nutrients. Meroplanktonic diatoms (mostly Aulacoseira spp.) declined approximately 20%, perhaps because of superior competitive ability for light in a polymictic, turbid water column. Canonical Correspondence Analysis (CCA) suggested that reduction in planktonic cyanobacteria after compression of the photic zone and the persistence of meroplanktonic diatoms were related to light utilization traits for the key algal taxa and indicated that pre-existing light limitation and crustacean grazing pressure were intensified in the post-hurricane period.  相似文献

Tm (tropomyosin) is an evolutionarily conserved α-helical coiled-coil protein, dimers of which form end-to-end polymers capable of associating with and stabilizing actin filaments, and regulating myosin function. The fission yeast Schizosaccharomyces pombe possesses a single essential Tm, Cdc8, which can be acetylated on its N-terminal methionine residue to increase its affinity for actin and enhance its ability to regulate myosin function. We have designed and generated a number of novel Cdc8 mutant proteins with N-terminal substitutions to explore how stability of the Cdc8 overlap region affects the regulatory function of this Tm. By correlating the stability of each protein, its propensity to form stable polymers, its ability to associate with actin and to regulate myosin, we have shown that the stability of the N-terminal of the Cdc8 α-helix is crucial for Tm function. In addition we have identified a novel Cdc8 mutant with increased N-terminal stability, dimers of which are capable of forming Tm polymers significantly longer than the wild-type protein. This protein had a reduced affinity for actin with respect to wild-type, and was unable to regulate actomyosin interactions. The results of the present paper are consistent with acetylation providing a mechanism for modulating the formation and stability of Cdc8 polymers within the fission yeast cell. The data also provide evidence for a mechanism in which Tm dimers form end-to-end polymers on the actin filament, consistent with a co-operative model for Tm binding to actin.  相似文献

The SERCA (sarcoplasmic/endoplasmic reticulum Ca2+-ATPase) is probably the most extensively studied membrane protein transporter. There is a vast array of diverse inhibitors for the Ca2+ pump, and many have proved significant in helping to elucidate both the mechanism of transport and gaining conformational structures. Some SERCA inhibitors such as thapsigargin have been used extensively as pharmacological tools to probe the roles of Ca2+ stores in Ca2+ signalling processes. Furthermore, some inhibitors have been implicated in the cause of diseases associated with endocrine disruption by environmental pollutants, whereas others are being developed as potential anticancer agents. The present review therefore aims to highlight some of the wide range of chemically diverse inhibitors that are known, their mechanisms of action and their binding location on the Ca2+ ATPase. Additionally, some ideas for the future development of more useful isoform-specific inhibitors and anticancer drugs are presented.  相似文献

This Biochemical Society Annual Symposium on Recent Advances in Membrane Biochemistry was organized to bring together experts from across the spectrum of biomembrane disciplines from the biological to the biophysical/structural, with the intention of promoting interactions and collaborations across the field. We were keen that the potential for improving human health that stems from a deeper understanding of membrane structure/function should be acknowledged, especially in the light of the increasing numbers of membrane protein structures that continue to be made available to the biomembrane community. This foreword provides an idea of what was communicated in the various sessions and, we hope, gives an impression of the excitement generated by the speakers and delegates at this over-subscribed Symposium.  相似文献
