Isoprenylated proteins bear an isoprenylcysteine methyl ester at the C terminus. Although isoprenylated proteins have been implicated in meristem development and negative regulation of abscisic acid (ABA) signaling, the functional role of the terminal methyl group has not been described. Here, we show that transgenic Arabidopsis thaliana plants overproducing isoprenylcysteine methyltransferase (ICMT) exhibit ABA insensitivity in stomatal closure and seed germination assays, establishing ICMT as a negative regulator of ABA signaling. By contrast, transgenic plants overproducing isoprenylcysteine methylesterase (ICME) exhibit ABA hypersensitivity in stomatal closure and seed germination assays. Thus, ICME is a positive regulator of ABA signaling. To test the hypothesis that ABA signaling is under feedback regulation at the level of isoprenylcysteine methylation, we examined the effect of ABA on ICMT and ICME gene expression. Interestingly, ABA induces ICME gene expression, establishing a positive feedback loop whereby ABA promotes ABA responsiveness of plant cells via induction of ICME expression, which presumably results in the demethylation and inactivation of isoprenylated negative regulators of ABA signaling. These results suggest strategies for metabolic engineering of crop species for drought tolerance by targeted alterations in isoprenylcysteine methylation.  相似文献

The survival of coxsackievirus B3 was studied under various conditions of incubation. The comparative study demonstrated that coxsackievirus B3 was stable for 24h (less than 0.4-log decrease in titer) when suspended at neutral pH (6 or 23 degrees C) in the presence of 0.25% bovine serum albumin in saline regardless of whether the preparations were subjected to evaporation. Bovine serum albumin provided increased stability to the virus for each of the conditions tested. At 37 degrees C, evaporation greatly reduced the virus infectivity between 6 and 20 h of incubation. Nevertheless, coxsackievirus B3 was found to be stable for at least 24 h under conditions similar to those of a household environment, and its presence represents a potential biohazard to nonimmune persons. These data provide a rationale for using coxsackievirus B3 as a model for investigating the role of environmental surfaces in the transmission of enteroviral diseases.  相似文献

Evidence for reduction of clutch size in insular passerines is presented. Average clutch sizes for the Catbird, Cardinal and White-eyed Vireo were reduced 14.0, 18.2 and 30.7%, respectively as compared with coastal North American populations from similar latitudes. Differences are highly significant. Clutch size in the Eastern Bluebird was not reduced. Trends in latitudinal and longitudinal variations of clutch size in North America are examined. With the exception of a slight latitudinal gradient in coastal populations, no consistent trends existed. There is no evidence for reduced variability in clutch size or increased egg size in the Bermudan populations, but both these parameters may show some response to combined effects of latitude and longitude in continental populations.
Reduction of clutch size in insular nidicolous birds is consistent with both the Lack-Ashmole resource limitation hypothesis and Cody's energy allocation hypothesis. In the absence of experimental evidence for life history optimization in response to K-selection, the modified Lack hypothesis seems sufficient.  相似文献

Solid-state deuterium ((2)H) NMR spectroscopy was used to study the reorientation of magnetically ordered bicelles in the presence of the paramagnetic lanthanide Eu(3+). Bicelles were composed of 1,2-dimyristoyl-sn-glycero-3-phosphocholine (DMPC) plus 1,2-dihexanoyl-sn-glycero-3-phosphocholine plus either the anionic lipid 1,2-dimyristoyl-sn-3-phosphoglycerol, or the cationic lipid 1,2-dimyristoyl-3-trimethyl ammonium propane. Alignment of the bicelles in the magnetic field produced (2)H NMR spectra consisting of a pair of quadrupole doublets, one from the alpha-deuterons and one from the beta-deuterons of DMPC-alpha,beta-d(4). Eu(3+) addition induced the appearance of a second set of quadrupole doublets, having approximately twice the quadrupolar splittings of the originals, and growing progressively in intensity with increasing Eu(3+), at the expense of the intensity of the originals. The new resonances were attributed to bicelles having a parallel alignment with respect to the magnetic field, as opposed to the perpendicular alignment preferred in the absence of Eu(3+). Therefore, the equilibrium degree and kinetics of reorientation could be evaluated from the (2)H NMR spectra. For more cationic initial surface charges, higher amounts of added Eu(3+) were required to induce a given degree of reorientation. However, the equilibrium degree of bicellar reorientation was found to depend solely on the amount of bound Eu(3+), regardless of the bicelle composition. The kinetics of reorientation were a function of lipid concentration. At high lipid concentration, a single fast rate of reorientation (minutes) described the approach to the equilibrium degree of orientation. At lower lipid concentrations, two rates processes were discernible: one fast (minutes) and one slow (hours). The data indicate, therefore, that bicelle reorientation is a phase transition made critical by bicelle-bicelle interactions.  相似文献

