Various daily photon doses (daily net number of photons perunit area), provided by varying both photon flux density andphotoperiod, were applied to dormant seed populations of fourgraminaceous species tested for germination in alternating-temperatureregimes on filter papers moistened with either water or a solutionof potassium nitrate. In all four species germination was dependentupon daily photon dose, the product of photon flux density andduration, and quantified by linear relations between the probitof percentage germination and the logarithm of the daily photondose determined by probit analysis. Photon flux density andphotoperiod only influenced germination indirectly as factorsof daily photon dose, except in Echinochloa turnerana (Domin)J.M. Black at high daily photon doses where lower germinationwas observed in continuous light than in 8 h d–1 photoperiod.The low energy reaction, in which increasing dose promotes seedgermination, was observed in Echlnochloa turnerana and Eragrostistef(Zucc.) Trotter but not in the populations of Brachiariahumidicola (Rendle) Schwcickerdt and Panicum maximum Jacq. usedin this study. The high irradiance reaction, in which increasingthe photon dose reduces germination, was, however, observedin all four species. In Eragrostis tef potassium nitrate influencedthe response of seed germination to light, but in the remainingspecies its influence on this relationship was either additive(Panicum maximum) or nil (Brachiaria humidicola and Echinochloaturnerana) in the alternating-temperature regimes used in thisstudy. The implications of the quantal response to photon dosefor devising optimal light regimes to be applied in laboratorytests intended to maximize seed germination are discussed. The relationship between vegetational type and a number of soil chemical factors was examined in secondary successions from fire-maintained eucalypt/grass to climax rainforest communities growing on uniform granitic soil parent material. Canonical variates analysis, which utilized the following variables: pH; loss on ignition; total N, P, K, Ca, and Mg; cation exchange capacity and exchangeable Ca, K, and Mg; and potentially mineralizable N, revealed close overall similarity between surface soils of adjacent types, and significant differences among those of types distant from each other in the successional sequences. Exchangeable Ca, mineralizable N, total N. P, and Mg, and pH all differed significantly among soils of the vegetational types. However, the only identifiable gradients in soil properties that were detected within a successional sequence were in total and mineralizable N, which tended to increase, and pH, which generally tended to decrease with progression towards the climax vegetation. Nitrification was promoted by the presence of Acacia dealbata and apparently inhibited by the presence of Leptospermum lanigerum; it was more rapid in soils beneath late successional vegetation than in those from climax vegetation or early stages of succession, and was inhibited in soil from old (> 200 years) grassland. It was concluded that differences among soils in chemical composition and rates of mineralization of N were due to differences in species composition of the vegetational types that they carried for the time being.  相似文献

A DNA directed, cell-free system which synthesized L-ribulokinase coded by the L-arabinose operon ara OIBAD, has been developed. L-arabinose but not the araC gene product is required for the expression of this operon and, in addition, cyclic AMP and guanosine tetraphosphate are needed for optimal synthesis.  相似文献

Small experimental swards of three types-healthy, 50% and 100% streak-virus-infected-were established from single tillers of randomly selected cocksfoot genotypes. In these swards 44% of infected plants died within 2 years compared with 21% of healthy plants. Mortality of infected plants was not increased by the presence of healthy plants in the same sward. In swards containing both healthy and infected plants, increased growth of healthy plants compensated for the low yield of infected ones only when the swards were frequently defoliated. Infrequent cutting apparently allowed infected plants to check the growth of healthy ones, and 50% infected swards yielded little more than 100% infected swards. This difference in response was attributed to differences in the growth habits of healthy and infected plants. Few initially healthy plants became infected during 2 years of the experiment.  相似文献

Electron microscope examination of myosin-extracted psoas muscleof the rabbit reveals a number of thin filaments, about 25 Äin diameter, bridging the gap between the ends of the actinsand the remaining myosin of the M band. Extraction of actinfrom the region of the Z disc shows similar thin filaments nearthe Z disc. Myosin-extraction of several invertebrate musclesreveals a remaining network of thin filaments in the regionof the A band. These thin filaments are probably elastic, andcould contribute to the functioning of these muscles.  相似文献

