Phosphatidic acid (PA) is an important signalling lipid involved in various stress‐induced signalling cascades. Two SnRK2 protein kinases (SnRK2.4 and SnRK2.10), previously identified as PA‐binding proteins, are shown here to prefer binding to PA over other anionic phospholipids and to associate with cellular membranes in response to salt stress in Arabidopsis roots. A 42 amino acid sequence was identified as the primary PA‐binding domain (PABD) of SnRK2.4. Unlike the full‐length SnRK2.4, neither the PABD‐YFP fusion protein nor the SnRK2.10 re‐localized into punctate structures upon salt stress treatment, showing that additional domains of the SnRK2.4 protein are required for its re‐localization during salt stress. Within the PABD, five basic amino acids, conserved in class 1 SnRK2s, were found to be necessary for PA binding. Remarkably, plants overexpressing the PABD, but not a non‐PA‐binding mutant version, showed a severe reduction in root growth. Together, this study biochemically characterizes the PA–SnRK2.4 interaction and shows that functionality of the SnRK2.4 PABD affects root development.  相似文献

In this study we examined ecosystem respiration (RECO) data from 104 sites belonging to FLUXNET, the global network of eddy covariance flux measurements. The goal was to identify the main factors involved in the variability of RECO: temporally and between sites as affected by climate, vegetation structure and plant functional type (PFT) (evergreen needleleaf, grasslands, etc.). We demonstrated that a model using only climate drivers as predictors of RECO failed to describe part of the temporal variability in the data and that the dependency on gross primary production (GPP) needed to be included as an additional driver of RECO. The maximum seasonal leaf area index (LAIMAX) had an additional effect that explained the spatial variability of reference respiration (the respiration at reference temperature Tref=15 °C, without stimulation introduced by photosynthetic activity and without water limitations), with a statistically significant linear relationship (r2=0.52, P<0.001, n=104) even within each PFT. Besides LAIMAX, we found that reference respiration may be explained partially by total soil carbon content (SoilC). For undisturbed temperate and boreal forests a negative control of total nitrogen deposition (Ndepo) on reference respiration was also identified. We developed a new semiempirical model incorporating abiotic factors (climate), recent productivity (daily GPP), general site productivity and canopy structure (LAIMAX) which performed well in predicting the spatio‐temporal variability of RECO, explaining >70% of the variance for most vegetation types. Exceptions include tropical and Mediterranean broadleaf forests and deciduous broadleaf forests. Part of the variability in respiration that could not be described by our model may be attributed to a series of factors, including phenology in deciduous broadleaf forests and management practices in grasslands and croplands.  相似文献

The effect of hybridization on morphological variation was investigated in 120 western house mice, Mus musculus domesticus , from the hybrid zone between the Barcelona and standard chromosomal races. The incidence of 37 non-metric cranial traits was calculated for standard mice (2 n  = 40) and Barcelona-standard hybrids (2 n  = 27–39). Subsequent analyses were conducted on several karyological subgroups, established by grouping the animals according to either their diploid number or their degree of chromosomal heterozygosity. Results revealed no significant difference by sex, asymmetry, or geographical distance. Significant phenetic divergences were found between the karyotypes studied in relation to several variants. Differences were especially substantial between the standard race and hybrid mice, even with respect to those hybrids with karyotypes close to that of the standard race. Within the hybrids, the maximum divergence corresponded to the 28-chromosome homozygotes, chromosomally close to the Barcelona race, and to the heterozygotes with more than two fusions. Since differences in non-metric trait frequencies are generally considered a measure of genetic divergence, the results suggest the occurrence of a barrier to gene flow in the Barcelona hybrid zone. The decrease of genetic exchange between the chromosomally differentiated mice might be due to reduced fertility in hybrids, associated with chromosomal heterozygosity.  © 2003 The Linnean Society of London, Biological Journal of the Linnean Society , 2003, 80 , 313–322.  相似文献

1. A comparative study of fatty acid (FA) profiles in particulate matter (seston) and the key grazer Daphnia was performed in six high Arctic ponds (79°N, Svalbard). The ponds were all small and shallow, but followed a strong gradient with respect to nutrient content and optical properties. 2. A distinct locality‐specific pattern was detected by principal component analysis of FA profiles, where samples from each locality clustered both with regard to seston and Daphnia. Linear discriminant analysis using nine sestonic fatty acids as discriminant functions gave on average a correct prediction of the Daphnia lake membership in 47% of cases, with very high predictability in some lakes but poor predictability in others. 3. No significant correlation was detected between FA and nutrient concentration or levels of dissolved organic carbon. The major determinant of FA profiles as judged from a redundancy analysis was the taxonomic composition of phytoplankton communities, notably the biomass of Chlorophyceae. 4. The FA profiles of Daphnia were for some FAs strongly enriched relative to the seston, while diluted for others. Among the polyunsaturated fatty acids (PUFAs), a pronounced magnification of eicosapentaenoic acid (EPA, 20 : 5 n‐3), and to some extent 18 : 3 n‐3 and 20 : 4 n‐6 was found, while docosahexaenoic acid (DHA, 22 : 6 n‐3) contributed in general less to FAs in Daphnia than in seston and was hardly detectable in Daphnia from most localities. 5. The overall content of PUFAs in Daphnia was consistently high, close to 40% of total FA in all investigated localities, despite major differences in seston PUFA content. Daphnia thus acts as a regulator with regard to overall PUFAs, reflecting its physiological constraints and relatively fixed demands for PUFAs in general. The distinct locality‐specific profiles in Daphnia strongly suggest a kind of FA‐fingerprint, but our data do not allow strict statements on the use of specific FAs as trophic markers.  相似文献

