The first collagen recognizable in the embryo is in the formof an incomplete basal lamina under the epiblast and hypoblast.We suggest that this collagen acts as a railroad track to guidethe migration of the primitive streak mesenchyme. The mesenchymeaggregates into chordamesoderm, a layer which is said to "induce"the overlying epiblast (now ectoderm) to develop into neuralfolds. This tissue interaction may be mediated by the formationof complete basal laminas separating the two tissues and bydeposition of sulfated mucopolysaccharides in the interveningextracellular space. At the very least, the collagenous basallamina serves to give the elongating cells of the developingneural tube a firm foothold. The fully formed neural tube andadjacent notochord are said to induce the sclerotome of thesomite to migrate medially and differentiate into cartilage.Notochord and neural tube basal lamina and collagen fibrilsmay play a role by guiding the migrating cells and stabilizingthe already existing chondrogenic bias of the cells. We wereunable to prove this hypothesis directly (by adding collagento somite cultures), because in our hands the somites died invitro even in the presence of neural tube and notochord. Wedid obtain direct evidence, however, that the basal lamina ofthe lens can promote the differentiation of the cornea in vitro.  相似文献

Electron microscopy of infected D. stramonium cells showed that the virus particles occurred in the cytoplasm. Particles were mostly bacilliform and measured 297 ± 18 times 64 ± 4 nm. In negatively stained leaf homogenates, particles were mostly disrupted; intact particles measured 267 ± 20 times 69 ± 6 nm. In brussels-sprout cells containing BNYV and CIMV, BNYV particles were rarely found compared with those of CIMV, and they occurred within and near CIMV inclusion bodies. BNYV particles were also found in extracts of virus-carrying Brevicoryne brassicae. Broccoli necrotic yellows (BNYV) and cauliflower mosaic (CIMV) viruses occurred together in naturally infected Brussels sprout plants, which showed conspicuous symptoms, and in cauliflower. BNYV was transmitted to and maintained in Datura stramonium and Hyoscyamus niger. It was partially purified from D. stramonium. Using these preparations, from which inhibitor had been removed, BNYV was manually transmitted to cauliflower, causing mild symptoms, and to Brussels sprout, causing a symptomless infection. BNYV also infected Sinapis alba but not cabbage, lettuce, Sonchus oleraceus or Plantago major. BNYV was transmitted by Brevicoryne brassicae but not by Myzus persicae, Hyperomyzus lactucae or Aleyrodes proletella.  相似文献

The types of opaline silica-bodies (opal phytoliths) which occurin the mature prophylls, radical and culm leaves, culms, andinflorescence bracts of rye (Secale cereale L.) are describedand figured. Silica-bodies are absent from the coleoptile, andthe adaxial epidermis of the prophylls, leaf sheaths, and inflorescencebracts. THE average content of DNA in Bacillus spores is unaffected by the growth medium and is constant for each species1. Within the Bacillus group, however, the average amount of DNA per spore varies from species to species1. A cell of B. cereus contains on average twice as much DNA as the spore and during sporulation this DNA is divided equally between the spore and the sporangium2. Estimates3–5 of genome size vary from 1.3 × 109 to 10 × 109 daltons and the number of genomes per cell from 2 to 44–8. Some of these variations may be associated with differences within the cells rather than differences of methodolqgy; spores within a population vary not only in size and shape, but also in their content of stainable chromatin9. Moreover, in ‘Renografm’ density gradients10, spores band within a range of densities. If spores are taken from a narrow part of this range, regrown and rebanded, the original pattern of dispersal occurs, suggesting that spores in the same population normally show variation in density as well as in size (A. IN 1969, after carbenicillin had been in use for three years in this unit, highly resistant strains of Pseudomonas aeruginosa were isolated for the first time1. Because these resistant strains included, from their first appearance, representatives of two unrelated types, it seemed likely that the resistance was transferable; this hypothesis was supported by experiments showing the transfer of carbenicillin resistance between Ps. aeruginosa and Escherichia coli K12 in vitro and in vivo2–4;. The resistant Ps. aeruginosa produced a penicillinase (β lactamase) similar to that normally produced by some strains of Enterobacteria and different from that normally produced by Ps. aeruginosa2,3, so it seemed likely that the Ps. aeruginosa had initially acquired resistance by the transfer of an R factor from a carbenicillin-resistant member of the Enterobacteriaceae colonizing the same burn. This hypothesis is now supported by a study on strains of Enterobacteria and Ps. aeruginosa isolated in a number of hospitals. We have also found evidence suggesting that Ps. aeruginosa which has acquired this R factor may not show resistance until it has been exposed repeatedly to carbenicillin.  相似文献

Inactivation of Calcitonin by Specific Organs   总被引:2,自引:0,他引:2  
THE greater biological potency of salmon calcitonin (SCT) as compared with mammalian calcitonins may be due to the relative resistance of SCT to inactivation in vivo1,2. SCT infused into dogs disappears from the circulation more slowly than does porcine calcitonin (PCT) or human calcitonin (HCT)1–3. For example, the metabolic clearance rate (MCR) of PCT in the dog is approximately 10 times greater than that of SCT1,2. Neither renal excretion3,4 nor inactivation by plasma1,2 is sufficient to account for the rapid clearance of the calcitonins that we have observed in vivo and thus it seemed likely that inactivation of the hormones must occur during passage through one or more organs. Here we present data that suggest the kidney, the liver and muscle and/or bone as the sites of inactivation of the calcitonins in the dog. SCT is relatively resistant to inactivation in the latter two sites.  相似文献

