Salt marshes of the Indian River Lagoon, Florida (USA) were once prolific producers of mosquitoes. Mosquitoes lay their eggs on the infrequently-flooded high marsh surface when the soil surface is exposed. The eggs hatch when the high marsh is flooded by the infrequent high tides or summer rains. To control mosquito production, most of the salt marshes (over 16.200 ha) were impounded by the early 1970s. Flooding, usually by pumping water from the Lagoon, effectively controlled mosquitoes. However, impounding had a profoundly negative impact on the wetland plant, fish, and invertebrate communities. Isolation from the Lagoon cut off aquatic access by transient estuarine species that used the wetlands for feeding or as nursery area. In one study, the number of fish species dropped from 16 to 5 after impounding. Wetland vegetation within some impoundments was totally eliminated; other impoundments developed into freshwater systems. When tidal exchange is restored through hydrologic connection, usually by culverts installed through the perimeter dike, recovery to more natural conditions is often rapid. In one impoundment where wetland vegetation was totally eliminated, recovery of salt-tolerant plants began almost immediately. In another, cover of salt-tolerant plants increased 1,056% in less than 3 years. Fisheries species that benefitted the most were snook, ladyfish, and striped mullet. Over 1,500 juvenile snook were captured in a single 3-hr flood-tide culvert trap as they attempted to migrate into an impoundment. The zooplankton community rapidly returned to the more typical marsh-Lagoon community. Water quality and sediment sulfides returned to typical marsh values. Overall, reconnection enhances natural productivity and diversity, although water quality in the perimeter ditch, an artifact of dike construction, remains problematic. Earlier experiments demonstrated that flooding only during the summer mosquito breeding season provided as effective mosquito control as year-round flooding. In standard management, the impoundment is flooded in summer, then left open to the Lagoon through culverts the rest of the year. Culverts are typically opened when the fall sea level rise first floods the high marsh. Impoundment reconnection is being implemented by a multi-agency partnership. The total reconnected area is expected to reach 9,454 ha by the end of 1998, representing 60% of the impounded wetlands in the entire IRL system. Considerable evidence seems to show that emotional and reflex reactions to feared situations are mediated by the amygdala. It might therefore seem plausible to expect that amygdala-coded fear should also influence decisions when animals make choices about instrumental actions. However, there is not good evidence of this. In particular, it appears, though the literature is conflicted, that once learning is complete, the amygdala may often not be involved in instrumental avoidance behaviours. It is therefore of interest that we have found in rats living for extended periods in a semi-naturalistic ‘closed economy’, where they were given random shocks in regions that had to be entered to obtain food, choices about feeding behaviour were in fact influenced by amygdala-coded fear, in spite of the null effect of amygdalar lesions on fear of dangerous location per se. We suggest that avoidance of highly motivated voluntary behaviour does depend in part on fear signals originating in the amygdala. Such signalling may be one role of well-known projections from amygdala to cortico-striate circuitry.  相似文献

Outbreaks of yaws-like ulcerative skin lesions in children are frequently reported in tropical and sub-tropical countries. The origin of these lesions might be primarily traumatic or infectious; in the latter case, Treponema pallidum subspecies pertenue, the yaws agent, and Haemophilus ducreyi, the agent of chancroid, are two of the pathogens commonly associated with the aetiology of skin ulcers. In this work, we investigated the presence of T. p. pertenue and H. ducreyi DNA in skin ulcers in children living in yaws-endemic regions in Cameroon.Skin lesion swabs were collected from children presenting with yaws-suspected skin lesions during three outbreaks, two of which occurred in 2017 and one in 2019. DNA extracted from the swabs was used to amplify three target genes: the human β2-microglobulin gene to confirm proper sample collection and DNA extraction, the polA gene, highly conserved among all subspecies of T. pallidum, and the hddA gene of H. ducreyi. A fourth target, the tprL gene was used to differentiate T. p. pertenue from the other agents of human treponematoses in polA-positive samples. A total of 112 samples were analysed in this study. One sample, negative for β2-microglobulin, was excluded from further analysis. T. p. pertenue was only detected in the samples collected during the first 2017 outbreak (12/74, 16.2%). In contrast, H. ducreyi DNA could be amplified from samples from all three outbreaks (outbreak 1: 27/74, 36.5%; outbreak 2: 17/24, 70.8%; outbreak 3: 11/13, 84.6%). Our results show that H. ducreyi was more frequently associated to skin lesions in the examined children than T. p. pertenue, but also that yaws is still present in Cameroon. These findings strongly advocate for a continuous effort to determine the aetiology of ulcerative skin lesions during these recurring outbreaks, and to inform the planned mass treatment campaigns to eliminate yaws in Cameroon.  相似文献

The epithelial ovarian carcinomas arise in the ovarian surface epithelium (OSE) which is the mesothelial covering of the ovary. Studies of human USE have been hampered by the small amounts and limited lifespan of this epithelium in culture. OSE cells expressing SV40 large T antigen (Tag) or the HPV genes E6 and E7 have increased growth potentials but lack some of the normal characteristics of OSE. In this study, we used conditional SV40 Tag expression to produce OSE cells with increased proliferative potentials but relatively normal phenotypes. Primary OSE cultures from three women, one of whom had a BRCA1 mutation, were infected with a temperature-sensitive Tag construct (tsTag), and from these, 28 monoclonal and four polyclonal lines were isolated. The effects of temperature changes were examined in two monoclonal and two polyclonal lines. At the permissive temperature (34 degrees C), these cell lines underwent 52-71 population doublings (PD) compared to 15-20 PD for normal OSE. Nuclear SV40-Tag and p53 expression, demonstrated by immunofluorescence, showed that tsTag was uniformly present and biologically active in all lines. At 34 degrees C, culture morphologies ranged from epithelial to mesenchymal. The mean percentage of cells expressing the epithelial differentiation marker, keratin. varied between lines from 20 to 97%. Collagen type III, a mesenchymal marker expressed by OSE in response to explantation into culture, was present in 24-43% of cells. At 39 degrees C, tsTag was inactivated by 2 d while nuclear p53 staining diminished to control levels over 2 wk. Over 3 d. the cells assumed more epithelial morphologies, keratin expression reached 85-100% in all lines and collagen expression increased significantly in two lines. The cultures with the BRCA1 mutation expressed the most keratin and the least collage n III at both temperatures. As indicated by beta-galactosidase staining at pH 6.0, changes leading to senescence were initiated at 39 degrees C by 6 h and were present in all cells after 24 h. However, the cells underwent 1-3 population doublings over up to 1 wk before growth arrest and widespread cell death, thus providing an experimental system where large numbers of OSE cells with different genetic backgrounds and growth potentials can be studied without the concurrent influence of Tag.  相似文献