MAPK phosphatases (MKPs) are critical modulators of the innate immune response, and yet the mechanisms regulating their accumulation remain poorly understood. In the present studies, we investigated the role of post-translational modification in the accumulation of MKP-1 and MKP-2 in macrophages following LPS stimulation. We found that upon LPS stimulation, MKP-1 and MKP-2 accumulated with different kinetics: MKP-1 level peaked at ∼1 h, while MKP-2 levels continued to rise for at least 6 h. Accumulation of both MKP-1 and MKP-2 were attenuated by inhibition of the ERK cascade. Interestingly, p38 inhibition prior to LPS stimulation had little effect on MKP-1 and MKP-2 protein levels, but hindered their detection by an M-18 MKP-1 antibody. Studies of the epitope sequence recognized by the M-18 MKP-1 antibody revealed extensive phosphorylation of two serine residues in the C terminus of both MKP-1 and MKP-2 by the ERK pathway. Remarkably, the stability of both MKP-1 and MKP-2 was markedly decreased in macrophages in the presence of an ERK pathway inhibitor. Mutation of the two C-terminal serine residues in MKP-1 and MKP-2 to alanine decreased their half-lives, while mutating these residues to aspartate dramatically increased their half-lives. Deletion of the C terminus from MKP-1 and MKP-2 also considerably increased their stabilities. Surprisingly, enhanced stabilities of the MKP-1 and MKP-2 mutants were not associated with decreased ubiquitination. Degradation of both MKP-1 and MKP-2 was attenuated by proteasomal inhibitors. Our studies suggest that MKP-1 and MKP-2 stability is regulated by ERK-mediated phosphorylation through a degradation pathway independent of polyubiquitination.  相似文献

For the mammalian faunas of 24 landbridge islands in the Gulf of Maine (0.003–279 km2 in size), area accounts for 86% of variance in species richness. The slope, z , of the species-area curve is 0.247. For the seven largest islands (>10km2), the non-equilibrium hypothesis of relaxation following saturation in the post-Pleistocene is suggested by (1) elevated slope of the species-area curve (0.353), (2) correlation of species richness with island age ( r =- 0.81) and water depth to mainland ( r = -0.70), (3) highly non-random nested subsets of species ranked by island area, and (4) discontinuity with the extremely depauperate faunas of oceanic islands of the eastern North Atlantic. The alternative hypothesis of a dynamic equilibrium determined by recurrent immigration and extinction is supported by (1) documented turnover in 16 species, (2) correlation of species-area residuals with distance ( r = - 0.90), (3) distribution dependent upon vagility with reduction or absence of hibernators and other poor dispersers, (4) low levels of endemism, and (5) congruence of community structure with that of mainland fauna for both trophic level and body size.
I conclude that while some insular populations may be relictual, the faunal composition of most of these islands is dependent on recurrent colonization, much of which takes place over ice bridges. However, true equilibrium is perturbed by climatic shifts, range expansions, and human disturbance.  相似文献

