Background

Impaired cutaneous wound healing is common in humans, and treatments are often ineffective. Based on the significant emotional and economic burden of impaired wound healing, innovative therapies are needed. The potential of mesenchymal stromal cell (MSC)–secreted factors to treat cutaneous wounds is an active area of research that is in need of refinement before effective clinical trials can be initiated. The aims of the present study were to (i) study which MSC-secreted factors stimulate dermal fibroblast (DF) migration in vitro and (ii) evaluate the potential of these factors to promote wound healing in vivo.

Methods

To this end, MSCs were isolated from the peripheral blood of healthy horses, a physiologically relevant large animal model appropriate for translational wound-healing studies. Conditioned medium (CM) from cultured equine MSCs was analyzed using liquid chromatography-mass spectrophotometry (LC-MS/MS) to identify secreted proteins of interest. Double-stranded RNA-mediated interference (RNAi) was used to silence the genes encoding selected proteins, and the effects of CM from these transfected MSCs on migration of cultured equine DF cells in vitro and full-thickness wounds in mice were evaluated.

Results

We found that MSC-derived plasminogen activator inhibitor-1 (PAI-1) and tenascin-C significantly increased DF migration in vitro and improved wound healing in vivo by decreasing time to wound closure.

Discussion

These results suggest that in a complex wound environment, MSC-secreted factors PAI-1 and tenascin-C contribute to the positive effect of therapeutically applied MSC CM on wound healing.  相似文献

Abstract The recovery of fynbos vegetation after invasion by dense stands of alien trees, and clearing by either ‘burn standing’, ‘fell and burn’, or ‘fell, remove and burn’ treatments, was investigated in two watersheds in the Western Cape Province, South Africa. Native plant density, cover, functional and biological guilds and species richness were compared with matched control sites that were not invaded, but were burnt in the same fires. Species richness was lower for invaded sites compared to controls, at all scales measured (up to 2000m2). Species area curves for invaded sites did not converge with those of controls, indicating that lower richness at smaller scales was not compensated by increased species survival at a larger scale. Indigenous plant density and cover were lower for invaded sites compared to controls. Overall, treatment differences were non‐significant, but the ‘burn standing’ treatment caused the least change to vegetation variables, and the ‘fell, remove and burn’ and ‘fell and burn’ treatments caused greater, similar changes. Changes to the guild structure of the recovering fynbos stands differed among treatments, and indicated that the ‘fell and burn’ treatment had the greatest negative effect on guild survival. In the ‘fell and burn’ treatment, which resulted in an exceptionally intense fire, only non‐mycorrhizal graminoids (predominantly myrmecochores) persisted relatively well. Because of practical problems associated with the ‘burn standing’ and ‘fell, remove and burn’ treatments, managers often have little option but to apply the ‘fell and burn’ treatment. Our results illustrate the dangers of this, and highlight the need for intervention before areas become densely invaded. They also highlight the need for effective biological control agents to reduce rates of spread of aggressively invasive species.  相似文献

Evaluation of PAPNET-assisted cervical rescreening
We have compared the results of targeted manual rescreening of 1211 randomly selected smears with the results of PAPNET-assisted rescreening of 1613 cervical smears, containing at least 6.3% low-grade squamous intraepithelial lesion (SIL). PAPNET diagnosis and the targeted rescreening diagnosis were compared with the initial report, issued on the corresponding smear. Reproducibility scores for inadequacy, presence of endocervical and endometrial cells, specific infections and squamous cell abnormalities were determined. The reproducibility scores for the diagnosis of inadequate smears and specific infections were lower with the PAPNET-assisted rescreening. The detection of squamous cell abnormalities was excellent for both methods (>0.95), with a higher detection rate for false-negative smears with the PAPNET testing system.  相似文献

In this paper we demonstrate the study of plant water balanceby the non-invasive measurement of tissue water content andwater flow using proton nuclear magnetic resonance (NMR). Sapvelocity and flux were measured independently in the presenceof an excess of stationary tissue water. The instrumentationdescribed allows automated and unattended measurement of flow-and water content-variables in a well-defined region of theplant over periods of several days, with a time resolution betweensuccessive measurements of c. 5 s. Using this apparatus theeffect of changes in light intensity (day/night rhythm) andrelative humidity on stem tissue water content as well as onthe velocity and flux of xylem sap in the stem were investigatedin a cucumber plant. The results are in agreement with predictionsfrom a simple model for plant water balance, which is basedon water potential, flow rate and resistance to flow. As longas only transpiration is varied, flow rate and water content(or potential) are affected in opposite ways as demonstratedin this paper. In contrast, the model predicts that changesin uptake (resulting from changes in, for example, root resistance)will induce changes in water content and flow in the same direction.An experimental verification of this prediction is given ina subsequent paper, where, in addition, the NMR results arecompared to those obtained with a dendrometer. The effects of carbon dioxide and the induction of morbidity on aversion learning in larvae of the Pacific Coast wireworm Limonius canus LeConte (Coleoptera: Elateridae) are discussed. Wireworms preconditioned by exposing them one or four times to odour of Tefluthrin 20SC and Dividend XLRTA [Syngenta Crop Protection (Canada), Inc., Canada] during the induction of temporary morbidity subsequently contact tefluthrin‐treated wheat seeds in soil bioassays for as long as naïve (i.e. not preconditioned) larvae but are repelled four to five‐fold more frequently by Dividend‐treated seeds in soil bioassays than naïve wireworms, suggesting that wireworms are capable of associating a novel odour (i.e. Dividend) with morbidity but require a minimum of 10–15 min subsequent contact time with treated seeds before being repelled. Wireworms preconditioned by exposure to peppermint odour during the induction of morbidity are not subsequently repelled by peppermint odour in soil bioassays, suggesting that wireworms are either not capable of aversion learning or that the presence of a CO2 source and/or a suitable host plant may override a negative cue (i.e. peppermint odour). In studies conducted in the absence of soil, a host plant and CO2 production, wireworms are repelled slightly by droplets of 1.0% but not 0.1% peppermint oil. Previous exposure to peppermint odour or contact with peppermint oil‐treated filter paper during one induction of morbidity does not increase the repellency of wireworms to 1.0% peppermint oil significantly. Repellency to 1.0% peppermint oil is almost eliminated when morbidity is induced five times in the absence of peppermint odour but is restored when peppermint odour is present during preconditioning. These findings suggest that wireworm sensitivity to repellent compounds decreases when repeatedly made moribund, although the results are not sufficient to conclude that wireworms are capable of associative learning.  相似文献
