Molecular cloning of Proteus morganii phenylalanine deaminase gene in Escherichia coli 总被引:1,自引:0,他引:1
The gene for phenylalanine deaminase (PAD) of Proteus morganii strain 2815 has been isolated on a 6.3-kb HindIII restriction fragment and cloned within RP4-prime plasmids, pYB2321 and pYB2322, in both orientations. Expression of the cloned gene in Escherichia coli strains was comparable to that in P. morganii 2815. The hybrid plasmids mobilized the 2815 chromosome with trajectories in reverse directions from an origin between ser-2 and ade-1, suggesting the map location of the PAD gene. 相似文献

We have determined the prevalence of familial hypercholesterolaemia (FH) in a rural Afrikaner community by means of direct
DNA screening for three founder-related Afrikaner low density lipoprotein (LDL) receptor gene mutations. A random sample of
1612 persons, aged 15–64 years, was selected as a subsample of 4583 subjects from an Afrikaner community living in the south-western
Cape, South Africa. Participants who had a total serum cholesterol (TC) in the high TC category as defined in the consensus
recommendations by the Southern African Heart Foundation, were screened for three founder-related LDL receptor gene mutations,
causing FH in 90% of Afrikaners. Of the subsample, 201 participants (12.5%) had TC levels above the 80th percentile. In this
group the combined prevalence of the three common Afrikaner LDL receptor gene defects (D206E, FH Afrikaner-1; V408M, FH Afrikaner-2;
D154N, FH Afrikaner-3) was calculated as 1 : 83. When taking into account the reported background prevalence of other FH gene
defects of 1 : 500 in this community, their overall prevalence of FH was estimated to be 1 : 72. The significant differences
found between the FH patients and other high risk patients with raised cholesterol levels were higher TC and LDL cholesterol
levels and lower high density lipoprotein cholesterol levels in FH patients. The treatment status of the molecularly identified
FH patients and other hypercholesterolaemic persons suggests that this condition is inadequately diagnosed and poorly managed
in this study population. An extrapolation to the entire South African population suggests that there are about 112 000 FH
patients in the country who are underdiagnosed as a group and therefore not receiving the care that would help to reduce the
burden of FH-associated ischaemic heart disease in South Africa.

An Immunogold labeling procedure and experimental data are presented, which demonstrate that antibodies produced against a bovine serum albumin-abscisic acid conjugate can be used both to characterize the cellular and subcellular localization of abscislc acid (ABA), and to permit quantitative comparisons of this hormone in the subcellular compartments prior to and at times of drought stress. At the control leaf water potential (approximately -0.45 MPa), a quantitatively similar positive labeling pattern was observed in the chloroplasts and apoplast. A twofold drought stress-induced increase in the apoplastic ABA concentration was observed in the drought stressed leaf tissue (i.e., at a leaf water potential of approximately -1.55 MPa), while the ABA concentration in the chloroplasts did not differ from that of the controls. Three histochemical controls and the physiological observations validated the specificity of the procedure. Based on the labeling patterns we observed and literature cited, the validity of the hypothesis that drought stress induces a release of chloroplastic ABA is questioned. We interpreted our results as providing indirect evidence for a drought stress-induced root source origin for the increased apoplastic ABA concentrations. 相似文献

Water lettuce, Pistia stratiotes, and red water fern, Azolla filiculoides, are floating aquatic macrophytes that have become problematic in South Africa. Two weevils, Neohydronomus affinis and Stenopelmus rufinasus, are successful biological control agents of these two species in South Africa. The aim of this study was to investigate the thermal requirements of these two species to explain their establishment patterns in the field. Laboratory results showed that both weevils are widely tolerant to cold and warm temperatures. The critical thermal minima (CTmin) of N. affinis was determined to be 5.58?±?0.31°C and the critical thermal maxima (CTmax) was 44.52?±?0.27°C, while the CTmin of S. rufinasus was 5.38?±?0.33°C and the CTmax?44.0?±?0.17°C. In addition, the lower lethal temperatures were ?9.85?±?0.06°C for N. affinis and ?6.85?±?0.13°C for S. rufinasus, and the upper lethal temperatures were 42.7?±?0.85°C for N. affinis and 41.9?±?2.52°C S. rufinasus. Using the reduced major axis regression method, the development for N. affinis was described using the formula y?=?12.976x?+?435.24, while the development of S. rufinasus was described by y?=?13.6x?+?222.45. These results showed that S. rufinasus develops twice as fast as N. affinis. Using these formulae and temperature data obtained from the South African Weather Service, N. affinis was predicted to complete between 4 and 9 generations per year in South Africa, while S. rufinasus was predicted to complete between 5 and 14 generations per year around the country. These results suggest that both species should not be limited by cold winter, nor warm summer temperatures, and should establish throughout the ranges of the weeds in South Africa. 相似文献

The wall of the stomach of the tigerfish is described and compared with that of other vertebrates. Light microscopic and ultrastructural characteristics of the stomach wall correspond to a large extent to those of other vertebrates, although some differences are found. The mucosa contains (1) surface epithelium characterized by narrow columnar cells with abundant mucous granules; (2) gastric glands consisting of pepsinogenic cells of variable height, containing tubulovesicles and bearing microvilli; (3) five granulated cell types located basally in the epithelium (types 1–5); and (4) lamina propria and muscularis mucosae. Connective tissue separating smooth muscle fibers of the muscularis mucosae constitutes a stratum compactum. The submucosa contains a loose connective tissue, a tunica muscularis of inner circular and outer longitudinal layers, and a serosa of mesothelium and subjacent connective tissue. Immunocytochemical tests with antisera to five polypeptides show gastrin/cholecystokinin (CCK), vasoactive intestinal polypeptide (VIP) immunoreactivities in some cells of the gastric glands, and somatostatin in cells lying among epithelial cells lining the gastric luminal surface or gastric pits. 相似文献