An Intermediate Category of Seed Storage Behaviour?: I. COFFEE   总被引:15,自引:3,他引:12  
Seeds of four cultivars of arabica coffee (Coffea arabica L.)were tested for germination following hermetic storage for upto 12 months at several different combinations of temperaturesbetween –20 °C and 15 °C and moisture contentsbetween 5% and 10% (wet basis). Most of the seeds from one cultivarwithstood desiccation to between 5% and 6% moisture content,a seed water potential of approximately –250 MPa, butthose of the remaining three cultivars were much more sensitiveto desiccation damage. Moreover, in all four cultivars, seedlongevity at cool and sub-zero temperatures, and at low moisturecontents did not conform with orthodox seed storage behaviour:viability was lost more rapidly under these conditions thanat either warmer temperatures or higher moisture contents. Theresults confirm that coffee seeds fail to satisfy the definitionsof either typical orthodox or recalcitrant seed storage behaviour.These results, therefore, point to the possibility of a thirdcategory of storage behaviour intermediate between those oforthodox and recalcitrant seeds. Various photon doses (net number of photons per unit area perday), provided by varying both photon flux density and photoperiod,were applied to imbibing seeds of seven lots of four speciesof Compositae in various germination test regimes. In all fourspecies germination was dependent upon photon dose, the productof photon flux density and daily duration of exposure. The responsewas quantified by linear relations between the probit of percentagegermination and the logarithm of photon dose. In general, photonflux density and photoperiod only influenced the stimulationof germination by the low energy reaction indirectly (as factorsof daily photon dose), whereas there was a tendency for photoperiodto have a direct influence on the inhibition of germinationby the high irradiance reaction. Reducing the germination testtemperature from 25?C to 20?C and 15?C not only increased thedark germination of L. sativa L., but also broadened the photondose range at which full germination occurred by reducing theminimum value necessary for the germination of the most dormantseeds, and increasing the maximum value which failed to inhibitthe germination of any seeds. Differences between L. sativaand L. serriola L. in the response of germination to white lightwere only quantitative, rather than qualitative. The singlemost promotory dose for all four species was 3 ? 10–3mol m–2 d–1, although the inhibitory action of dosesup to 10– mol m– d– was generally only slight. Mixed life cycle stages of rat-derived Pneumocystis carinii were isolated from host lungs and their sterols were compared with those present in lungs from normal and immunosuppressed uninfected rats. Gas-liquid chromatography consistently detected, resolved, and quantified 9, 10, and 20 sterol components in the total nonsaponifiable neutral lipid fraction of lungs from normal rats, lungs from immunosuppressed uninfected rats, and P. carinii preparations, respectively. In all samples, cholesterol was the most abundant sterol present, comprising 97%, 93%, and 78% of total sterols in lungs from normal rats, lungs from immunosuppressed uninfected rats, and P. carinii , respectively. Tentative identifications of several rat lung and P. carinii minor sterols were made based on gas-liquid chromatogram retention times and fragmentation patterns from mass spectral analyses. Campesterol (ergost-5-en-3-ol), cholest-5-en-3-one, and β -sitosterol (stigmast-5-en-3-ol) were among the minor components present in both types of lung controls, and were also components of P. carinii sterols. In contrast to lung controls, the sterols of P. carinii were enriched in C28 and C29 sterols with one or two double bonds, and a hydroxyl group at C-3 (ergost-5-en-3-ol, ergost-7-en-3-ol, ergosta-dien-3-ol, stigmast-5-en-3-ol, stigmast-7-en-3-ol and stigmasta-dien-3-ol). Steryl esters of P. carinii , probably stored in cytoplasmic lipid droplets, were dominated by those present in the host lung. In separate studies. 3-hydroxy-3-methylglutaryl coenzyme A activity, a key enzyme in the regulation of sterol biosynthesis, was detected in purified P. carinii preparations and incorporation of radiolabeled squalene and mevalonate was observed. Together, these results suggest that the parasite readily takes up and incorporates host sterols, and that the organism synthesizes some of its own "metabolic sterols"  相似文献