Bacillus popilliae NRRL B-2309S forms several million refractile spores/ml in liquid shaken cultures. A suitable medium includes glucose, K2HPO4, water, and three selected ingredients-activated carbon, a yeast autolyzate, and a casein hydrolyzate. Only 4 out of 26 lots of commercial yeast autolyzates tried were sporogenic. However, spore formation in the presence of the four was poor and erratic unless a compatible casein hydrolyzate also was present. Five of eight lots of casein hydrolyzates improved the sporogenicity of selected yeast products to various degrees. A comparison of amino acid compositions of yeast and casein lysates sheds no light on differences between suitable and unsuitable ones. Less refined yeast and casein products seem preferable.  相似文献

Heritable variation in metabolic traits is likely to affect fitness. In this study, white-footed mice from wild-derived photoresponsive [R, infertile in short day length (SD)] and non-photoresponsive (NR, fertile in SD) selection lines were maintained under short-day (SD 8Light:16Dark), sub-thermoneutral conditions (22 or 12 °C). Mice had significantly higher levels of food intake and resting metabolic rates (RMR) at low temperature. RMR differed significantly between lines (greater in NR mice). In contrast to previous work under thermoneutral conditions, there was no significant difference in overall activity or average daily metabolic rates (ADMR) of mice from the two lines. Reduced activity may reflect behavioral changes under cooler conditions (e.g., nest building) reducing the overall energetic cost of fertility (for NR mice). There was no significant difference in maximal rate of oxygen consumption ( $\dot V \text{O}_{\text {2max}} $ ) between lines. R mice had significantly greater brown adipose tissue and white abdominal fat mass due to both line and temperature. Reaction norms for intake, resting metabolism (RMR/BMR) and level of activity from current (12 and 22 °C) and previously published data (28 °C) demonstrate independent effects of genetics (line) and environment (temperature) for resting metabolism, but a clear interaction between these for activity. The results suggest that fertility under winter conditions imposes metabolic costs that are related to the level of reproductive development. Under the coldest conditions tested, however, mice that remained fertile in SD reduced activity, ADMR and food requirements, decreasing the differential between selection lines. Heritable variation in reaction norms suggests a genetic by environment effect that could be subject to selection.  相似文献

Salivary gland duct ligation is an alternative to gland excision for treating sialorrhea or reducing salivary gland size prior to tumor excision. Duct ligation also is used as an approach to study salivary gland aging, regeneration, radiotherapy, sialolithiasis and sialadenitis. Reports conflict about the contribution of each salivary cell population to gland size reduction after ductal ligation. Certain cell populations, especially acini, reportedly undergo atrophy, apoptosis and proliferation during reduction of gland size. Acini also have been reported to de-differentiate into ducts. These contradictory results have been attributed to different animal or salivary gland models, or to methods of ligation. We report here a bilateral double ligature technique for rabbit parotid glands with histologic observations at 1, 7, 14, 30, 60 days after ligation. A large battery of special stains and immunohistochemical procedures was employed to define the cell populations. Four stages with overlapping features were observed that led to progressive shutdown of gland activities: 1) marked atrophy of the acinar cells occurred by 14 days, 2) response to and removal of the secretory material trapped in the acinar and ductal lumens mainly between 30 and 60 days, 3) reduction in the number of parenchymal (mostly acinar) cells by apoptosis that occurred mainly between 14–30 days, and 4) maintenance of steady-state at 60 days with a low rate of fluid, protein, and glycoprotein secretion, which greatly decreased the number of leukocytes engaged in the removal of the luminal contents. The main post- ligation characteristics were dilation of ductal and acinar lumens, massive transient infiltration of mostly heterophils (rabbit polymorphonuclear leukocytes), acinar atrophy, and apoptosis of both acinar and ductal cells. Proliferation was uncommon except in the larger ducts. By 30 days, the distribution of myoepithelial cells had spread from exclusively investing the intercalated ducts pre-ligation to surrounding a majority of the residual duct-like structures, many of which clearly were atrophic acini. Thus, both atrophy and apoptosis made major contributions to the post-ligation reduction in gland size. Structures also occurred with both ductal and acinar markers that suggested acini differentiating into ducts. Overall, the reaction to duct ligation proceeded at a considerably slower pace in the rabbit parotid glands than has been reported for the salivary glands of the rat.  相似文献
