The tumor promoter 12–0-tetradecanoylphorbol-13-acetate (TPA) dramatically modifies the differentiative program of myotubes, developed in culture from chick embryo skeletal myogenic cells. In fact TPA selectively decreases the expression of differentiative parameters with a lag of 8–10 h from its administration to the cultures. We have tested whether the reported effect of TPA depends on the synthesis of specific products during the lag phase of TPA action. The data presented indicate that inhibition of protein synthesis by the use of cycloheximide prevents the appearance of TPA induced inhibition of the expression of differentiative products, such as creatine phosphokinase (CPK) activity and acetylcholine receptors (AChR). Following removal of cycloheximide and reinitiation of normal protein synthesis, the TPA induced inhibitory effect on CPK and AChR appears after a delay of about the same length as the time lag of TPA action. Our results indicate that inhibition of protein synthesis during the lag phase of TPA action prevents the effect of this tumor promoter on myotube differentiative parameters, and suggest that the expression of differentiative traits in cultured myotubes is affected by TPA via a regulatory step implying protein synthesis.  相似文献

Despite formal separation based on molecular and morphological evidence, the genera Prosthechea Knowles & Westc. and Encyclia Hook. have not been studied in terms of their vegetative anatomy. In this study we examine 16 Brazilian species of these genera. Additionally, one species of subtribe Laeliinae and another from subtribe Oncidiinae were studied in order to evaluate the taxonomic consistency of the anatomic characters observed. Except for Epidendrum crassifolium Lindl., all species possess a velamen differentiated into epivelamen and endovelamen. Endodermal cells with uniformly thickened walls, calcium oxalate raphides and cells with phi thickenings in the cortical region were observed in all species. Silica bodies, raphides and fibre bundles in the mesophyll were common in leaves of all species studied. By contrast, flavonoid crystals were observed in both roots and leaves of Prosthechea only. Encyclia is characterized by the presence of fibre bundles on the subepidermal layer, and a cuticle, which is clearly thicker than that of Prosthechea . Lowland tropical habitats harbour an unexplored genetic diversity of epiphyllous fungi. In the shade of rainforest understoreys, lichenized fungi are specialized to an ephemeral habitat where they produce little vegetative biomass and develop reproductive structures early. In a first population genetic study of epiphyllous lichen fungi, we analysed the intraspecific genetic diversity of five leaf-colonizing lichen mycobiont species. Sampling focused on a lowland perhumid forest plot in Costa Rica, with additional collections from other localities throughout the country. In all species we detected sympatric occurrence of highly diverged haplotypes. Haplotypes belonging to distinct clades in networks were also found on the same leaf, clearly indicating multiple independent colonization events on single leaves. Despite the unusually high genetic diversity of these leaf-colonizing tropical fungi, we did not detect pronounced spatial structure of the haplotype distribution between geographical regions. The observed patterns suggest that the diversity of foliicolous lichens could be much higher than expected, with several cryptic genetic lineages within each morphologically characterized species.  相似文献

Fires set for slash‐and‐burn agriculture contribute to the current unsustainable accumulation of atmospheric greenhouse gases, and they also deplete the soil of essential nutrients, which compromises agricultural sustainability at local scales. Integrated assessments of greenhouse gas emissions have compared intensive cropping systems in industrialized countries, but such assessments have not been applied to common cropping systems of smallholder farmers in developing countries. We report an integrated assessment of greenhouse gas emissions in slash‐and‐burn agriculture and an alternative chop‐and‐mulch system in the Amazon Basin. The soil consumed atmospheric methane (CH4) under slash‐and‐burn treatment and became a net emitter of CH4 to the atmosphere under the mulch treatment. Mulching also caused about a 50% increase in soil emissions of nitric oxide and nitrous oxide and required greater use of fertilizer and fuel for farm machinery. Despite these significantly higher emissions of greenhouse gases during the cropping phase under the alternative chop‐and‐mulch system, calculated pyrogenic emissions in the slash‐and‐burn system were much larger, especially for CH4. The global warming potential CO2‐equivalent emissions calculated for the entire crop cycles were at least five times lower in chop‐and‐mulch compared with slash‐and‐burn. The crop yields were similar for the two systems. While economic and logistical considerations remain to be worked out for alternatives to slash‐and‐burn, these results demonstrate a potential ‘win‐win’ strategy for maintaining soil fertility and reducing net greenhouse gas emissions, thus simultaneously contributing to sustainability at both spatial scales.  相似文献

This paper presents the mitotic chromosome numbers of 18 species of Bromeliaceae. The diploid number 2n = 50 was observed in Aechmea comata, A. caudata, A. correia‐araujoi, A. recurvata, A. marauensis, A. bicolor, A. pineliana, Hohenbergia catingae, H. blanchetti, Alcantarea imperialis, Al. nahoumi, Neoregelia tenebrosa, Nidularium lyman‐smithii, N. scheremetiewii, N. innocentii var. innocentii, and N. innocentii × Neoregelia johannis hybrid, whereas 2n = 34 was observed in Cryptanthus maritimus and C. warren‐loosei. All of the determinations presented in this study are previously unpublished, except A. comata and H. catingae. Stone-curlews Burhinus oedicnemus have a vulnerable population status in the UK after a large population decline and range contraction since the 1930s. Much Stone-curlew breeding habitat is open-access land designated under the Countryside and Rights of Way Act 2000. In order to guide the conservation and habitat management for this species whilst allowing recreational access, a tool known as the Stone-Curlew Access Response Evaluator (SCARE) has been developed. SCARE offers a method to assess the effects of scenarios for future changes in disturbance type, routes and frequency and will be valuable in making informed decisions about the management of public access to Stone-curlew breeding sites and the deployment of habitat creation measures.  相似文献

Mitotic chromosomes of eight populations of six species of Vernonia sensu Baker (Asteraceae, Vernonieae, section Lepidaploa , group Axilliflorae ) were studied to assess the validity of maintaining the genus as proposed currently ( sensu Baker) or of splitting it into several smaller genera ( sensu Robinson). The species were collected in areas of 'cerrado' and 'campo rupestre' in southern and central-western Brazil. The chromosome numbers were 2 n  = 20–40 and karyotypic analysis showed a predominance of metacentric chromosomes, with some submetacentrics. B chromosomes were seen in one population of V. geminata . The chromosomes varied in length from 0.9 to 4.6 µm, with a total chromatin length of 29.7–45.0 µm and a symmetry index of 41.2–46.9%. The intrachromosomal asymmetry index varied from 0.13 to 0.29, and the interchromosomal asymmetry index varied from 0.15 to 0.21. The dispersion diagram showed that population 1 of V. geminata had the most asymmetrical karyotype. These results do not provide sufficient evidence for a conclusive cytotaxonomic discussion of Vernonia , mainly because of the impossibility of associating any infrageneric group with specific karyotypic characters, and also because of the small number of species analysed so far.  © 2007 The Linnean Society of London.