Lethaia, Vol. 45, pp. 210–226. The middle Miocene Rubielos de Mora Konservat‐Lagerstätte of northeast Spain is hosted within profundal, finely laminated, lacustrine mudstones. The diverse biota includes abundant salamanders. Most individuals died during separate episodes and sank rapidly postmortem. Specimens are typically preserved in dorso‐ventral aspect, the most hydrodynamically stable orientation. The near‐cylindrical morphology of the body, however, allowed some carcasses to settle in or subsequently re‐orientate into, lateral orientations. Loss of skeletal elements (i.e. reduced completeness) reflects their location within the body and followed a distal to proximal trend. Two stages are identified: initial loss of a small number of phalanges, followed by loss of more proximal limb bones plus additional phalanges. Disarticulation is more complex: it occurred via several mechanisms (notably, abdominal rupture and re‐orientation of part of the body and limbs during decay) and shows no consistent pattern among specimens. The physical taphonomy of the salamanders is controlled predominantly by intrinsic biological factors, i.e. the geometry of the body and of individual skeletal elements, the orientation, inherent strength and location of specific joints and the extent to which soft tissues, particularly the skin, persist during decay. 1. Changes in riparian vegetation owing to forest harvesting may affect the input of large wood, a major structural element, to streams. Studies of large wood impacts on stream fish have focused on population‐level responses, whereas little attention has been given to how wood affects fish behaviour. 2. In a laboratory stream experiment, we tested how two size classes of brown trout, Salmo trutta, (mean size of 85 and 125 mm), alone and together, responded to a gradient of large wood in terms of activity, foraging on terrestrial drift and interactions between conspecifics. 3. The results showed that the presence of large wood significantly reduced the overall activity of the fish, the number of agonistic interactions between individuals and the proportion of captured prey. However, activity decreased relatively more than the proportion of captured prey, resulting in a significant positive net effect of wood on the number of prey captures per time spent active (PTA). This indicates that trout living in habitats with high wood density may have a higher net energy gain than trout living in habitats with less wood. 4. There were no observable size‐class differences in the benefits of large wood or in the utilisation of surface‐drifting terrestrial prey. 5. These results suggest that the presence of large wood may be an important factor shaping stream communities and that a lack of structural complexity may decrease energy gain, increase agonistic interactions and, consequently, lower the production of brown trout.  相似文献

Background: This article describes the types of community‐wide benefits provided by investigators conducting public health research in South Asia as well as their self‐reported reasons for providing such benefits. Methods: We conducted 52 in‐depth interviews to explore how public health investigators in low‐resource settings make decisions about the delivery of ancillary care to research subjects. In 39 of the interviews respondents described providing benefits to members of the community in which they conducted their study. We returned to our narrative dataset to find answers to two questions: What types of community‐wide benefits do researchers provide when conducting public health intervention studies in the community setting, and what reasons do researchers give when asked why they provided community‐wide benefits? Findings: The types of community‐wide benefits delivered were directed to the health and well‐being of the population. The most common types of benefits delivered were the facilitation of access to health care for individuals in acute medical need and emergency response to natural disasters. Respondents' self‐reported reasons when asked why they provided such benefits fell into 2 general categories: intrinsic importance and instrumental importance.  相似文献

The majority of authors consider Festuca jubata Lowe as an endemic species common to Madeira and the Azores. Saint-Yves proposed that F. jubata was an Azorean endemic and described a geovicarious taxon in Madeira: F. filiformis C. Sm. ex Link in Buch ssp. mandonii St.-Yves. We undertook a complete bibliographical revision of the taxonomy, nomenclature, and chorology of F. jubata s.l. , and contrasted it with morphological and anatomical studies performed on samples from the Azores and Madeira. Azorean plants usually identified as F. jubata had a character combination distinct from that of those with a Madeiran provenance. Saint-Yves' proposal of two independent taxa was correct, but he erroneously considered F. jubata as an Azorean endemic because the name F. jubata was based on Madeiran plants.