GENETIC VARIATION IN THE SHEEP RED BLOOD CELL   总被引:2,自引:0,他引:2  
1. There are 7 well-established red-cell antigen (blood group) loci. The R-O system has 3 phenotypes, R, O and i, identified by the ‘naturally occurring’ antibodies, anti-R and anti-O. The R and O substances are also present in soluble form in some body secretions. The expression of R and O is controlled by a dominant gene I, epistatic in effect, at an independent locus from that of R. The systems, A, C, M-L, B, D and X-2 are identified by means of ‘immune-type’ antibodies, and several of the loci have multiple alleles. An isoenzymic form of serum alkaline phosphatase is associated with the R-O system. The frequency for the genes at the various loci has been determined in a limited number of breeds. 2. Some sheep red cells have high K+ and low Na+ concentrations (HK type, or Key), others have low K+ and high Na+ concentrations (LK type or Kea). Two other rare forms exist; Key type which is HK but with lower than normal K+ values, and Kep type which has approximately equal Na+ and K+ concentrations. The red cells of foetuses and newborn lambs have high K+ levels irrespective of their potassium genotype. HK cells have 3–4 times greater (Na+-K+)-activated ATPase activity, a 3–4 times increased rate of active K+ transport and a larger number of ouabain-binding sites than LK cells. Antigen M is present on homozygous HK and heterozygous LK red cells, and antigen L is present on homozygous and heterozygous LK red cells. Sensitization of LK cells with anti-L stimulates active K+ transport and ATPase activity and exposes a larger number of ouabain-binding sites in these cells. Anti-M has no effect. The red cells of newborn lambs only show weak L and M antigen activity. It is postulated that L antigen inhibits cation transport in LK cells by masking the pump sites on the membrane. Immature red cells in LK-type sheep have a high rate of active K+ transport and yet have L antigen present. No satisfactory explanation for this has yet been advanced. There is no conclusive evidence that the potassium types have any significance from the point of view of adaptation or sheep breeding. The potassium-gene frequencies are known for a large number of breeds. 3. Two allelic genes, Hb4 and HbB control 3 haemoglobin phenotypes, A, AB, and B. Foetal haemoglobin (HbF) is present in foetuses and newborn lambs. Sheep with HbA also synthesize small amounts of another haemoglobin (HbC) and under conditions of severe anaemia, synthesis of HbC takes over from that of HbA. No change in HbB is observed in anaemia. A rare haemoglobin (HbD) has been found in 3 Yugoslavian sheep. Hbs A, B, C and F differ in their physicochemical properties; they share the same alpha chains but their non-alpha chains differ in a number of amino acids. HbD differs from HbA in one amino acid in the alpha chain. Certain genetic aspects are discussed. There is some evidence that sheep with HbA are less fertile than those with HbB. The gene frequencies for Hb are known for a large number of breeds. 4. Two isoenzymic forms of carbonic anhydrase are found in red-cell lysates and these are controlled by a pair of allelic autosomal genes, producing 3 phenotypes, CAF, CAFS and CAS. Only a few breeds have been studied but CAF is apparently quite rare. 5. An unidentified protein, designated ‘X’ is present in electrophoretic separations of haemolysates from some sheep. Its presence is dominant to its absence. Polymorphism at this locus is present in all breeds so far studied. 6. A deficiency of reduced glutathione (GSH) in red cells is found in some sheep and is inherited as an autosomal recessive disorder. Sheep with this deficiency have lower red-cell K+ and Naf concentrations than normal and it is suggested that the HK GSH-deficient sheep may be Ked type sheep. This deficiency has so far only been found with certainty in one breed of sheep. 7. In sheep twin chimeras, admixture of red-cell antigens, haemoglobin and ‘X’ protein types has been found. Various aspects of chimerism, which occurs only rarely in sheep, are discussed. 8. The significance of the genetic variation is discussed in the light of the physiology and immunology of the red cell and of the sheep itself.  相似文献

SUMMARY: Many of 110 strains of Serratia , isolated from soil, water, milk and dairy equipment, were biochemically closely related to the coli-aerogenes bacteria. Acid and gas was formed from glucose in 14 days at 30° by 53% and from lactose and MacConkey's broth by about 40%. All except one strain gave——++ IMViC reactions.
An inverse relationship was observed between depth of pigmentation and carbohydrate fermentation. Complete loss of pigment in mutant strains was not uncommon, and was associated with loss of proteolytic properties and increase of saccharolytic activity.
The majority of the strains had psychrophilic characteristics: 75% grew at 3–5°. Most strains showed moderate growth at 37°, but only 7 formed red pigment at that temperature.
All strains resembled Serratia marcescens in morphology, containing minute coccoid rods smaller than those of coli-aerogenes bacteria.  相似文献

SUMMARY. The structure and life history of a new species of eugregarine, Gregarina garnhami n.sp., is described from the intestinal caeca and mid-gut of Schistocerca gregaria Forsk. The parasite destroys considerable areas of the caecal epithelium and in cases of heavy infection, the masses of parasites present in the mid-gut result in the formation of localized barriers between the gut wall and the food material in the lumen. The cephalont and sporont stages of gregarines from Locusta migratoria migratorioides R. & F. and Anacrydium aegyptium Linn. are shown to be similar to those from Schistocerca gregaria and are believed to belong to the same species.  相似文献
