Autophagy regulates cellular homeostasis through degradation of aged or damaged subcellular organelles and components. Interestingly, autophagy-deficient beta cells, for example Atg7-mutant mice, exhibited hypoinsulinemia and hyperglycemia. Also, autophagy response is diminished in heart of diabetic mice. These results implied that autophagy and diabetes are closely connected and affect each other. Although protein O-GlcNAcylation is up-regulated in hyperglycemia and diabetes, and O-GlcNAcylated proteins play an important role in metabolism and nutrient sensing, little is known whether autophagy affects O-GlcNAc modification and vice versa. In this study, we suppressed the action of mTOR by treatment of mTOR catalytic inhibitors (PP242 and Torin1) to induce autophagic flux. Results showed a decrease in global O-GlcNAcylation, which is due to decreased OGT protein and increased OGA protein. Interestingly, knockdown of ATG genes or blocking of lysosomal degradation enhanced protein stability of OGT. In addition, when proteasomal inhibitor was treated together with mTOR inhibitor, protein level of OGT almost recovered to control level. These data suggest that mTOR inhibition is a more efficient way to reduce protein level of OGT rather than that of CHX treatment. We also showed that not only proteasomal degradation regulated OGT stability but autophagic degradation also affected OGT stability in part. We concluded that mTOR signaling regulates protein O-GlcNAc modification through adjustment of OGT stability.  相似文献

Factorial combinations of three photoperiods (10, 13 and 16h), two day temperatures (18 and 28 °C) and two night temperatures(5 and 13 °C) were imposed on nodulated plants of six diversegenotypes of faba bean (Vicia faba L.). Plants were grown inpots in growth cabinets from both vernalized (1.5±0.5°C for 30 d) and non-vernalized seeds. The times from sowingto the appearance of first open flowers (f) were recorded. Seedvernalization decreased the subsequent time taken to flowerin almost all genotype x growing environment combinations (theexceptions were plants of the cv. Maris Bead grown in threecooler, short-day regimes). The influence of temperature andphotoperiod on the rate of flowering was quantified, using amodel applied previously to other long-day species of grainlegume in which positive linear relations between both temperatureand photoperiod and the rate of progress towards flowering areassumed to apply. A significant positive linear response ofrate of progress towards flowering to limited ranges of meandiurnal temperature was detected in all six genotypes, but inthree genotypes (Syrian Local Large, Aquadulce and Maris Bead)the 28 °C day temperature reduced the rate of progress towardsflowering - suggesting that the optimum temperature for floweringin these genotypes is below 28 °C. In four genotypes (MarisBead, Giza-4, Aquadulce and BPL 1722) a significant positiveresponse to photoperiod, typical of quantitative long-day plants,was observed only in plants grown from vernalized seeds. Incontrast, plants of the genotype Zeidab Local grown from bothnon-vernalized and vernalized seeds showed the same positiveresponse to photoperiod, whereas plants of the land-race SyrianLocal Large were consistently unresponsive to photoperiod. Theimplications of this range of responses amongst diverse genotypesare discussed in relation to screening germplasm. A survey amongst members of the British Society for Clinical Cytology (BSCC) was performed in 1990 and 1991 into their experience of fine needle aspiration (FNA) cytology of the breast. Here we present the findings of that part of the survey covering organizational aspects of the service and methods of specimen handling. the replies demonstrate differences in approach to many of the topics covered. There is a preference for the examination of direct smears (rather than cytospins) with air dried Giemsa staining and wet fixation Papanicolaou staining finding nearly equal favour. Some perceived problems were highlighted, including availability and funding of training and quality assurance as well as facilities for liaison between pathologists, clinicians and radiologists.  相似文献

The genus Volborthella is removed from synonymy under Salterella and placed in the extinct phylum Agmata. Volborthella is more restricted geographically and occurs in older Lower Cambrian strata than Salterella, but distinguishing younger specimens of that genus from poorly preserved Salterella is difficult. Vologinella is not related to Volborthella or other Agmata. Other genera assigned to the phylum by various workers are discussed, but their assignment to this phylum is either enigmatic or rejected. The published concept that specimens of Volborthella and Salterella are agglutinated sclerites or spines on a larger soft-bodied organism is discussed. The material on which this notion is based is re-illustrated and the notion rejected in favor of the view that these conical forms are the hard parts of discrete organisms.  相似文献

The developing caryopsis of rice (Oryza sativa L.) was examinedhistologically at successive stages of grain-filling in orderto identify the factors which determine the distribution ofstorage protein in the endosperm, and which terminate the depositionof endosperm protein. The storage protein was deposited at theperiphery of the endosperm, and this distribution was apparentlycaused by the radial pattern of cell development in the endosperm,and by the proximity of the peripheral endosperm cells to thenucellar epidermis. The nucellar epidermis directly surroundsthe endosperm and functions as the pathway for amino acid transportto the endosperm. During the later stages of caryopsis developmentthe nucellar epidermis became compressed by being ‘sandwiched’between the expanding endosperm and the rigid hull (the tightlylocked palea and lemma) which encloses the caryopsis. It isproposed that this compression of the nucellar epidermis blocksthe supply of amino acids to the endosperm and thereby terminatesthe deposition of storage protein in the rice grain.
